単粒子クライオEMは、生体高分子の高分解能構造決定のための主流の構造生物学技術となっています1。単粒子の再構築は、2次元(2D)粒子画像を抽出するために膨大な数のガラス化サンプルの顕微鏡写真を取得することに依存し、その後、生物学的高分子の3次元(3D)構造を再構築するために使用されます。DEDの開発前に、単粒子の再構成から達成された分解能は4から30Å4,5の範囲に及んだ。最近、単粒子クライオEMからの達成可能な解像度は1.8 Å6を超えています。DEDおよび自動データ収集ソフトウェアは、データ収集のための人間の介入が最小限に抑えられるこの解決革命7の主要な貢献者となっています。一般に、クライオEMイメージングは、低電子線量率(20-100 e/Å2)で行われ、生体試料の電子ビームによる放射線損傷を最小限に抑え、画像内の低信号対雑音比(SNR)に寄与します。この低いSNRは、単粒子分析を用いた生体高分子の高分解能構造の特性解析を妨げる。

User-friendly, High-throughput, and Fully Automated Data Acquisition Software for Single-particle Cryo-electron Microscopy
