このプロトコルは、アカゲザルのアルファベータおよびタウレベルに対するrTMSの影響を探索するために、連続槽マグナCSFサンプリング法を使用する。この技術は、有害事象のリスクが低い完全覚醒条件下でCSFサンプリングを繰り返すことを可能にする新しいCSFサンプリング方法を使用する。この手順を実証するのは、南西医科大学の生理学部門の研究助手であるYing-Qian Zhangと、四川大学の大学院生であるHui-Xin Tanです。鎮痛剤を投与し、麻酔が成功したことを確認した後、5歳の雄アカゲザルを側方褥瘡位置の手術台に置き、腰の周りの領域を消毒する。背中をひっくり返し、膝を胸の方に持っていきます。腰椎L4〜L5の間に脊髄針を挿入し、靭帯フラバが収容されている腰椎槽への進入を示すポップがあるまで押し込みます。硬膜への進入を示す2番目のポップがあるまで針をさらに押し続け、次に脊髄針からスタイレットを引き出し、CSFの滴を集める。次に、硬膜外カテーテルを穿刺針を介してくも膜下腔内に挿入し、大槽に浮力がつくまで透視誘導下でカテーテル挿入を行う。次にポート移植のために、穿刺部位から頭部に向かって5センチメートルの切開を行い、皮下組織から皮膚を単離してサンプリングポートを配置する。ポートを硬膜外カテーテルの端部に接続し、ポートを皮膚の下に埋め込み、切開部を縫合する。CSF収集の場合は、ケージバーを使用してサルを拘束し、背中を曲げたままにします。その後、アルコール綿棒で拭き取り、シリンジをサンプリングポートの中央に挿入して、カテーテルを通して胸郭マグナからCSFを抽出する。最初の 200 マイクロリットルの CSF を廃棄し、分析用に 1 ミリリットルの CSF を収集します。rTMS介入中の中断を避けるために実験前にサルをサルの椅子に固定し、麻酔薬の影響を避けるためにサルが目覚めているときにバイオマーカー分析のためにCSFを収集します。くも膜下カテーテル法の3日目及び実験開始の2週間前に、サルをサルチェアによる適応訓練に1日2回、毎回30分間供した。サルチェアによる適応トレーニングの1週間後、正式な実験開始の1週間前にrTMS適応トレーニングまたは偽刺激を行い、刺激プロセス中の振動や音によって実験の進行を妨げないようにします。偽刺激には、振動と音を発生させるが磁場を発生させない偽コイルを使用してください。刺激を受けた後、サルがプロセスに適応するのを助けるためにサルに食べ物を提供します。国際10~20系に従って両側背外側前頭前野を局在化し、24時間を超える休止期間を設けずに3つの異なる周波数を用いてrTMSの3つの異なるセッションを行う。第1,第2,第3のセッションを1日2回,3日間連続して行う。4つのCSFバイオマーカーを分析するには、低侵襲カテーテル法を使用してCSFを採取する。5 つの時点で CSF を収集し、各時点で 4 つのサンプルを 3 分間隔で収集します。3つの周波数で合計60個のサンプルを採取した後、それらを番号付けしてマイナス80°Cの冷蔵庫に最大1ヶ月間保管します。実験後、製造元の指示に従って、すべてのサンプルを液体チップ検出に供する。1ヘルツrTMS刺激後、Abeta42レベルは24時間にわたって徐々に増加した。20ヘルツのrTMSと同様に、Abeta42レベルは時間とともに増加し、刺激後6時間でピークに達した。対照的に、40ヘルツ刺激後、Abeta42レベルは、rTMS後の時点で直ちに有意に増加し、その後、ゆっくりと減少した。Abeta40に対するAbeta42の比率は、1および20ヘルツのrTMSによる刺激後に上昇傾向を示し、刺激後2時間後に有意に増加した。しかし、40ヘルツでの比率に有意差はなかった。CSF中のtTauレベルは、rTMS刺激20および40ヘルツ後に直ちに増加し、徐々に減少した。しかし、1ヘルツ刺激後に有意差はなかった。pTauレベルは、40ヘルツ刺激後に直ちに増加し、24時間後にベースラインレベル以下に低下した。これに対し、1及び20ヘルツ刺激については、pTauレベルは低下傾向を示した。サルCSFの複数のサンプルを必要とする研究者は、この連続槽マグナCSFサンプリング法を検討することができます。
