1. 建立 CE-ESI-MS 系统
<ol><li>设置一个 CE-ESI-MS 系统，该系统由商用 CE 系统和配备安捷伦喷射流 （AJS） 电喷雾电离 （ESI） 源的三重四极杆串联质谱仪组成。CE-ESI-MS喷雾器套件和等度LC（液相色谱）泵是必需的。</li>

</ol>2. 制备缓冲液、毛细管和CE-MS系统
<ol><li>制备CE电泳缓冲液：用氢氧化铵将40mM乙酸铵的pH调节至9.0。建议使用 250 mL 容量瓶。用 0.2 μm 孔径的膜过滤器过滤 250 mL 缓冲液。确保仅使用超纯去离子水和MS级试剂。 注意：该缓冲液可以在室温下保存2-3周或在冰箱中保存数月。</li>
	<li>准备护套液：在 250 mL 瓶中混合 100 mL 超纯水和 100 mL LC-MS 级异丙醇。每周至少更换一次鞘液。使用高分辨率质谱仪时，将质谱参考添加到鞘液中。</li>
	<li>安装护套液：以 5 mL/min 的速度吹扫 5 分钟，并将流速设置为 1 mL/min（10 μL/min 进入 CE-MS 喷雾器）。泵压约为 180 bar。确保回收管连接回护套液瓶中以重复使用溶剂。</li>
	<li>准备毛细管：购买具有紫外线检测窗口的CE-MS毛细管（内径50μm和外径365μM，长度为125厘米）。用户还可以从专业分销商处获得更便宜的条状熔融石英毛细管。切割长度为100厘米的毛细管。用带有旋转金刚石刀片的毛细管柱切割机正确切割毛细管两端，并用打火机去除两端 2-3 厘米的聚酰亚胺涂层。用异丙醇清洁毛细管表面。</li>
	<li>安装毛细管：将毛细管匹配到CE-MS盒中。单击 “更换暗 盒”按钮并将暗盒安装到 CE 设备中。毛细管的入口端比电极低约 2 mm。在进样过程中，确保入口端低于样品表面。</li>
	<li>激活毛细管：首次使用前，用1M NaOH冲洗毛细管，然后用水冲洗10分钟，CE电泳缓冲液冲洗15分钟。</li>
	<li>将毛细管端插入CE-MS喷雾器：将毛细管轻轻放入CE-MS喷雾器中，并确保毛细管端从喷雾器尖端突出约0.1毫米。确保用放大镜和喷雾器中的调节螺钉精确调整毛细管出口端。将喷雾器插回离子源，避免接触调节螺钉。执行此操作时，MS 处于待机模式。</li>
	<li>检查 ESI 喷雾：检查 ESI 喷雾器在全扫描模式下的稳定性。总离子电泳图的波动必须在5%以内。</li>
	<li>使用 InsP 标准品进行测试运行：使用2 μM InsP3-InsP 8 标准品（通过定量 31P NMR15，27调整）的混合物进行测试运行，在50 mbar下进样10秒（10 nL）。设置详细的ESI和MS参数，如 表1所示。CE 电流约为 26 μA。峰宽约为0.4-0.5分钟。确保信噪比至少达到 400。</li>
</ol>3. 从哺乳动物细胞中提取可溶性肌醇磷酸盐
注意：HCT116NIH 细胞是Stephen Shears28的一份礼物。HCT116UCL 细胞来自Saiardi的实验室26。
<ol><li>接种细胞<ol><li>在补充有10%胎牛血清（FBS）的Dulbecco改良鹰培养基（DMEM）的T75烧瓶中培养HCT116NIH 或HCT116UCL 细胞，在37°C的高湿度CO2 气氛（进一步称为标准条件）中培养。</li>
		<li>用磷酸盐缓冲盐水（PBS）（5mL）洗涤HCT116NIH 和HCT116UCL 储备培养物，并在标准条件下用胰蛋白酶 - 乙二胺四乙酸（EDTA）（3 mL，0.25%）孵育细胞，直到它们完全分离。通过加入培养基（7mL）淬灭胰蛋白酶活性，将细胞收集到离心管中，然后离心（200× g，3分钟）。</li>
		<li>取出上清液并将细胞重悬于培养基（10mL）中。计数细胞并通过台盼蓝排除法确定活力。</li>
		<li>将细胞（每次测定600万个HCT116细胞）接种到150 mm培养皿中，并总共调整20 mL细胞培养基。在接种之前，在离心管中预混合培养基和细胞，以实现培养皿中细胞的均匀分布。当需要按细胞数归一化时，准备一个平行培养皿。</li>
		<li>在标准条件下培养细胞72小时。细胞将达到约80%-90%的汇合。</li>
	</ol></li>
	<li>用NaF和细胞收获调节肌醇磷酸水平<ol><li>NaF处理：在收获到培养基中前1小时加入NaF（10mM）。通过旋转板/移液混合培养基，并在标准条件下孵育细胞60分钟。</li>
		<li>NaF处理后，从细胞中取出培养基并将细胞放在冰上。</li>
		<li>用PBS（5mL，4°C）洗涤细胞两次，并从培养皿中完全除去PBS。</li>
		<li>加入高氯酸（PA）（1毫升，1M，4°C）。确保用PA覆盖整个表面（随着蛋白质沉淀，细胞会变白）。将细胞在4°C的倾斜台上孵育10分钟。</li>
		<li>将PA收集到离心管中，并通过离心（17，000× g，5分钟，4°C）除去污染碎片。将上清液添加到制备的TiO2 珠中，用于下拉InsP。</li>
		<li>用PBS（5mL，r.t.）清洗提取后的培养皿两次以进行脱酸;从培养皿中完全去除PBS。</li>

		<li>从平行培养皿中测定细胞数：如步骤3.1.2中所述通过胰蛋白酶收获平行培养皿的细胞（对150 mm培养皿使用5 mL胰蛋白酶-EDTA）并除去培养基。将细胞沉淀重悬于PBS（5 mL）中，适当混合并计数细胞。在收获前通过直接淬灭执行此步骤以获得具有代表性的细胞计数。此外，使用适当的方法（例如，使用多尺寸机）测量细胞的体积。</li>
	</ol></li>
	<li>肌醇磷酸盐的TiO 2 富集 注意：为避免磷酸化化合物的酸性分解，请执行所有富集步骤直到在冰上洗脱，并将所有试剂冷却至4°C。 将提取时间保持在最短（1.5-2小时）。使用 1 M PA 执行所有提取步骤。<ol><li>珠子的制备：用ddH2O（1mL）洗涤TiO2珠（每个样品5mg）和离心机（3，500×g，1分钟，4°C）。取出 ddH2O 并用 PA （1 mL） 洗涤珠子。通过离心（3，500×g，1分钟，4°C）除去PA。将磁珠重悬于PA中（每个样品50 μL）。</li>

		<li>离心样品（3，500× g，1分钟，4°C）并弃去上清液。用PA（500μL）洗涤珠子并离心（3，500× g，1分钟，4°C）。弃去上清液并重复洗涤步骤。</li>
		<li>将 NH4OH（200 μL，3%）加入珠子中并重悬。以r.t旋转样品5分钟。</li>
		<li>离心样品（3，500 x g ，1分钟）并将上清液转移到新的离心管中。</li>
		<li>重复洗脱步骤3.3.4和3.3.5并合并洗脱液。丢弃珠子。</li>
		<li>离心合并的淋洗液（17，000× g，1分钟，4°C）以除去任何不溶性残留物。</li>
		<li>在真空蒸发（70分钟，60°C，v-AQ）下完全干燥上清液。向含有InsPs的干燥提取物中加入ddH2O（50μL）。涡旋混合样品直至完全溶解。将样品储存在-20°C直至CE-ESI-MS分析。</li>
	</ol></li>
</ol>4. 执行 CE-ESI-MS 运行
<ol><li>制备含有 40 μM [13 C 6]1，5-（PP）2-InsP 4、80 μM [13 C 6]5-PP-InsP 5、80 μM [13 C 6]1-PP-InsP 5、400 μM [13 C 6]InsP 6 和 400 μM [13C 6]Ins（1，3，4，5，6）P 5 的内标混合物。借助经认证的参考标准品（即磷酸乙酸），通过定量 31P 和 1H NMR 测定 SIL IS 溶液的浓度。 注：上述所有纯度高于96%的SIL内标（IS）均由Fiedler集团15，27合成和提供。与核苷酸一样，这些IS可以携带许多结晶水分子和不同的反离子。建议通过定量31P NMR测定13C6标准溶液的浓度，而不是称量物质并计算浓度。</li>
	<li>在CE样品瓶中将10 μL样品与0.5 μL内标混合物混合。2 μM [13 C 6]1，5-（PP）2-InsP 4、4 μM [13 C 6]5-PP-InsP 5、4 μM [13 C 6]1-PP-InsP 5、20 μM [13 C 6]InsP 6 和 20 μM [13C 6]Ins（1，3，4，5，6）P 5 是样品内的最终浓度。</li>
	<li>使用补给系统时，单击更换瓶按钮，将准备好的 250 mL CE 电泳缓冲液放入电解质 瓶 中，然后单击 清洁管。将补给针放在水瓶中。</li>
	<li>设置ESI和MS参数，如 表1所示。使用含有肌醇聚磷酸盐标准品混合物的源优化器优化源参数。使用包含所有标准的 Masshunter 优化器获取多反应监测 （MRM） 设置。调整不同仪器的 ESI 和 MS/MS 设置。</li>
	<li>运行 InsP 数据提取并检查结果（图 2）。设置有更多样本时的顺序。</li>
	<li>测量后让MS进入待机模式。请勿关闭液相色谱泵。鞘液的流动保护喷雾针。当没有护套液体供应时，将 CE-ESI-MS 喷雾器更换为 LC-ESI-MS 喷雾器。</li>
</ol>5. 数据分析
<ol><li>打开定量分析（用于QQQ）软件，为所有样品创建一个批次。</li>
	<li>根据获取的 MRM 数据创建新方法。将 [13C6]InsP 设置为内部标准 - （ISTD）。检查MRM化合物设置、保留时间设置、ISTD设置、浓度设置和限定符设置。通过验证并退出以将方法应用于当前批处理。保存方法。</li>
	<li>检查批次中每个峰是否积分正确;否则，请手动积分峰值。</li>
	<li>将结果导出到电子表格中。通过将分析物峰响应与已知浓度的SIL IS的相应峰响应进行比较，对肌醇（焦磷酸）进行定量。5-PP-InsP 5、InsP 6和Ins（1，3，4，5，6）P5的理论和实验校准曲线如表2所示，线性范围。 注意：应用理论校准曲线的先决条件是使用目标分析物的高质量同位素模式和可信浓度。评估线性范围。当完全获取上述同位素标准品不切实际时，校准曲线对于绝对定量至关重要。</li>
