Aminozuren zijn organische verbindingen die een amino (-NH2) en carboxyl (-COOH) functionele groepen bevatten met een variabele zijketen die specifiek is voor elk aminozuur. Over het algemeen staan aminozuren bekend als het basisbestanddeel van eiwitten. Meer recent zijn nieuwe toepassingen en functies van aminozuren opgehelderd. Individuele aminozuren kunnen bijvoorbeeld worden gemetaboliseerd om intermediaire metabolieten te genereren die bijdragen aan bio-energetica, functioneren als enzymatische cofactoren, reactieve zuurstofsoorten reguleren of worden gebruikt om andere aminozuren te synthetiseren 1,2,3,4,5,6,7,8,9,10 . Veel studies tonen aan dat aminozuurmetabolisme van cruciaal belang is voor celpluripotentie, proliferatie en differentiatie in verschillende contexten 3,6,11,12,13,14,15,16,17.
Osteoblasten zijn secretoire cellen die de collageen type 1 rijke extracellulaire botmatrix produceren en afscheiden. Om hoge snelheden van eiwitsynthese tijdens botvorming te ondersteunen, vragen osteoblasten om een constante toevoer van aminozuren. Om aan deze vraag te voldoen, moeten osteoblasten actief aminozuren verwerven. In overeenstemming hiermee onthullen recente studies het belang van aminozuuropname en metabolisme in osteoblastactiviteit en botvorming 15,16,17,18,19,20.
Osteoblasten verwerven cellulaire aminozuren uit drie belangrijke bronnen: extracellulair milieu, intracellulaire eiwitafbraak en de novo aminozuurbiosynthese. Dit protocol zal zich richten op de evaluatie van aminozuuropname uit extracellulaire omgeving. De meest gebruikelijke methoden om de opname van aminozuren te meten, zijn afhankelijk van radioactief gelabelde (bijv. 3H of 14C) of zware isotoop gelabelde (bijv. 13C) aminozuren. Zware isotopomesten kunnen de opname en het metabolisme van aminozuren grondiger en veiliger analyseren, maar zijn tijdrovender en het duurt meerdere dagen om te voltooien, omdat het een dag duurt om monsters voor te bereiden en te derivatiseren en meerdere dagen om te analyseren op de massaspectrometer, afhankelijk van het aantal monsters21,22. Ter vergelijking: radioactief gelabelde aminozuuropnametests zijn niet informatief over het downstream-metabolisme, maar zijn goedkoop en relatief snel, omdat ze binnen 2-3 uur na het begin van het experiment kunnen worden voltooid 23,24. Hier beschrijven we een gemakkelijk te wijzigen basisprotocol dat is ontworpen om de opname van radioactief gelabelde aminozuren in gekweekte primaire cellen of cellijnen in vitro of individuele botschachten ex vivo te evalueren. De toepassing van deze twee protocollen kan worden uitgebreid naar andere radioactief gelabelde aminozuren en andere bot geassocieerde celtypen en weefsels.

Botontwikkeling en homeostase is afhankelijk van de differentiatie en activiteit van botvormende osteoblasten. Osteoblastdifferentiatie wordt sequentieel gekenmerkt door proliferatie gevolgd door eiwitsynthese en uiteindelijk botmatrixsecretie. Proliferatie en eiwitsynthese vereisen een constante toevoer van aminozuren. Desondanks is er zeer weinig bekend over aminozuurconsumptie bij osteoblasten. Hier beschrijven we een zeer gevoelig protocol dat is ontworpen om aminozuurconsumptie te meten met behulp van radioactief gelabelde aminozuren. Deze methode is geoptimaliseerd om veranderingen in aminozuuropname te kwantificeren die geassocieerd zijn met osteoblastproliferatie of -differentiatie, medicijn- of groeifactorbehandelingen of verschillende genetische manipulaties. Belangrijk is dat deze methode door elkaar kan worden gebruikt om aminozuurconsumptie in gekweekte cellijnen of primaire cellen in vitro of in geïsoleerde botschachten ex vivo te kwantificeren. Ten slotte kan onze methode eenvoudig worden aangepast om het transport van een van de aminozuren te meten, evenals glucose en andere radioactief gelabelde voedingsstoffen.

Aminozuurtransport wordt gereguleerd door veel membraangebonden aminozuurtransporters die zijn gecategoriseerd in verschillende transportsystemen op basis van tal van kenmerken, waaronder substraatspecificiteit, kinetiek, evenals ion- en pH-afhankelijkheid25. Aminozuuropname en metabolisme zijn essentieel voor osteoblastspecificatie, differentiatie en botvorming. Dit protocol biedt een snelle en gevoelige methode om de opname van aminozuren te evalueren. Ons protocol maakt gebruik van radioactief gelabelde aminozuren om de opname van aminozuren te kwantificeren.Het is een goedkope, gevoelige en snelle methode die gemakkelijk kan worden aangepast aan verschillende omstandigheden. Dit protocol kan eenvoudig worden gewijzigd om de opname van andere radioactief gelabelde voedingsstoffen zoals glucose en vetzuren in verschillende weefsels en primaire of transformatiecellen te evalueren. Om te beginnen met aminozuuropname assay plaat vijf keer 10 tot de vierde ST2 cellen in elke put van een 12 put weefsel kweekplaat in alfa MEM medium aangevuld met FBS, penicilline en streptomycine.Plaat extra putten van cellen om het celgetal per voorwaarde voor de normalisaties te kwantificeren. Incubeer vervolgens cellen in een bevochtigde celkweekincubator bij 37 graden Celsius met 5% koolstofdioxide gedurende twee tot drie dagen totdat ze samenvloeien. Na incubatie aspirateer het medium en was de cellen tweemaal met één milliliter PBS bij pH 7,4.Aspirateer vervolgens de PBS voordat u de cellen eenmaal wast met een milliliter Krebs-Ringer's HEPES of KRH. Incubeer cellen met 0,5 milliliter vers bereide KRH met vier microcuries per milliliter L34 getriatificeerde glutamine werkmedia gedurende vijf minuten. Verzamel na incubatie het radioactieve medium om in de container met vloeibaar afval te doseren.Was de cellen vervolgens drie keer met ijskoude KRH om de reactie te beëindigen. Breng vervolgens de cellysaten over in een buis van 1,5 milliliter en verhelder het lysaat door gedurende 10 minuten met een minimumsnelheid van 10.000 G te centrifugeren.Breng het supernatant over naar een scintillatieflacon met acht milliliter van de scintillatieoplossing en lees de radioactiviteit en tellingen per minuut met behulp van de scintillatieteller. Van de resterende niet-radioactieve platen van cellen, trypsinize, resuspend, en tel de cellen om het aantal cellen in de gelyseerde radioactieve culturen te schatten. Tel met behulp van een hemacytometer het aantal cellen per niet-radioactieve put voor elke experimentele toestand.Om de opname van aminozuren te bepalen, spoelt u het merg uit het bot voordat u de botschacht weegt. Om het bot te decellulariseren, kookt u één opperarmbeen in PBS op 100 graden Celsius gedurende 10 minuten. Balanceer zowel levende als gekookte humeri in één milliliter KRH gedurende 30 minuten in de celkweekincubator bij 37 graden Celsius.Incubeer vervolgens de experimentele en gekookte controlebotten afzonderlijk in vers bereide KRH met vier microcurries per milliliter L234 getriageerd arginine gedurende maximaal 90 minuten bij 37 graden Celsius. Na het verwijderen van het radioactieve medium, beëindigt u de reactie door het opperarmbeen drie keer te wassen met één milliliter ijskoude KRH. Breng elk bot over in een buis van 1,5 milliliter met 500 microliter RIPA Buffer.Homogeniseer vervolgens het opperarmbeen en de RIPA-buffer door 100 keer met een schaar te hakken, gevolgd door ultrasoonapparaat bij 35% amplitude en één tweede puls gedurende 10 seconden. Het lysaat geklaard door centrifugeren gedurende 10 minuten bij 10.000 maal G bij kamertemperatuur. Breng na centrifugatie 200 microliter van het supernatant over naar scintillatieflacons met acht milliliter van de scintillatieoplossing.Lees radioactiviteit met behulp van de scintillatieteller. Kinetische eigenschappen van in vitro L34 tritamine opname en confluente ST2 cellen werden geëvalueerd in de aan- of afwezigheid van natriummethylamino-isoboterzuur of lithium. De verwijdering van natrium leidde tot een vermindering van 90% van de glutamine-opname.De aanwezigheid van lithium verhoogde de opname van glutamine met 2% in vergelijking met de natriumvrije toestand. Terwijl methylamino-isoboterzuur geen invloed had op de opname van glutamine. De geschatte procentuele bijdragen van systeem A, N, L of ASC en gamma plus L gaven aan dat de meeste glutamine-opname wordt gemedieerd door systemen ASC en gamma plus L.Systeem N is verantwoordelijk voor ongeveer 2% natriumafhankelijke glutamine-opname.En systeem A toonde 0% bijdrage en opname. X-VIVO L34 tritiele arginine opname in gekookte en levende neonatale humeri werd geanalyseerd over een tijdskuur van twee uur. Arginine opname nam lineair toe gedurende het experiment en levende humeri.De gekookte humeri vertoonde geen dynamische toename van arginineopname in het experiment. De representatieve analyse toont de genormaliseerde en gecorrigeerde arginine opname. Het is belangrijk om het contralaterale bot te koken als een actieve controle.Deze stap is essentieel omdat de botmatrix radioactieve aminozuren kan absorberen, wat resulteert in onnauwkeurige resultaten. Het protocol heeft ons in staat gesteld om de kinetiek van aminozuuropname te bevestigen, zowel in vitro als in vivo.
