stereotaxic viral injection and gradient-index lens implantation for deep brain in vivo calcium imaging

생체 내 칼슘 이미징의 결과는 바이러스 주입과 GRIN 렌즈 이식 수술의 성공에 달려 있습니다. 도 5는 생체내 칼슘 이미징 기록으로부터의 다양한 결과(즉, 성공적이지 못함, 최적이 아닌 및 양호함)를 보여준다. 실패한 경우, 칼슘 이미지는 어둡거나 밝게 보일 수 있지만 일반적으로 활성 뉴런이 없거나 거의 나타나지 않습니다. 우리는 일반적으로 다섯 개 미만의 활성 뉴런이있는 경우 생체 내 칼슘 기록 실험을 추구하지 않습니다. 좋은 생체 내 칼슘 이미징은 일반적으로 수백 개의 활성 뉴런을 나타냅니다. 기록에 백 개 미만의 활성 뉴런이 포함되어 있다면, 우리는 그것을 차선책으로 간주합니다.
차선책과 양호한 기록 에서 생체 내 칼슘 이미징 실험이 추구되었고, 후속 데이터 분석이 수행되었다. 무비 1은 마우스 mPFC로부터의 대표적인 생체내 칼슘 이미징 기록을 나타낸다. 행동 비디오 및 칼슘 이미징 데이터는 일반적으로 별도로 처리됩니다. 마우스 동작 비디오는 수동으로 점수를 매길 수 있습니다. 칼슘 이미징 파일은 CaIman 칼슘 이미지 프로세싱 툴박스(24)를 사용하여 처리된다. 도 6은 양호한 생체내 칼슘 영상 기록으로부터의 대표적인 세포 맵 및 몇몇 칼슘 트레이스를 도시한다.
