Dit protocol is bedoeld om duidelijke instructies te geven voor de functionalisatie van EV's op Ti-oppervlakken. De gepresenteerde methode is gebaseerd op een drop casting-strategie, een physisorptietype van functionalisatie. Er bestaat een slechte bibliografie met betrekking tot de functionalisering van EV's op Ti-oppervlakken, hoewel er weinig studies zijn die verschillende voordelen aantonen door EV's op Ti10 te immobiliseren. EV's zijn membraanblaasjes (30-200 nm) die door elke cel worden uitgescheiden en spelen een sleutelrol in cel-naar-cel communicatie door hun lading af te leveren. Ze bevatten een verscheidenheid aan actieve biomoleculen die nucleïnezuren, groeifactoren of bioactieve lipiden kunnen bevatten1. Om deze redenen zijn EV's geëvalueerd op hun potentiële gebruik in therapeutica. Op het gebied van orthopedie en botregeneratie zijn EV's uit verschillende bronnen getest. Onder hen is aangetoond dat van bloedplaatjes afgeleide EV's een differentiatie-effect op stamcellen induceren met behoud van een laag cytotoxisch profiel2,3. Daarom is verder onderzoek nodig om de mogelijkheid te onderzoeken om EV's te combineren met biomaterialen om ze in de dagelijkse klinische praktijk te gebruiken.
Op titanium gebaseerde biomaterialen worden veel gebruikt als steigers voor botgenezende klinische interventies vanwege hun mechanische eigenschappen, hoge biocompatibiliteit en duurzaamheid op lange termijn4. Niettemin zijn Ti-implantaten een bio-inert materiaal en vertonen daarom een slecht vermogen om zich te hechten aan het omliggende botweefsel5. Om deze reden worden titaniummodificaties bestudeerd om hun prestaties te verbeteren door een meer functionele micro-omgeving op het oppervlak te bereiken4,6,7. In die zin kunnen EV's aan titanium worden verankerd door chemische8 of fysische interacties9,10. Geïmmobiliseerde EV's afgeleid van stamcellen of macrofagen verbeteren de bioactiviteit van Ti door cellulaire adhesie en proliferatie te bevorderen en zo een osteogeen effect te induceren8,9,10.
Dit artikel zal zich in detail richten op een drop casting-strategie voor het coaten van Ti-oppervlakken met PL-afgeleide EV's. Daarnaast zullen we het afgifteprofiel van EV's van het gecoate oppervlak in de loop van de tijd evalueren en de cellulaire biocompatibiliteit in vitro bevestigen.

platelet-derived extracellular vesicle functionalization of ti implants

Extracellulaire blaasjes (EV's) zijn biologische nanovesicles die een sleutelrol spelen in de celcommunicatie. Hun inhoud omvat actieve biomoleculen zoals eiwitten en nucleïnezuren, die een groot potentieel bieden in de regeneratieve geneeskunde. Meer recent hebben EV's afgeleid van Platelet Lysate (PL) een osteogene capaciteit getoond die vergelijkbaar is met PL. Bovendien worden biomaterialen vaak gebruikt in orthopedie of tandheelkundige restauratie. Hier bieden we een methode om Ti-oppervlakken te functionaliseren met PL-afgeleide EV's om hun osteogene eigenschappen te verbeteren.
EV's worden geïsoleerd van PL door grootte-uitsluitingschromatografie en daarna worden Ti-oppervlakken gefunctionaliseerd met PL-EV's door drop casting. Functionalisatie wordt bewezen door ev-afgifte en de biocompatibiliteit ervan door de lactaatdehydrogenase (LDH) release assay.

De methode die in dit artikel wordt gepresenteerd, maakt het mogelijk om EV's gefunctionaliseerde titaniumschijven te verkrijgen. EV's zijn fysiek gebonden aan het oppervlak, wat een aanhoudende afgifte in de loop van de tijd mogelijk maakt. Het aantal vrijgegeven EV's kan worden gemeten door NTA op dag 2, 6, 10 en 14. Hier presenteren we een methode voor de isolatie van extracellulaire blaasjes (EV's) afgeleid van de bloedplaatjeslysaten (PL) en hun gebruik voor het coaten van titanium (Ti) implantaatoppervlakken. Extracellulaire blaasjes combinatie met biomaterialen is een wenselijke aanpak voor regeneratieve geneeskunde studies. Deze studie rapporteert dus titanium functionalisatie met EV's als een praktisch hulpmiddel voor orthopedie en botregeneratie. De druppelgiet functionalisatie is een eenvoudige en goedkope methode in vergelijking met andere strategieën zoals polymere beknelling of biochemische binding.Begin met de functionalisering van het titaniumoppervlak door de titaniumschijven te wassen met gedeïoniseerd water in een glazen bekerglas. Gooi vervolgens het water weg voordat u de schijven wast met 70% ethanol. Decanteer de oplossing om de gewassen implantaten gedurende vijf minuten in gedeïoniseerd water bij 50 C te soniceren.Na het weggooien van het water incubeert u titaniumimplantaten in een 40% natriumhydroxideoplossing bij 50 C gedurende 10 minuten met agitatie en vervolgens soniceren de implantaten in gedeïoniseerd water zoals aangetoond. Voer vervolgens ten minste vijf wasbeurten van de implantaten uit met gedeïoniseerd water. Totdat de pH neutraal is op de pH-indicator.Herhaal het ultrasoonapparaat en incubatieproces, zoals eerder uitgelegd, door natriumhydroxide te vervangen door 50% salpeterzuur. Voer de wasbeurten en ultrasoonapparaat uit met gedeïoniseerd water voordat u de implantaten opslaat in een 70% ethanoloplossing. Incubeer de titanium implantaten in een 30% salpeterzuuroplossing gedurende 30 minuten bij kamertemperatuur met zachte agitatie.Voer na incubatie ten minste vijf wasbeurten uit met gedeïoniseerd water totdat de pH neutraal is op pH-indicatoren. Incubeer vervolgens titanium implantaten 's nachts bij kamertemperatuur in gedeïoniseerd water. Droog de volgende dag de implantaten af onder vacuüm bij 40 C gedurende 10 minuten.Werk onder de celkweekkast en plaats titanium implantaten in een 96-well plaat met de machinezijde naar boven gericht. Zorg ervoor dat de extracellulaire blaasjes EV's vallen door ev-oplossing te ontdooien en vortexoplossing met een puls van drie seconden. Zet na het mengen 40 microliter van de EV-oplossing af op het titaniumoppervlak om maximaal 4 x 10 ^ 11 EV's per implantaat te immobiliseren, volgens de concentratie bepaald door nanodeeltjesvolganalyse.Plaats de platen vervolgens onder vacuümomstandigheden op 37 C gedurende ongeveer twee uur of totdat de druppels volledig droog zijn. Pas de droogtijd aan afhankelijk van het aantal titanium implantaten en het water dat in de vacuümkamer aanwezig is. De hoeveelheid EV's die vrijkwamen uit titaniumschijven werd gemeten door nanodeeltjes tracking analyse.Op dag 2 werden ongeveer 10^9 EV's uitgebracht, gevolgd door een aanhoudende release op dag 6, 10 en 14. In vitro celbiocompatibiliteit van de titanium EV's werd beoordeeld met lactaatdehydrogenase, of LDH-afgiftetest, 48 uur na celzaaien op de implantaten. Titanium EV's vertoonden een lagere hoeveelheid LDH dan de maximaal geaccepteerde cytotoxische waarde.Terwijl de titaniumcontrolegroep hogere LDH-activiteitsniveaus vertoonde. De belangrijkste stap is het vacuümdrogen van het gecoate implantaat. Het is belangrijk om ervoor te zorgen dat al het water wordt verdampt om een optimale lichaamsbouw te krijgen.
