アミノ酸の取り込みと代謝は、骨芽細胞の仕様、分化、および骨形成に不可欠です。このプロトコルは、アミノ酸の取り込みを評価するための迅速かつ高感度な方法を提供します。私たちのプロトコルは、放射性標識されたアミノ酸を使用してアミノ酸の取り込みを定量化します。これは、さまざまな条件に合わせて簡単に変更できる、安価で敏感で高速な方法です。このプロトコルは、初代細胞や形質転換細胞だけでなく、さまざまな組織におけるグルコースや脂肪酸などの他の放射性標識栄養素の取り込みを評価するために簡単に変更できます。FBS、ペニシリンおよびストレプトマイシンを添加したαMEM培地中の12穴組織培養プレートの各ウェルで10〜4番目のST2細胞のアミノ酸取り込みアッセイプレートを5回開始する。細胞の余分なウェルをプレートして、正規化の条件ごとの細胞数を定量化します。次に、加湿した細胞培養インキュベーターで摂氏37度、5%二酸化炭素で細胞をコンフルエントになるまで2〜3日間インキュベートします。インキュベーション後、培地を吸引し、pH 7.4で1ミリリットルのPBSで細胞を2回洗浄します。次に、PBSを吸引してから、1ミリリットルのクレブスリンガーHEPESまたはKRHで細胞を1回洗浄します。1ミリリットルあたり4マイクロキュリーを含む0.5ミリリットルの新たに調製したKRHで細胞をインキュベートします L34 トリチウム化グルタミン作動培地 5分間。インキュベーション後、放射性媒体を収集して液体廃棄物容器に分注します。次に、細胞を氷冷KRHで3回洗浄して反応を終了させます。次に、1ミリリットルの1%ドデシル硫酸ナトリウムを各ウェルに加え、混合物を10回粉砕して細胞を溶解および均質化します。次に、細胞ライセートを1.5ミリリットルのチューブに移し、最低速度10, 000 Gで10分間遠心分離してライセートを清澄化します。上清を8ミリリットルのシンチレーション溶液を含むシンチレーションバイアルに移し、シンチレーションカウンターを用いてラジオアクティビティと毎分カウントを読み取る。細胞の残りの非放射性プレートから、細胞をトリプシン処理、再懸濁、およびカウントして、溶解した放射性培養中の細胞数を推定します。血球計算盤を用いて、実験条件ごとに非放射性ウェル当たりの細胞数をカウントする。アミノ酸の取り込みを決定するには、骨シャフトの重量を量る前に骨髄を骨から洗い流します。骨を脱細胞化するには、上腕骨をPBSで摂氏100度で10分間煮沸します。生上腕骨と煮上腕骨の両方を、摂氏37度の細胞培養インキュベーターで30分間、1ミリリットルのKRHで平衡化します。次に、実験用および煮沸した対照骨を、1ミリリットルのL234トリシエーションアルギニンあたり4つのマイクロカレーを含む新たに調製したKRHで、摂氏37度で最大90分間インキュベートします。放射性媒体を除去した後、上腕骨を1ミリリットルの氷冷KRHで3回洗浄して反応を終了する。各骨を500マイクロリットルのRIPAバッファーを入れた1.5ミリリットルのチューブに移します。次に、ハサミで100回刻んで上腕骨とRIPAバッファーを均質化し、続いて35%振幅で超音波処理を行い、1秒間1秒間パルスします。室温で10, 000倍Gで10分間遠心分離することにより溶解液を清澄化した。遠心分離後、200マイクロリットルの上清を8ミリリットルのシンチレーション溶液を含むシンチレーションバイアルに移す。シンチレーションカウンタを使用してラジオアクティビティを読み取ります。in vitro L34 トリチウム化グルタミン取り込みおよびコンフルエントST2細胞の動態特性を、アミノイソ酪酸メチルナトリウムまたはリチウムの存在下または非存在下で評価した。ナトリウムの除去は、グルタミンの取り込みの90%の減少につながりました。リチウムの存在は、ナトリウムを含まない状態と比較して、グルタミンの取り込みを2%増加させた。一方、アミノイソ酪酸メチルはグルタミンの取り込みに影響を与えなかった。システムA、N、LまたはASCとガンマプラスLの推定寄与率は、ほとんどのグルタミン取り込みがシステムASCとガンマプラスLによって媒介されることを示しました。そして、システムAは0%の寄与と取り込みを示しました。X-VIVO L34トリチウム化アルギニンの煮沸および生きた新生児上腕骨における取り込みを2時間の経時変化にわたって分析した。アルギニンの取り込みは、実験全体を通して直線的に増加し、上腕骨を生きた。茹でた上腕骨は、実験においてアルギニン取り込みの動的増加を示さなかった。代表的な分析は、正規化および補正されたアルギニン取り込みを示す。アクティブコントロールとして反対側の骨を沸騰させることが重要です。骨基質が放射性アミノ酸を吸収し、不正確な結果をもたらす可能性があるため、このステップは不可欠です。このプロトコルにより、in vitroとin vivoの両方でアミノ酸取り込み動態を確認することができました。
