変形性関節症(OA)は、関節軟骨1の細胞外マトリックスの進行性および不可逆的な炎症および破壊を意味する関節変性疾患である。関節炎の様々な形態は、多数の治療法を持っていますが2,3,4,これらは、その副作用と限られた有効性によって制限されています.自家軟骨細胞移植を用いた組織工学技術は、OA初期の軟骨病変における負傷軟骨の再生治療のために日常的に適用される。細胞ベースの治療法は、主に、軟骨3を効果的に修復することができる限られた数の平種安定な軟骨細胞またはチョンドロゲニターのために制限される。したがって、疾患の進行を予防し、大きな軟骨病変を再生するための新しい治療戦略の開発が最も重要である。
細胞外小胞(EV)は、異なる著者5,6によってOAの治療法として提案されている。EVは、細胞型の大部分によって分泌される膜状体であり、細胞間シグナル伝達に関与しており、そして、それらは最近再生医療への関心を引き出した幹細胞の治療効果7、8、9を発揮することが示されている。間葉間質細胞(MSC)に由来するEVは、OAに関して調査された主な治療用EVであり、他の関節関連細胞はEV源として使用されているが、例えば、チョンドロゲニタまたは免疫細胞11、12。
血小板リサート(PLs)などの血小板濃縮物は、角膜潰瘍13、14、15、腱組織再生16などの異なる傷害における創傷治癒を増強するために使用され、血小板濃縮物のEV成分がこれらの効果に関与する可能性があるという仮説のため17 .関節関連疾患に関連するいくつかの研究は、骨関節炎の状態を改善するための治療法として血小板由来のEV(PL-EV)を使用しています。PL-EVは、Wnt/β-カテニン経路18を活性化することによって軟骨細胞増殖および細胞遊行を改善し、変形性関節症軟骨球19における軟骨形成マーカーの発現を促進し、または、軟骨原性タンパク質のより高いレベルを示し、PL-EV18で治療した骨関節症ウサギの骨関節異常を少なく示す。

このシンプルなプロトコルは、1つのラベルを持って、共焦点顕微鏡を介してそれらを監視することができます。例えば、EVイメージングは、治療薬で使用する場合のEV分布の評価に役立つ可能性があります。この技術により、EV移動と軟骨外植の取り込みが、特別な機能を持たない共焦点顕微鏡で容易にモニタリングできます。同じプロトコルを、in vitroまたはin vivo実験用のEVに使用できます。列が事前に用意されていること、および EV の初期量が、精製工程で粒子の約 10% が失われることを考慮して、目的の最終粒子濃度に達するのに十分であることを確認してください。細胞外小胞、またはEV、サンプルおよびコントロールを濃縮することから始めます。EVサンプルの開始容積に応じて、サンプルを15ミリリットルまたは500マイクロリットルの濃縮チューブに入れます。ほとんど乾燥したサンプルが得られるまで、製造業者の指示に従ってチューブを遠心分離します。濃縮されたEVサンプルを200マイクロリットル、対照群に100マイクロリットルの希釈Cを再中断し、新しい1.5ミリリットル遠心管に移します。EVサンプルをそれぞれ100マイクロリットルの2つのアリコートに分ける。染料でアリコートの1つをマークし、治療としてそれを使用します。もう一方は無印のままにして、EVサンプルのように処理し、ナノ粒子追跡解析によりEV濃度を定量化する。2x色素溶液を調製し、希釈C中のPKH26溶液の濃度が8マイクロモルになるようにした。1ミリモルPKH26リンカーの1マイクロリットルを、サンプル中の希釈剤Cの125マイクロリットルにつき混合します。サンプルに追加するために必要なボリュームを 1:1 の比率で準備します。PKH血小板溶解物由来のEVと対照試料に2x色素溶液を1:1比で添加し、1x色素濃度、および4マイクロモルPKH26濃度を達成します。同じ量のPBSをNTA血小板溶地由来EVサンプルに加えます。室温で5分間インキュベートします。5%ウシ血清アルブミン-PBS溶液を1:1の比率でサンプルに加え、最終体積が約400マイクロリットルであることを確認します。ラベル付きの EV と、非バウンド色素と非特異的な色素の相互作用を列と分離します。カラムからキャップを取り出し、PKH血小板溶菌由来のEV、NTA血小板溶菌由来のEVを400マイクロリットル加えるか、溶出した液体をすべて制御して廃棄します。次の手順に進む前に、サンプルが列に完全に入るのを待ちます。600マイクロリットルのPBSを加え、溶出した液体をすべて捨てます。次の手順に進む前に、PBS が列に完全に入るのを待ちます。PBSの600マイクロリットルを追加し、1.5ミリリットル遠心管に600マイクロリットルの分画を収集します。新しい列の場合は、プロトコルの最初に言及されているサンプルの濃度に関する手順を繰り返します。600マイクロリットルの以前に溶出したEVをカラムに加え、溶出したボリュームを捨てます。その後、PBSの400マイクロリットルを追加し、すべての溶出ボリュームを捨てます。600マイクロリットルのPBSをカラムに加え、1.5ミリリットルの遠心管に600マイクロリットルの分画を集めます。PBSで軟骨を2回洗浄し、無菌条件下で直径3ミリの生検パンチを使用して切除します。エクスプラントをDMEM-F12培地付きの96ウェル培養プレートに入れ、摂氏37度、5%CO2、湿度80%で1%ペニシリンレンストレプトマイシンを補います。炎症駆動型OAモデルを確立するには、オンコスタチンMの1ミリリットル当たり10ナノグラムと腫瘍壊死因子アルファの1ミリリットル当たり2ナノグラムで細胞培養培地を補う。PKH血小板ライセート由来のEVのウェル当たり10〜9番目の粒子で外植を治療し、またはコントロールして、オンコスタチンMおよび腫瘍壊死因子αを添加した細胞培養培地で治療する。軟骨外植を含む96ウェル細胞培養プレートから培地を取り除きます。200マイクロリットルの細胞培養培地を、原稿に記載した各々に加える。1時間ごとにインビトロアッセイをゼロから5時間に停止します。200マイクロリットルのPBSで軟骨外植を含む細胞培養井戸を2回洗浄する。4%パラホルムアルデヒドの100マイクロリットルを組織に加え、摂氏4度で3時間固定します。PFAを取り出し、PBSを100マイクロリットル加え、固定組織を摂氏4度で保存し、48時間以内にサンプルを処理します。異なる時点で撮影された画像は、EVが軟骨細胞に到達するまで組織に入り、時間の経過とともに軟骨細胞に入る様子を示しています。ラベル付きのEVは、1時間のインキュベーションの後、既に軟骨細胞の周りに局在していた。EV の初期ボリュームが、目的の最終パーティクル濃度に達するのに十分であることを確認します。覚えておかなもう1つの重要なことは、次の24時間以内にラベル付きのEVを使用することです。24時間以内に使用しないと、蛍光の低下が起こることがある。この技術は、研究者がEVイメージングを改善し、生物学や治療のEVを研究するのに役立ちます。
