根本的な問題であるアルボウイルスのベクター宿主感染様式は、生物科学の最前線にあります。農業上重要な多くの植物ウイルスは、昆虫媒介動物によって水平に伝染します1。植物ウイルスの半分以上は、アブラムシ、コナジラミ、ヨコバイ、ウンカ、アザミウマなどの半翅目昆虫によって媒介されています。これらの昆虫は、植物ウイルスを効率的に感染させることを可能にする明確な特徴を持っています1。彼らは穿孔吸引口器を持ち、師部と木部からの樹液を食べ、唾液を分泌します1,2,3,4。技術の開発と改善に伴い、唾液成分の同定と機能分析は、集中的な研究の新たな焦点になりつつあります。唾液中の既知の唾液タンパク質には、ペクチネステラーゼ、セルラーゼ、ペルオキシダーゼ、アルカリホスファターゼ、ポリフェノールオキシダーゼ、スクラーゼなどの多数の酵素が含まれます5,6,7,8,9,10,11,12,13 .唾液中のタンパク質には、宿主防御応答を誘発し、それによって昆虫の性能を変化させるエリシター、および昆虫の適応度を高める宿主防御を抑制するエフェクターおよび宿主病理学的応答を誘導する成分も含まれる14,15,16,17。したがって、唾液タンパク質は昆虫と宿主の間のコミュニケーションに不可欠な物質です。ウイルスの伝染中、穿刺吸引ウイルス性の昆虫の唾液腺から分泌される唾液にもウイルスタンパク質が含まれています。ウイルス成分は唾液の流れを利用して昆虫から植物宿主に放出します。したがって、昆虫の唾液は、ウイルス-ベクター-宿主の三栄養相互作用を橋渡しします。ウイルス性昆虫が分泌する唾液タンパク質の生物学的機能を調査することは、ウイルス-ベクター-宿主の関係を理解するのに役立ちます。
動物ウイルスの場合、蚊の唾液がウエストナイルウイルス(WNV)とデングウイルス(DENV)の伝染と病原性を媒介することが報告されています。唾液タンパク質AaSG34はデング熱ウイルスの複製と伝播を促進し、唾液タンパク質AaVA-1はオートファジーを活性化することによりDENVおよびジカウイルス(ZIKV)の伝播を促進します18,19。蚊の唾液タンパク質D7は、DENVビリオンおよび組換えDENVエンベロープタンパク質20との直接相互作用を介して、インビトロおよびインビボでDENV感染を阻害することができる。植物ウイルスでは、ベゴモウイルストマト黄葉カールウイルス(TYLCV)は、植物宿主のWRKY33を介した免疫を抑制するコナジラミ唾液タンパク質Bsp9を誘導して、コナジラミの嗜好と性能を高め、最終的にウイルスの感染を増加させます21。昆虫の唾液タンパク質が植物宿主で果たす役割の研究は動物宿主の研究に遅れをとっており、植物宿主中の唾液タンパク質を検出するための安定した信頼性の高いシステムが緊急に必要とされています。
イネ矮性ウイルス(RDV)として知られる植物ウイルスは、ヨコバイネフォテティクス・シンクティセプス(半翅目:Cicadellidae)によって高効率で持続的に増殖して伝染します22,23。RDVは昆虫媒介動物によって伝染し、アジアでイネの重篤な病気を引き起こすことが最初に報告されました24,25。ビリオンは二十面体で二重層の球状であり、外層はP8外側キャプシドタンパク質22を含む。N. cincticepsにおけるRDVの循環伝達期間は14日26,27,28,29,30である。RDVが唾液腺に到達すると、ビリオンはエキソサイトーシス様機構を介して唾液腺の唾液貯蔵空洞に放出される23。ビテロゲニン(Vg)は、雌昆虫の卵母細胞発生に必須の卵黄タンパク質前駆体です31、32、33。ほとんどの昆虫種は、約220kDaの前駆タンパク質をコードする6〜7kbの少なくとも1つのVg転写産物を有する。Vgのタンパク質前駆体は、通常、卵巣に入る前に、大きな断片(140〜190 kDa)と小さな断片(<50 kDa)に切断することができます18,19。これまでのプロテオミクス解析では、ヨコバイの分泌唾液中にVg由来のペプチドが存在することが明らかになったが、その機能は不明である(未発表データ)。ウンカから経口分泌されるVgが、植物の防御を損傷するエフェクターとして機能することが新たに報告されている34。N. cincticepsのVgも唾液流で植物宿主に放出され、植物防御を妨害する役割を果たすことができるかどうかは不明です。N. cincticepsがVgなどの唾液タンパク質を利用して植物の防御を阻害または活性化するかどうかに対処するための最初のステップは、摂食中に植物に放出されるタンパク質を特定することです。植物中に存在する唾液タンパク質の同定方法を理解することは、唾液タンパク質の機能や半翅目と植物の相互作用を説明するために不可欠である可能性があります。
