Selectief doorlatende hydrogel-microcapsules geladen met verschillende soorten DNA-gecodeerde sensorontwerpen kunnen volgens dit protocol worden bereid. Een van de onderscheidende kenmerken van de LbL-benadering is het vermogen om de complexiteit van microcapsules aan te passen tijdens de bottom-up assemblage, die meestal begint met de adsorptie van moleculaire soorten op offersjablonen. Het gebied van synthetische biologie biedt unieke mogelijkheden om detectiemogelijkheden te ontwikkelen door gebruik te maken van natuurlijke mechanismen die door micro-organismen zijn ontwikkeld om hun omgeving en potentiële bedreigingen te monitoren. Belangrijk is dat deze detectiemechanismen meestal gekoppeld zijn aan een reactie die deze micro-organismen beschermt tegen schadelijke blootstelling, genexpressie reguleert om negatieve effecten te verminderen of inname van giftige materialen te voorkomen. Er zijn aanzienlijke inspanningen geleverd om deze micro-organismen te ontwikkelen om sensoren met hele cellen te maken die profiteren van deze natuurlijke reacties, maar ze opnieuw richten om nieuwe doelen te herkennen en / of een meetbaar signaal te produceren dat kan worden gemeten voor kwantificeringsdoeleinden (meestal fluorescentie)1,2. Momenteel wijzen zorgen over het gebruik van genetisch gemodificeerde micro-organismen (GGO's), met name bij introductie in het milieu of het menselijk lichaam, als gevolg van lekkage van hele cellen of een deel van hun genetisch materiaal, zelfs als het is ingekapseld in een polymeermatrix, erop dat alternatieve manieren nodig zijn om deze detectiebenaderingen te benutten3.
Een krachtige aanpak om de voordelen van op micro-organismen gebaseerde detectie te benutten zonder de zorg voor de inzet van GGO's is het gebruik van in vitro transcriptie/translatie (IVTT) systemen. Vanuit een praktisch perspectief bestaan IVTT-systemen uit een mengsel dat de meeste celcomponenten in een actieve toestand bevat en dat op verschillende manieren uit cellen is "geëxtraheerd", waaronder ultrasoonapparaat, kralenkloppen of andere4. Het eindproduct van dit proces is een biochemisch reactiemengsel dat al is geoptimaliseerd om transcriptie en translatie uit te voeren en dat kan worden gebruikt om verschillende sensoren in een "open vat" -formaat te testen, zonder de beperkingen die gepaard gaan met het gebruik van hele cellen (membraandiffusie, transformatie-efficiëntie, celtoxiciteit, enz.). Belangrijk is dat verschillende sensorcomponenten kwantitatief kunnen worden toegevoegd en hun effect kan worden bestudeerd door verschillende optische en spectrometrische technieken, zoals we hebben aangetoond5. Het is opgevallen dat de prestaties van IVTT-systemen inconsistent kunnen zijn; Recente studies hebben echter benaderingen aangetoond om hun voorbereiding en karakterisering te standaardiseren, wat van grote hulp is bij het bestuderen van hun prestaties in sensorontwerp6. Onlangs zijn veel voorbeelden van IVTT-systemen die worden gebruikt om op papier gebaseerde assays te maken door de lyofilisatie van hun componenten in papieren matrices gedemonstreerd, waaronder detectie van zware metaalionen, geneesmiddelen, quorumdetectie-elementen en andere 7,8,9. Een opwindende toepassingsruimte voor IVTT-gebaseerde sensoren is hun gebruik in detectietoepassingen in verschillende soorten omgevingen, waaronder bodem, water en het menselijk lichaam. Om deze IVTT-systemen in deze uitdagende omgevingen te implementeren, moet een inkapselingsbenadering worden geïmplementeerd om de IVTT-componenten te bevatten en te beschermen tegen degradatie.
De meest voorkomende inkapselingsbenaderingen voor IVTT-systemen omvatten het gebruik van lipidecapsules, micellen, polymersomen en andere nauw gesloten microcontainers10,11,12. Een nadeel van deze aanpak is de noodzaak om passieve of actieve mechanismen in te bouwen om materialen in en uit de containers te transporteren om communicatie met de externe omgeving mogelijk te maken en detectiemogelijkheden te bieden. Om enkele van deze problemen op te lossen, rapporteert de studie hier een methode die een eenvoudige maar effectieve aanpak biedt om de coderingsmaterialen voor verschillende sensorontwerpen in te kapselen die in IVTT-systemen moeten worden uitgedrukt. Deze aanpak is gebaseerd op het gebruik van Layer-by-Layer (LbL) depositie van een biopolymeer in de aanwezigheid van de plasmiden van belang om holle microcapsules met hoge porositeit te creëren, waardoor het beschermde genetische materiaal kan interageren met de verschillende componenten van de IVTT van keuze. De studie toonde aan dat ingekapselde plasmiden transcriptie en translatie konden sturen wanneer ze binnen deze polymere matrix werden geactiveerd, zoals blijkt uit de respons van een plasmide-gecodeerde aptamer en een riboswitch op hun overeenkomstige doelen. Bovendien beschermt deze LbL-coating de plasmiden maandenlang zonder speciale bewaarcondities.

preparation of multifunctional silk-based microcapsules loaded with dna plasmids encoding rna aptamers and riboswitches

We introduceren een protocol voor de bereiding van met DNA beladen zijdefibroïnemicrocapsules via de Layer-by-Layer (LbL) assemblagemethode op offerende bolvormige kernen. Na adsorptie van een eerste laag en DNA-plasmiden werd de vorming van robuuste microcapsules vergemakkelijkt door β-vellen in de secundaire zijdestructuur te induceren tijdens acute uitdroging van een enkele zijdelaag. Vandaar dat de gelaagdheid plaatsvond via meerdere waterstofbindingen en hydrofobe interacties. Na adsorptie van meerlaagse schillen kunnen de kernschilstructuren verder worden gefunctionaliseerd met gouden nanodeeltjes (AuNP's) en/of antilichamen (IgG) om te worden gebruikt voor teledetectie en/of gerichte toediening. Het aanpassen van verschillende belangrijke parameters tijdens sequentiële depositie van belangrijke macromoleculen op silicakernen, zoals de aanwezigheid van een polymeerprimer, de concentratie van DNA en zijde-eiwit, evenals een aantal geadsorbeerde lagen, resulteerde in biocompatibele, DNA-beladen microcapsules met variabele permeabiliteit en DNA-belastingen. Bij het oplossen van silicakernen toonde het protocol de vorming aan van holle en robuuste microcapsules met DNA-plasmiden geïmmobiliseerd op het binnenoppervlak van het capsulemembraan. Het creëren van een selectief doorlaatbaar biocompatibel membraan tussen de DNA-plasmiden en de externe omgeving bewaarde het DNA tijdens langdurige opslag en speelde een belangrijke rol in de verbeterde outputrespons van ruimtelijk beperkte plasmiden. De activiteit van DNA-sjablonen en hun toegankelijkheid werden getest tijdens in vitro transcriptie- en translatiereacties (celvrije systemen). DNA-plasmiden die coderen voor RNA-oplichtende aptameren en riboswitches werden met succes geactiveerd met overeenkomstige analyten, zoals werd gevisualiseerd tijdens de lokalisatie van fluorescerend gelabelde RNA-transcripten of GFPa1-eiwit in de schaalmembranen.

Hier richt de studie zich op de functionaliteit van DNA-sjablonen die coderen voor verschillende sensorontwerpen (twee soorten RNA-gereguleerde transcriptie / translatie-elementen) na inkapseling in zijden eiwitcapsules.
