1. 对渐变进行分层

</ol>2. 红细胞沉降

</ol>3. 残留红细胞的裂解
<ol><li>轻轻吸出上清液而不破坏沉淀。</li>

</ol>4. 确定中性粒细胞分离质量

已经发表了许多关于人类中性粒细胞分离的方法10，22。它们主要分为两类，有一些共同的战略。第一类是基于抗体的，通过阳性或阴性选择。阳性选择将直接标记嗜中性粒细胞，因此提供高纯度的细胞群，尽管它也会导致快速细胞活化，细胞死亡以及嗜中性粒细胞23的不需要标记。阴性选择，尽管使细胞未标记并给出非常纯净的群体，但加速了中性粒细胞死亡，尽管确切的机制尚不清楚（图5）。基因或蛋白质表达在阳性或阴性选择后是否也改变需要进一步研究。此外，由于消耗其他类型的细胞所需的抗体量，这些方法不能输出大量的嗜中性粒细胞。然而，基于抗体的测定仍然可以用于短期培养和较小规模的实验，并且是需要非常高细胞纯度的实验的首选方法，例如基因和蛋白质表达研究。
第二种类型的隔离方法是基于梯度和密度的。它通常涉及Percoll，Ficoll-Paque或其他多糖/聚乙烯吡咯烷酮组分，并利用离心力根据细胞密度分离不同类型的血细胞。这些方法通常与葡聚糖沉淀红细胞相辅相成。这些方法可以处理更大规模的起始材料，也可以实现高纯度。基于密度的分离的一个警告是将其他远不那么丰富的粒细胞（主要是嗜酸性粒细胞）与嗜中性粒细胞分离效率低下，因此，这是所提出的方案的主要局限性，因为即使存在小细胞污染也会影响中性粒细胞反应24。
这里介绍的是一种基于梯度隔离的总结方法，对先前的方法10、22进行细化。我们利用目前对中性粒细胞特异性的低估来可靠地分离具有有限残留血小板和红细胞的纯人嗜中性粒细胞，从而防止中性粒细胞活化和加速死亡。最关键的步骤是梯度的分层，通过在血液下方添加密度梯度介质以获得尖锐的界面，可以更有效地获得梯度。快速检查PBMC环和离心后的梯度可以揭示可能的污染，活化和低产量。当使用白细胞分离膜或白蛋白涂层时，稀释血液很重要，因为过多的细胞密度会导致细胞聚集，导致杂质和细胞活化。
该方案应在2小时内完成以确保细胞新鲜度，并且涉及密度梯度培养基，葡聚糖和裂解的步骤应立即完成，因为暴露于这些溶液可以改变嗜中性粒细胞。使用该协议，嗜中性粒细胞的预期产率至少为约1000万/ 10mL全血和至少约6000万/ 10 mL的黄褐色外套。分离质量的评估应如下：活化的中性粒细胞应小于10%，淋巴细胞污染应低于5%，嗜酸性粒细胞最小（同侧散射但前向散射群体较低），细胞活力应大于90%。较低的纯度可能是由于密度梯度培养基的分层或储存不当或由于起始血液产品的质量和新鲜度造成的。

嗜中性粒细胞用来对抗感染的分子对微生物和宿主有毒。因此，寿命和老化/垂死嗜中性粒细胞的适当去除受到高度调节，并且它们在循环中的寿命有限（<48小时）3。由于这种短暂的生存期，人体平均每天产生1000亿个新的嗜中性粒细胞来维持人口稳态4。紧急造粒术可以进一步增加炎症和感染期间血液中嗜中性粒细胞的释放，包括成熟和未成熟5。中性粒细胞在先天免疫反应中的重要性由获得性或先天性中性粒细胞减少症患者强调，他们容易受到细菌和真菌感染6。

当使用密度梯度纯化嗜中性粒细胞时，血液和密度梯度培养基之间的界面尽可能锋利，并且在离心后保持明显的层分离至关重要（步骤1.4）。红细胞裂解后，缓冲液应为红色且不浑浊（步骤3.3）。如果制剂浑浊，可能需要第二轮裂解（步骤3），尽管这可能会影响中性粒细胞的存活率（图1）。裂解后，当纯度优先时，建议使用低速离心（200×g），因为它可以大大减少血小板污染。 然而，高速离心（400×g）以牺牲纯度为代价提高了产量（步骤3.4，图2）。中性粒细胞分离后，荧光激活的细胞分选可用于评估分离纯度（步骤4.1），并且应选择显微镜检查。虽然仅细胞的FSC/SSC分布提供了细胞分离质量的估计（图3A），但使用特定的细胞标志物应该是首选。在这种情况下，最常见的污染细胞群用特异性抗体和CD66b染色，特异性地在粒细胞上表达。CD45染色用于区分白细胞（CD45 +）和红细胞和血小板（CD45-）。
其他污染物包括淋巴细胞（CD3 +或CD19 +，图3F），单核细胞（CD14 +，图3D）和嗜酸性粒细胞（CD193 +，图3G）。CD11b是在骨髓谱系上表达的整合素;嗜中性粒细胞和单核细胞是CD11b +，而淋巴细胞是CD11b-（图3C）。由于中性粒细胞活化会影响下游实验，因此应评估CD62L的表达;中性粒细胞一旦被激活就会变成CD62L-（步骤4.1）。肽fMLP可用作CD62L脱落的阳性对照（图4）。在进行测定之前评估嗜中性粒细胞的健康状况也很重要;中性粒细胞的半衰期相对较短，活化可以进一步缩短它（步骤4.2）。标准的膜联蛋白V和PI染色可以在指定时间提供有关嗜中性粒细胞培养物的活/死状态的信息（图5）。
