嚢胞性線維症患者に対する個別化治療は、ベースラインにおけるCFTR活性を理解するためのインビトロ疾患モデルによって達成することができる。ヒト鼻上皮オルガノイド、またはHNEオルガノイドは、CFTR活性と相関する様々なサイズのルーメンを産生し、CFオルガノイドと非CFオルガノイドを区別する。ここでは、これらのHNEオルガノイドを培養・イメージングするための方法論を詳細に説明した。機能不全の上皮イオン輸送、特に塩化物および重炭酸塩の輸送は、上皮ライニング流体の体積の減少をもたらす。これはまた、粘液分泌物に影響を及ぼし、粘膜炎および閉塞につながる。したがって、私たちのHNEモデルは、研究者の実験計画とリソースに応じて、さまざまなアプリケーション向けに開発されています。ベースライン時または治療薬に応答してCFTR活性を評価することとは別に、この技術は、上皮細胞機能、特に上皮細胞液輸送を伴う他の疾患にも適用することができる。これらの方法は、主に嚢胞性線維症研究における使用のために開発された。原発性毛様体ジスキネジアなどの気道上皮を評価する他の研究も、これらの方法が有用であると認める可能性がある。これらの手法には、細部と精度に注意が必要です。また、細胞の初期膨張が適切であることを確認するために、注意深い観察も必要です。毎日すべての文化を監視すると、成功の可能性が高まります。開始するには、細胞診ブラシを1ミリリットルの大口径ピペットチップに数回通すことによって、鼻ブラシ生検を15ミリリットルの円錐形チューブ内の8ミリリットルのRPMI培地に解離させる。細胞を500倍gで摂氏4度で5分間遠心分離する。上清を捨て、次いで細胞ペレットを3ミリリットルの細胞剥離溶液に再懸濁する。室温で16分間インキュベートし、消化する。5ミリリットルの膨張培地を使用して、細胞を2回洗浄する。次いで、それらをT75フラスコに加え、照射、不活化、および50〜60%コンフルエントな3T3線維芽細胞を予め播種する。細胞を共培養し、拡張培地中で7〜14日間膨張させる。嚢胞性線維症患者由来の細胞に抗生物質を導入した場合、培養の最初の3日後に培地を抗生物質を含まない拡張培地に交換し、80%コンフルエントになるまで2日ごとに培地を交換し続けることができます。1x DPBSで細胞を洗浄します。次いで、1.5ミリリットルの0.05%トリプシン-EDTAをT75フラスコに加え、摂氏37度で4分間インキュベートして、照射および不活化された3T3線維芽細胞を培養物から除去した。T75フラスコを1x DPBSで2回すすぎ、残りの3T3線維芽細胞を完全に洗い流します。1.5ミリリットルの0.05%トリプシン-EDTAをT75フラスコに加え、摂氏37度で10分間インキュベートしてHNEを剥離する。ダイズトリプシン阻害剤でトリプシンを1対1の比率で中和する。細胞を5分間500回giで遠心分離する。その後、上清を捨て、5ミリリットルの膨張培地で細胞を1回洗浄する。細胞はオルガノイドを成長させるために播種する準備ができています。オルガノイド培養細胞外マトリックスまたはECMを摂氏4度で一晩解凍する。15ウェル血管新生スライドを同じ温度で一晩冷却する。次に、ピペットチップを摂氏4度に冷却します。氷の上に5マイクロリットルの冷たい100%ECMでスライドをコーティングします。それらを摂氏37度の細胞培養インキュベーターに入れ、少なくとも30分間固結させる。共培養から採取したHNEsを血球計数器を用いて計数する。次いで、細胞を総数当たり500個の細胞に希釈し、氷上の分化培地により希釈した20%ECMを用いた。ECMコーティングされたスライドをインキュベーターから取り出し、5マイクロリットルの冷細胞ECM混合物をそれぞれのウェルに播種する。スライドを直ちに摂氏37度の培養インキュベーターに1時間移し、細胞ECM混合物を固結させた。その後、インキュベーターからスライドを取り出し、50マイクロリットルの分化培地を用いて15ウェル血管新生スライドの各ウェルに細胞を送り込む。スライドを摂氏37度の培養インキュベーターに戻し、オルガノイドがさらなる実験の準備が整うまで1日おきに培地を交換します。8ウェルガラス底チャンバースライドを細胞接着剤で30分間前処理する。溶液を廃棄し、井戸をさらに30分間風乾する。次に、ECMの上部からメディアを廃棄し、氷上の15ウェルスライドの各ウェルに50マイクロリットルの冷たい1x PBSを追加します。200マイクロリットルの大口径ピペットチップを使用してピペットを3〜5回上下させます。8ウェルチャンバースライドのウェルの中央に溶液を分配します。余分な液体をすぐに細かいチップピペットでウェルから取り除きます。次いで、チャンバースライドを37°Cのインキュベーターに40分間置き、ガラス底に付着したオルガノイドを増強する。40分後、オルガノイドを1x PBSで穏やかに2回洗浄し、各ウェル中の300マイクロリットルの4%パラホルムアルデヒドで室温に設定して30分間固定する。1x PBSで2回洗浄し、オルガノイドを1x PBSセットの摂氏4度に保存し、免疫染色を最大2週間行う。組織学的研究のためにオルガノイドを採取するには、培養物から培地を除去し、氷上のスライドの各ウェルに50マイクロリットルの冷たい1つの1x PBSを加える。200マイクロリットルの大口径ピペットチップを使用してピペットを3〜5回上下させます。15ウェルスライドまたは培養インサートからのすべての溶液を氷上の15ミリリットルの円錐形チューブに組み合わせます。冷たい1x PBSを加えて、チューブ内の総溶液量を10ミリリットルに調整する。チューブを摂氏4度で5分間300回g遠心分離する。次いで、上清を吸引し、60マイクロリットルの温かいヒストゲルを加えて、200マイクロリットルの大口径ピペットチップを用いてオルガノイドペレットと混合する。懸濁液を直ちに組織学鋳型に移す。ヒストゲルを室温で固結させた後、モールドブロックを4%パラホルムアルデヒドに入れ、摂氏4度で一晩固定する。ソフトウェアを開き、画面の下部を右クリックします。次に、[分析コントロール]を選択し、[自動測定結果]を選択します。オルガノイド画像を開き、ルーメンが見える5~10個のオルガノイドを選択します。画像を右クリックしたままメニューを開き、[多角形ROI]を選択してオルガノイド全体の輪郭を描き、オルガノイドの総表面積を取得します。次に、ルーメンを輪郭を描き、ルーメン領域を求める。ウェル内の残りのオルガノイド、およびアッセイ中のすべてのウェルについてこれを繰り返す。最後に、データを Excel にエクスポートします。ルーメン面積を総表面積で割り、サンプルからのすべてのオルガノイドを平均してベースラインルーメン比を求めます。明視野画像は、成功したサンプル コレクションと失敗したサンプル コレクションからの HNE を表します。HNEは大きなクラスターでよく成長します。対照的に、HNEは、照射された3T3細胞を囲む2つの小さなクラスターではあまり成長しない。15ウェルスライドのオルガノイドは、培養インサート内のものよりも正確で鮮明な画像を有する。これ以外にも、これら2つの培養方法において有意な形態学的差異は認められなかった。非CFオルガノイドは、典型的には、CFオルガノイドよりも大きな内腔およびより多くの流体を有する。非CF、F508del/F508del被験者からのオルガノイドにおけるHNE染色、およびオルガノイドにおける繊毛の免疫蛍光染色をこれらの画像に示す。アセチル化チューブリンで染色された繊毛、FITC標識二次抗体、およびDAPIで標識された核がここに示されている。ホールマウント免疫蛍光染色による2つの代表的なオルガノイドの最大投影像およびそれらに対応する三次元再構成像がここに示されている。オルガノイドの内腔内の粘液および繊毛が示されている。グラフィカル画像は、初代鼻上皮細胞上のCFTR機能を試験するためのフォルスコリン誘導腫脹アッセイを表す。非CFボランティアの代表的なフォルスコリン用量反応実験がここに示されている。FSK用量反応は、1時間時と8時間時の平均分数変化とを比較し、これは、8時間のアッセイが1時間のアッセイよりも異なるFSK用量間でより有意な腫脹差を生じ得ることを示唆している。曲線の下の面積は、8時間時の平均分数変化と比較して腫脹のわずかな差を示すことができる。この手順を試みている間、オルガノイドは収集、固定および染色プロセス中に失われることに留意してください。したがって、各ステップでは細心の注意を払い、成功を確実にするために十分な開始番号を取得する必要があります。培養インサート中のオルガノイドの成長は、これらの技術が開発されるにつれて、この点で助けることができる。ここでは、市販の試薬と供給品を使用して、他の研究者への拡大を容易にしました。一般的な顕微鏡技術とより特殊な装置を使用した機能アッセイも開発されました。
