流域变换通常与分割技术相结合，以准备图像以进行更清晰的分析，从而提高细胞计数的准确性。对于此过程，分水岭变换用于在分割之前突出显示潜在的感兴趣区域。分水岭变换通过识别图像前景中的细胞提供了独特的优势，这可以通过消除细胞的潜在误报（例如背景的不均匀斑块）来提高分割分析的准确性。然而，当试图使基于细胞的图像适应分水岭变换时，可能会出现困难。具有高细胞密度的图像可能会受到下分段的困扰，其中细胞的聚集体被识别为单一组而不是单个组分。噪声或剧烈强度变化的存在也可能导致过度分段，其中算法过度隔离细胞，导致细胞计数过多和不准确8。
在本文中，我们详细介绍了一种方法，通过将阈值分析的组件合并到基于面积的量化算法中来最小化分水岭变换的主要缺点，如图 1所示。值得注意的是，该算法是使用开源和/或广泛使用的软件实现的，并且无需昂贵的试剂或复杂的细胞制备技术即可应用该细胞计数框架。RAW264.7巨噬细胞用于证明该方法，因为它们在调节结缔组织维持和伤口愈合过程中起着关键作用9。此外，由于NIH / 3T3成纤维细胞在组织维护和修复中的关键作用，对其进行了分析。成纤维细胞通常与巨噬细胞共存并支持巨噬细胞，因此需要在共培养研究中区分这些表型不同的细胞类型。
通过计算细胞覆盖的面积和单一细胞占用的平均面积，可以可靠有效地量化具有高活细胞密度（VCD）的图像中的细胞计数。使用阈值而不是分割进行细胞鉴定也使更复杂的分析成为可能，例如同时分析共培养中不同细胞类型的实验。NIH / 3T3成纤维细胞，通常被发现与伤口愈合部位内的RAW264.7巨噬细胞共定位，被发现在与巨噬细胞10的焦平面不同的焦平面上生长。因此，根据所分析的细胞类型，运行多种阈值算法来定义背景和前景，从而能够准确计数同一图像中的两种不同细胞类型。
