Alle dierproeven werden goedgekeurd door de dierenwelzijnscommissie van de Shanghai University of Traditional Chinese Medicine. Vier weken oude mannelijke BALB / c athymische muizen werden geacclimatiseerd gedurende een week vóór de operatie voor orthotopische injectie van osteosarcoomcellen. Muizen werden gehuisvest in individueel geventileerde muizenkooien met vijf muizen per kooi in een licht/ donkercyclus van 12 uur met ad libitum toegang tot SPF-voer en steriel water.

Orthotopische injectie van osteosarcoomcellen is een ideaal model om de functie en het mechanisme van specifieke factoren in osteosarcoomcarcinogenese en ontwikkeling te bestuderen om de therapeutische werkzaamheid te evalueren. Om verschillen in tumorgroei te voorkomen, worden de meeste actieve osteosarcoomcellen bij 80% -90% confluent met hetzelfde aantal zorgvuldig geïnjecteerd in het scheenbeen van elke muis en wordt de cel trypsinisatietijd strikt gecontroleerd zonder de levensvatbaarheid van de cel te beïnvloeden...

Osteosarcoom is de meest voorkomende primaire botkanker bij kinderen en adolescenten 1,2, die voornamelijk het omliggende weefsel infiltreert en zelfs uitzaait naar de longen wanneer de patiënten worden gediagnosticeerd. Pulmonale metastase is de belangrijkste uitdaging voor osteosarcoomtherapie en de vijfjaarsoverleving van osteosarcoompatiënten met pulmonale metastase blijft zo laag als 20% -30% 3,4,5. De vijfjaarsoverleving van primair osteosarcoom is echter sinds de jaren 1970 verhoogd tot ongeveer 70% als gevolg van de introductie van chemotherapie6. Daarom is het dringend nodig om het fundamentele mechanisme van osteosarcoomcarcinogenese en pulmonale metastase te begrijpen om nieuwe therapieën te ontwikkelen. De toepassing van muismodellen die de progressie van osteosarcoom bij mensen het beste nabootsen, is van groot belang7.
De osteosarcoom diermodellen worden gegenereerd door spontane, geïnduceerde genetische manipulatie, transplantatie en andere technieken. Het spontane osteosarcoommodel wordt zelden gebruikt vanwege de lange tumorvormingstijd, inconsistente tumorvoorval, lage morbiditeit en slechte stabiliteit 8,9. Hoewel het geïnduceerde osteosarcoommodel toegankelijker is om te verkrijgen dan het spontane osteosarcoom, is de toepassing van het geïnduceerde osteosarcoommodel beperkt omdat de inducerende factor de micro-omgeving, de pathogenese en pathologische kenmerken van osteosarcoom zal beïnvloeden10. Transgene modellen helpen de pathogenese van kankers te begrijpen, omdat ze de menselijke fysiologische en pathologische omgevingen beter kunnen simuleren; de transgene diermodellen hebben echter ook hun beperkingen vanwege de moeilijkheid, lange termijn en hoge kosten van transgene modificatie. Bovendien, zelfs in de meest algemeen aanvaarde transgene diermodellen gegenereerd door p53- en Rb-genmodificatie, kwam slechts 13,6% van het sarcoom voor in de botten van de vier ledematen11,12.
Transplantatie is een van de meest gebruikte primaire en verre gemetastaseerde kankermodelproductiemethoden in de afgelopen jaren vanwege de eenvoudige manoeuvre, stabiele tumorvormingssnelheid en betere homogeniteit13. Transplantatie omvat heterotope transplantatie en orthotopische transplantatie volgens de transplantatieplaatsen. Bij osteosarcoom heterotope transplantatie worden de osteosarcoomcellen geïnjecteerd buiten de primaire osteosarcoomplaatsen (bot) van de dieren, meestal onder de huid, subcutaan14. Hoewel de heterotope transplantatie eenvoudig is zonder de noodzaak om een operatie bij dieren uit te voeren, vertegenwoordigen de plaatsen waar de osteosarcoomcellen worden geïnjecteerd niet de werkelijke menselijke osteosarcoommicro-omgeving. Osteosarcoom orthotopische transplantatie is wanneer de osteosarcoomcellen worden geïnjecteerd in de botten van dieren, zoals tibia15,16. In vergelijking met de heterotope grafts worden orthotopische osteosarcoomtransplantaten gekenmerkt door een korte incubatietijd, snelle groei en een sterke erosieve aard; daarom zijn het ideale diermodellen voor osteosarcoom-gerelateerde studies17.
De meest gebruikte dieren zijn muizen, honden en zebravissen18,19. Het spontane model van osteosarcoom wordt meestal gebruikt bij honden omdat osteosarcoom een van de meest voorkomende tumoren bij honden is. De toepassing van dit model is echter beperkt vanwege de lange tumorvormingstijd, de lage tumorigenesesnelheid, slechte homogeniteit en stabiliteit. Zebravissen worden vaak gebruikt om transgene of knock-out tumormodellen te construeren vanwege hun snelle reproductie20. Maar zebravisgenen verschillen van menselijke genen, dus hun toepassingen zijn beperkt.
Dit werk beschrijft de gedetailleerde procedures, voorzorgsmaatregelen en representatieve beelden voor het produceren van het primaire osteosarcoom in het scheenbeen met pulmonale metastase via intratibia-injectie van osteosarcoomcellen in athymische muizen. Deze methode werd toegepast om het primaire osteosarcoom in het tibia van muizen te creëren voor therapeutische werkzaamheidsevaluatie, die een hoge reproduceerbaarheid vertoonde21,22.

intratibial osteosarcoma cell injection to generate orthotopic osteosarcoma and lung metastasis mouse models

Osteosarcoom is de meest voorkomende primaire botkanker bij kinderen en adolescenten, met longen als de meest voorkomende gemetastaseerde plaats. De vijfjaarsoverleving van osteosarcoompatiënten met pulmonale metastase is minder dan 30%. Daarom is het gebruik van muismodellen die de ontwikkeling van osteosarcoom bij mensen nabootsen van groot belang voor het begrijpen van het fundamentele mechanisme van osteosarcoomcarcinogenese en pulmonale metastase om nieuwe therapieën te ontwikkelen. Hier worden gedetailleerde procedures gerapporteerd om de primaire osteosarcoom- en pulmonale metastasemuismodellen te genereren via intratibia-injectie van osteosarcoomcellen. In combinatie met het bioluminescentie- of röntgensysteem voor live beeldvorming, worden deze levende muismodellen gebruikt om de groei en metastase van osteosarcoom te monitoren en te kwantificeren. Om dit model vast te stellen, werd een keldermembraanmatrix met osteosarcoomcellen geladen in een microvolumespuit en geïnjecteerd in één scheenbeen van elke athymische muis na te zijn verdoofd. De muizen werden opgeofferd toen het primaire osteosarcoom de groottebeperking in het door de IACUC goedgekeurde protocol bereikte. De benen met osteosarcoom en de longen met uitzaaiingen werden gescheiden. Deze modellen worden gekenmerkt door een korte incubatietijd, snelle groei, ernstige laesies en gevoeligheid bij het monitoren van de ontwikkeling van primaire en pulmonale gemetastaseerde laesies. Daarom zijn dit ideale modellen voor het verkennen van de functies en mechanismen van specifieke factoren in osteosarcoom carcinogenese en pulmonale metastase, de tumormicro-omgeving en het evalueren van de therapeutische werkzaamheid in vivo.

Succesvolle orthotopische (primaire) osteosarcoom en gemetastaseerde longmodellen zijn afhankelijk van de nauwkeurige orthotopische injectie van osteosarcoomcellen. Dit protocol beschrijft de techniek om muismodellen te genereren met primair osteosarcoom en de pulmonale metastaselaesies. Door intratibiale injectie van osteosarcoomcellen bootst dit model het beste de klinische ontwikkelingskenmerken van tumorostepinoom na. Nauwkeurige aantallen osteosarcoomcellen worden rechtstreeks in het scheenbeen geïnjecteerd met behulp van een microvolumespuit, waardoor identieke tumorvormingssnelheden en tumorvolumes mogelijk zijn.Op de dag van osteosarcoomcelinjectie, was de cellen twee keer met PBS. Trypsinize de cellen met 1,5 milliliter van 0,25% trypsine gedurende drie minuten. Voeg vervolgens zes milliliter 10% serumbevattende MEM-media toe.Verzamel de cellen in een centrifugebuis van 15 milliliter. Zuig 20 microliter celsuspensie op in de kamer van de celtelplaat en bereken de celconcentratie met behulp van een automatische celteller. Centrifugeer de cellen gedurende vijf minuten bij kamertemperatuur op 800 maal g.Adem het supernatant op met een pipet, resuspend de celkorrel in een keldermembraanmatrix van 8,5 milligram per milliliter en plaats ze op ijs. Plaats de muis in rugligging. Houd de enkel van de muis vast met de duim en wijsvinger en desinfecteer de injectieplaats van het scheenbeen met een 70% ethanol wattenstaafje.Draai het enkelgewricht van elke muis naar buiten om het scheenbeen en kuitbeen te bewegen en buig het kniegewricht naar een geschikte positie totdat het proximale tibiaplateau zichtbaar is door de huid. Bevestig de naald aan een spuit van één milliliter en richt de punt van de naald in de richting van de injectieplaats, evenwijdig aan de as van het scheenbeen. Breng de naald percutaan in om een gat door het tibiaplatform naar het distale uiteinde van het scheenbeen te boren.Laad de osteosarcoomcelsuspensie die eerder op ijs is geplaatst in een microvolumespuit en vervang de spuit van één milliliter in het scheenbeen door de met osteosarcoomcellen geladen microvolumespuit. Injecteer langzaam ongeveer 10 microliter osteosarcoomcelsuspensie in het scheenbeen van elke athymische muis zonder veel druk uit te oefenen. Verwijder vervolgens de microvolumespuit van de injectieplaats en druk ongeveer 30 seconden met het wattenstaafje op de plaats.Plaats de muis terug in een schone kooi en observeer het herstel gedurende 10 minuten. Deze figuur toont het muismodel met orthotopisch osteosarcoom ontwikkeld uit intratibiale osteosarcoomcelinjectie en de geïsoleerde vorm ervan. Een geleidelijke toename van het tumorvolume wordt elke week waargenomen.De röntgenfoto's verkregen van de eerste tot en met de zesde week na injectie van luciferase-gelabelde osteosarcoomcellen toonden een geleidelijke toename van het volume. Bovendien toonde het beeld verkregen na de injectie in het scheenbeen van de muis orthotopische osteosarcoomgroei in vivo. De pulmonale metastase veroorzaakt door intratibiale injectie van de osteosarcoomcellen werd met succes in vivo gevolgd met behulp van een bioluminescentie live imaging-systeem.Observatie onder een stereomicroscoop onthulde gemetastaseerde kolonies in de geïsoleerde longweefsels. He-kleuring op paraffine-ingebedde longweefsels toonde gemetastaseerde laesies. De naald moet percutaan via het patellape ligament in het gewrichtskapsel worden ingebracht om een gat door het tibiaplatform naar het digitale scheenbeen te boren door de spuit te draaien.
