細胞外小胞(EV)は、ナノサイズの脂質膜結合粒子で、そのサイズによって分類されます: エクトソーム (100-500 nm) およびエキソソーム (50-150 nm)1.EVには、タンパク質、核酸、脂質など、さまざまな生体分子が含まれています。これらの生体分子は、貨物としてカプセル化される前に細胞から発生し、EV1,2,3を介して細胞外空間に放出されます。
その貨物の多様性により、EVは細胞間通信において積極的な役割を果たすと考えられています。細胞によるEVの放出および取り込みにより、細胞間の生体分子の移動が可能となる4,5.EV貨物をセルに導入すると、受信者細胞の機能やホメオスタティック状態が変われ得る4,5,6.EV は複数の経路を通じて内部化されます。しかし、正確なメカニズムは正確には実証されていません。
遺伝子タグ付けなどのEV取り込みアッセイの大部分は、個々のEV7に蛍光標識を付ける。得られた信号は、マイクロプレート光度計、フローサイトメトリー、または顕微鏡検査によって測定することができ、各技術は大きな制限を有する。マイクロプレートフォトメーター、フローサイトメトリー、または標準的な2次元(2D)顕微鏡では、内部化されたEVsと表面的に取り付けられたEVsを区別できません。さらに、これらの各技術に必要なサンプル調製は、EV取り込み評価に追加の問題を導入する可能性があります。例えば、EV取り込み分析の前にトリプシンで付着した細胞を持ち上げると、表面的に取り付けられたEVが細胞表面10,11に切断されることがあります。トリプシンはまた、細胞表面と相互作用し、細胞およびEV表現型に影響を与える。さらに、トリプシンは表面的なEVを完全に切り離すものではなく、孤立した集団を歪める可能性があります。
蛍光色素でEVに正確にラベルを付けるには、残留色素7を除去するために追加の洗浄工程が必要です。受け入れられた分離技術は、EVの分離中に起こる凝固による偽陽性シグナルにも寄与する可能性がある。例えば、シリアル超遠心分離(UC)は、EVを分離し、固定染め染料を除去するために広く使用されています。しかし、UCはEVを共沈殿させ、残存色素は偽陽性シグナル12,13を引き起こす可能性がある。カラムベースのろ過などの他のナノ濾過方法も、非固定化色素除去にも広く使用されています。カラムマトリックス内で相互作用するEVと色素の複雑な性質により、複雑な入力14,15,16によってカラムの分子カットオフが変化するため、残存染料が不完全に除去される可能性があります。
現在のプロトコルは、蛍光標識された単離されたEVを単離して洗浄するためのナノ濾過ベースのマイクロ流体装置を提案している。ナノ濾過ベースのマイクロ流体デバイスは、流体支援分離技術(FAST)17,18を介して効率的なろ過を提供することができます。FAST はフィルター全体の圧力降下を低減し、EV と色素の間の潜在的な凝集を減らします。残留色素を効率よく除去することで、蛍光標識されたEVの品質とアッセイの特異性を高めることができる。
共焦点顕微鏡は、細胞表面に内部化されたEVと表面的に結合したEVを区別し、時空間的な解像度におけるEV取り込みの細胞機構を包括的に調べることができます19,21,22,23,24,25。例えば、Sungらは、開発された生細胞レポーターを用いてエキソソームライフサイクルの可視化を説明した。3次元(3D)およびポスト画像処理ツール20の共焦点顕微鏡を用いて、内部化されたEVの位置を検出し、解析した。小さなEV(40〜200nm)のサイズは光学顕微鏡の解像度限界を下回っていますが、光検出器は蛍光放射の増強を検出できるため、蛍光標識されたEVは共焦点顕微鏡で検出できます。したがって、細胞内の蛍光標識されたEVの細胞内局在化は、EVおよび周囲の細胞小器官の複数のZ積層画像を取得することによって正確に決定することができる。
さらに、3D再構成とポストデータ処理により、内部化、表面化、自由浮遊型のEVの位置に関するさらなる洞察を得ることができます。これらのプロセスを共焦点顕微鏡で提供するタイムラプスライブセルイメージングと組み合わせることで、EV取り込みのレベルを正確に評価でき、リアルタイムでのEV取り込みの追跡も可能です。また、EVの入稿解析は、細胞内機能に対するインターナライズされたEVの共同局在化を評価することによって、共焦点顕微鏡を用いて行うことができる。本プロトコルは、ナノ濾過ベースのマイクロ流体デバイス17,26、共焦点顕微鏡法、およびポスト画像解析を用いてEV取込アッセイを行う方法論を記述する。

このプロトコルは、細胞レベルでEVを分析するための堅牢な方法論と、効率的なEV取り込みおよびEV追跡分析のための実用的なアプローチを提供します。EV標識のこの方法は、EVと色素の共沈殿を排除し、EV信号を増強します。EV取り込みは、内部化されたEVと表面的なEVを区別するために、体積解析によって測定されます。EVは活動的な貨物であり、従って、その取り込みは生化学的分子の移動を可能にする。EVベースの薬物送達とがん治療に関する研究では、この技術を評価ツールにすることができます。EV取り込みは細胞間コミュニケーションの役割を果たし、がん生物学、神経科学、薬物送達など様々な研究分野で研究されています。このプロトコルは、効率的なEV取り込みアッセイを促進する可能性がある。細胞外小胞(EV)の単離のために、ナノ濾過ベースのマイクロ流体装置のサンプルチャンバーに、前処理された細胞培養培地を1ミリリットル注入する。デバイスを操作するには、ベンチトップ回転機でデバイスを3000 RPMで10分間回転させます。紡糸後、配管または吸引によって廃液を除去し、次に、細胞培養培地の更に2ミリリットルを処理するためにこの手順を2回繰り返す。次に、分離したEVを洗浄し、PBSを1ミリリットルサンプルチャンバに注入し、ベンチトップスピニングマシンでデバイスを回転させます。EVの免疫蛍光標識の場合、100マイクロリットルの単離されたEVを含むデバイスの溶出穴にEV特異的抗体の1ミリリットル当たり1マイクログラムを注入します。その後、プレートシェーカーの暗闇の中で1時間インキュベートし、サンプル全体の抗体の均等な分布を確保します。次に、溶出孔に粘着テープを貼り付けます。PBSを1ミリリットルのサンプルチャンバーに注入して残留抗体を洗い流し、サンプルチャンバーが空になるまでデバイスを回転させます。サンプルチャンバーに別のミリリットルのPBSを注入し、先に示したようにデバイスを回転させて洗浄を繰り返します。チャンバーから残りの液体を取り除いた後、蛍光標識されたEVを膜室から琥珀色のチューブまたはマイクロチューブにピペットします。使用するまで、チューブを光から保護します。4回10から4番目のPC3細胞を1ミリリットルの培地で35ミリリットル皿に種分します。最適な細胞培養条件で細胞が一晩接着することを可能にする。翌日、付着した細胞をエキソソーム枯渇した培地で2回洗います。エキソソーム枯渇培地を用いて蛍光標識したEVを適切な濃度に希釈した後、希釈したEVを接着された標的細胞に加え、所望の実験時間にインキュベートする。エキソソームフリー培地で細胞を3回洗浄して非内在性EVを除去した後、CMTMRのミリリットル当たり1マイクログラムで接着した細胞の細胞質にラベルを付け、最適な細胞培養条件でインキュベートする。生細胞イメージングの場合は、準備した細胞をステージ上のインキュベーターに入れる。制御サンプルに基づいて撮像パラメータを設定した後、ターゲットセルの深さとZ方向の積層サイズの範囲を決定して3D共焦点画像を取得し、画像取得を細胞特異的な色素とEV特異的染料の両方の複数のZスタック画像に同時に設定します。画像処理には、自動画像処理ソフトウェアを利用します。セルの仮想サーフェスを作成するには、[新しいサーフェスを追加] をクリックします。仮想セルサーフェスを設定するには、[追加]ボタンをクリックし、フィルタタイプとして[品質]を選択します。目視検査による下限の適切な値をしきい値にし、上限の最大値を設定し、[完了]ボタンをクリックして[次へ]をクリックして、EVの仮想ドットを構築し、[新しいスポットを追加]ボタンをクリックします。アルゴリズム設定で、異なるスポットサイズと最短距離計算を選択し、[次へ]をクリックして、ソースチャンネルとしてチャンネル1アレクサFluor 488を選択します。[スポット検出]で推定 XY 直径に適切な値を入力し、[次へ]をクリックします。仮想 EV ドットを設定するには、[追加] ボタンをクリックし、フィルターの種類として [品質] を選択します。目視検査で下限しきい値を設定し、[スポット領域タイプ] の [次へ] をクリックし、[絶対強度] を選択して、[次へ] をクリックします。EV ドットの領域をしきい値にするには、目視検査で [領域のしきい値] に適切な値を入力します。[領域ボリューム]の下の直径を選択し、[完了]をクリックします。構築したサーフェス内のグループ化されたスポットを分割するには、[スポットを構築]をクリックして[フィルタ]に移動します。次に、[追加]ボタンをクリックし、フィルタ タイプとして[サーフェス、サーフェス 1 までの最短距離]を選択します。下限の最低しきい値と上限値の適切な値を設定した後、[セクションの複製]をクリックして新しいスポットボタンをクリックします。セル内の EV の自動カウントの場合は、[作成されたスポット 1] の選択をクリックし、[統計情報] に移動して、[スポットの合計数] から値をエクスポートします。セルの数を取得するには、[作成サーフェス 1] をクリックし、[統計情報] に移動して[全体]から[サーフェスの総数]の値をエクスポートします。最後に、[統計] の [詳細] タブに移動してボリュームをエクスポートします。ナノ濾過ベースのマイクロ流体デバイスを用いてPC3細胞培養培地から分離されたEVを、フルオロ-4-共役EV特異的抗体で標識し、3D共焦点顕微鏡を用いて可視化した。内部化されたEVは、プンクタとして視覚化され、これらの画像の個別の EV.Post 処理により、細胞内へのEV内在化の可視化と定量が可能になります。EV取り込みアッセイの結果は、EV取り込みのレベルがインキュベーション期間の長さに依存することを示しています。内部化された EV の数を受信者のセル ボリュームに正規化して、指定したセルの EV の実際の取り込み速度を決定できます。この手順により、非内部化された EV を体系的に除外して、内部化された EV の数を正確に測定できます。内部化されたEVドットのサイズ分布を以下に示します。したがって、重要なステップは、EV標識用マイクロ流体デバイスの利用、蛍光EVの視覚化時のバックグラウンド蛍光の最小化、そして内部化されたEVと表面的なEVを決定するためのポストイメージング分析です。したがって、細胞内EV追跡は3D平面上でリアルタイムで行うことができるため、空間的/時間的解像度に関するEVの人身売買解析や、細胞内小器官とのEVの共同局在化も可能です。したがって、この技術は、EVの細胞通信分野の研究者に対して、細胞内および細胞外マトリックス内のEVを調査するより簡単で効率的な方法を提供すると考えています。
