このプロトコルは、火傷の様々な側面と結果とその治療に関する研究に適しており、この研究分野に重要な情報をもたらすことが期待されています。このプロトコルは、温水誘発性火傷動物モデルを確立し、関連する疼痛、歩行障害、およびうつ病様行動応答を測定する方法を提案する。手順を実演するのは、キム博士の研究室の修士課程の学生であるキム・ジェヒョクです。まず、体重20〜25グラムの雄ICRマウスを光と温度制御された部屋に収容します。マウスが食物と水に自由にアクセスできるようにし、実験開始前に少なくとも1週間は順応させる。1週間後、マウスを実験群または対照群にランダムに割り当て、動物番号を盲検実験を行うためのコードとして使用する。火傷誘発を行う前に、手術用ガウン、手袋、マスクを着用してください。次に、マウスを深く麻酔した後、右後足の周りを70%エタノールで消毒し、眼軟膏を眼に塗布して角膜乾燥を防ぎ、各マウスの足首に印を付け、右後足を摂氏65度のお湯に3秒間浸します。火傷後、麻酔から回復するまで、マウスを清潔な家庭用ケージ内の加熱パッドの上に置きます。マウスを行動試験室に持ってきて、試験前に少なくとも30分間順応させてください。マウスが順応している間、手術用ガウン、手袋、マスクを着用します。次に、マウスを金属メッシュの床の上の正方形の箱に入れ、さらに30分間順応させます。次に上昇刺激法を用いて後足の機械的閾値を評価するために、一連のフォン・フレイフィラメントで足を5〜8秒間隔で穏やかに突いて後足底を刺激する。5つの試験を行い、各同側後足の機械的閾値を評価する。歩行解析のために、熱傷の5日前から10〜15分間、歩行解析システムにマウスを毎日順応させる。試験当日は、手術用ガウン、手袋、マスクを着用した後、マウスを行動試験室に持ち込み、試験前に30分間順応させた後、歩行解析プログラムを起動し、新規実験作成メニューをクリックしてデータ保存フォルダを指定します。指定後、最大走行時間を5秒、最大許容速度変動を50%に設定し、次にプログラムの設定タブで登録カメラを選択し、歩道の長さを30センチに設定します。次に、プログラムメニューの取得タブで[取得を開く]を選択し、ステータスメッセージに基づいて[背景のスナップ]ボタンをクリックして、空の歩道の背景画像を取得します。次に、取得開始ボタンをクリックし、左右の横断可能な歩道の入り口にマウスを置きます。記録は、マウスの自由な動きに続いて自動的に開始されます。記録が成功したら、プログラムメニューの取得タブから[実行の分類]を選択し、分析する実行を選択して自動分類ボタンをクリックします。自動分類が完了したら、鼻、生殖器の認識、および足の誤認識を各実行でジャンク データから削除し、データを分析します。行動試験室で30分間の順応期間を過ごした後、マウスを15センチメートルの水を含む透明なプレキシグラスシリンダーに15分間置く。翌日、同じ条件下でマウスを再びシリンダーに入れ、マウスが登山や水泳をやめて頭で水面上に浮かぶ不動時間を測定します。次に、正常な運動機能を測定するために、装置の底部から16センチメートル上に吊り下げられた回転円筒形のプラットフォーム上に動物を置く。火傷を誘発する前に、マウスにロタロッドで1日1回少なくとも5日間訓練させる。薬物投与後、20分毎にロタロッド試験を2分間、2分間のカットオフ時間で実施する。マウスが落下することなく毎分15回転の一定速度で回転ロッド上を走る時間の長さを測定します。火傷誘発マウスの足の離脱閾値は、対照群と比較して1日目以降に減少し、7日間持続した。アセトアミノフェン投与は、足離脱閾値の火傷誘発性減少を有意に減少させた。さらに、曲線下面積分析は、アセトアミノフェン投与が火傷傷害誘発性機械的アロディニアを有意に減少させることを示した。後足の印刷面積も火傷の誘発後に有意に減少し、7日間持続した。アセトアミノフェン投与は、ビヒクル治療と比較して後足印刷面積を有意に改善した。さらに、火傷は、1日目以降に同側後肢の単一姿勢を減少させ、この減少は7日間維持された。後足単姿勢はアセトアミノフェン投与により改善された。強制水泳試験では、火傷誘発マウスの不動時間は、対照群と比較して傷害後7日間増加した。アセトアミノフェン投与は、火傷傷害誘発性不動時間の増加を有意に減少させた。ロタロッド試験において、熱傷誘発マウスの走行時間は、火傷誘発後7日目の対照群のものと類似していた。対照的に、アルファキソロン処置マウスの実行時間は約60分有意に減少し、この熱傷傷害法が運動障害を引き起こさないことを示している。正確な結果を得るために、各動物に適用される火傷の程度は一定に保たれなければなりません。本研究で提案された様々な行動実験的解析方法は、疼痛、歩行障害、うつ病様行動に関連する他の動物モデルにも適用することができる。提案する解析手法は、火傷誘発後の様々な症状を定量化することができ、今後の火傷関連研究・治療法の開発に資すると考えられる。
