Alle Tierversuche wurden vom Tierschutzkomitee der Shanghai University of Traditional Chinese Medicine genehmigt. Das Osteosarkom ist der häufigste primäre Knochenkrebs bei Kindern und Jugendlichen1,2, der hauptsächlich das umliegende Gewebe infiltriert und sogar in die Lunge metastasiert, wenn die Patienten diagnostiziert werden. Lungenmetastasen sind die größte Herausforderung für die Osteosarkom-Therapie, und die Fünf-Jahres-Überlebensrate von Osteosarkom-Patienten mit Lungenmetastasen bleibt so niedrig wie 20% -30% 3,4,5. Die Fünf-Jahres-Überlebensrate des primären Osteosarkoms ist jedoch seit den 1970er Jahren aufgrund der Einführung der Chemotherapie auf etwa 70% gestiegen6. Daher ist es dringend notwendig, den grundlegenden Mechanismus der Osteosarkom-Karzinogenese und Lungenmetastasierung zu verstehen, um neuartige Therapien zu entwickeln. Die Anwendung von Mausmodellen, die das Fortschreiten des Osteosarkoms beim Menschen am besten nachahmen, ist von großer Bedeutung7.
Die Osteosarkom-Tiermodelle werden durch spontane, induzierte Gentechnik, Transplantation und andere Techniken erzeugt. Das Modell des spontanen Osteosarkoms wird aufgrund der langen Tumorbildungszeit, der inkonsistenten Tumorauftretensrate, der geringen Morbidität und der schlechten Stabilität selten verwendet 8,9. Obwohl das induzierte Osteosarkommodell zugänglicher ist als das spontane Osteosarkom, ist die Anwendung des induzierten Osteosarkommodells begrenzt, da der induzierende Faktor die Mikroumgebung, die Pathogenese und die pathologischen Merkmale des Osteosarkoms10 beeinflusst. Transgene Modelle helfen, die Pathogenese von Krebs zu verstehen, da sie die physiologischen und pathologischen Umgebungen des Menschen besser simulieren können; Die transgenen Tiermodelle haben jedoch auch ihre Grenzen aufgrund der Schwierigkeit, der langfristigen und der hohen Kosten der transgenen Modifikation. Darüber hinaus traten selbst in den am weitesten verbreiteten transgenen Tiermodellen, die durch p53- und Rb-Genmodifikation erzeugt wurden, nur 13,6% des Sarkoms in den vier Gliedmaßenknochenauf 11,12.
Die Transplantation ist aufgrund ihres einfachen Manövers, der stabilen Tumorbildungsrate und der besseren Homogenität in den letzten Jahren eine der am häufigsten verwendeten primären und fernmetastasierenden Krebsmodell-produzierenden Methoden13. Die Transplantation umfasst die heterotope Transplantation und die orthotope Transplantation entsprechend den Transplantationsstellen. Bei der heterotopen Osteosarkomtransplantation werden die Osteosarkomzellen außerhalb der primären Osteosarkomstellen (Knochen) der Tiere, üblicherweise unter die Haut, subkutan14 injiziert. Obwohl die heterotope Transplantation unkompliziert ist, ohne dass eine Operation an Tieren durchgeführt werden muss, stellen die Stellen, an denen die Osteosarkomzellen injiziert werden, nicht die tatsächliche Mikroumgebung des menschlichen Osteosarkoms dar. Osteosarkom orthotope Transplantation ist, wenn die Osteosarkom-Zellen in die Knochen der Tiere, wie Tibia15,16, injiziert werden. Im Vergleich zu den heterotopen Transplantaten zeichnen sich orthotope Osteosarkomtransplantate durch eine kurze Inkubationszeit, schnelles Wachstum und starke erosive Natur aus; Daher sind sie ideale Tiermodelle für osteosarkombezogene Studien17.
Die am häufigsten verwendeten Tiere sind Mäuse, Hunde und Zebrafische18,19. Das spontane Modell des Osteosarkoms wird normalerweise bei Hunden verwendet, da das Osteosarkom einer der häufigsten Tumoren bei Hunden ist. Die Anwendung dieses Modells ist jedoch aufgrund der langen Tumorbildungszeit, der niedrigen Tumorentstehungsrate, der schlechten Homogenität und Stabilität begrenzt. Zebrafische werden aufgrund ihrer schnellen Reproduktion häufig zur Konstruktion transgener oder Knockout-Tumormodelleverwendet 20. Aber Zebrafischgene unterscheiden sich von menschlichen Genen, so dass ihre Anwendungen begrenzt sind.
Diese Arbeit beschreibt die detaillierten Verfahren, Vorsichtsmaßnahmen und repräsentativen Bilder zur Erzeugung des primären Osteosarkoms in der Tibia mit pulmonaler Metastasierung durch Intratibia-Injektion von Osteosarkom-Zellen in athymischen Mäusen. Diese Methode wurde angewendet, um das primäre Osteosarkom in Maus-Tibia für die therapeutische Wirksamkeitsbewertung zu erzeugen, die eine hohe Reproduzierbarkeitzeigte 21,22.

intratibial osteosarcoma cell injection to generate orthotopic osteosarcoma and lung metastasis mouse models

Das Osteosarkom ist der häufigste primäre Knochenkrebs bei Kindern und Jugendlichen, wobei die Lunge die häufigste metastasierende Stelle ist. Die Fünf-Jahres-Überlebensrate von Osteosarkom-Patienten mit Lungenmetastasen beträgt weniger als 30%. Daher ist die Verwendung von Mausmodellen, die die Osteosarkomentwicklung beim Menschen nachahmen, von großer Bedeutung für das Verständnis des grundlegenden Mechanismus der Osteosarkomkarzinogenese und Lungenmetastasierung zur Entwicklung neuartiger Therapeutika. Hier werden detaillierte Verfahren berichtet, um die primären Osteosarkom- und Lungenmetastasen-Mausmodelle über die Intratibia-Injektion von Osteosarkom-Zellen zu erzeugen. In Kombination mit dem Biolumineszenz- oder Röntgen-Live-Imaging-System werden diese lebenden Mausmodelle verwendet, um das Wachstum und die Metastasierung von Osteosarkomen zu überwachen und zu quantifizieren. Um dieses Modell zu etablieren, wurde eine Basalmembranmatrix, die Osteosarkomzellen enthielt, in eine Mikrovolumenspritze geladen und nach der Anästhesie in eine Tibia jeder athymischen Maus injiziert. Die Mäuse wurden geopfert, als das primäre Osteosarkom die Größenbeschränkung im IACUC-genehmigten Protokoll erreichte. Die Beine mit Osteosarkom und die Lunge mit Metastasenläsionen wurden getrennt. Diese Modelle zeichnen sich durch eine kurze Inkubationszeit, schnelles Wachstum, schwere Läsionen und Empfindlichkeit bei der Überwachung der Entwicklung primärer und pulmonaler metastasierender Läsionen aus. Daher sind dies ideale Modelle, um die Funktionen und Mechanismen spezifischer Faktoren in der Osteosarkom-Karzinogenese und Lungenmetastasierung, der Tumormikroumgebung und der Bewertung der therapeutischen Wirksamkeit in vivo zu untersuchen.

Erfolgreiche orthotope (primäre) Osteosarkom- und metastasierende Lungenmodelle hängen von der genauen orthotopen Injektion von Osteosarkomzellen ab. Dieses Protokoll beschreibt die Technik zur Generierung von Mausmodellen mit primärem Osteosarkom und den pulmonalen Metastasenläsionen. Durch die intratibiale Injektion von Osteosarkomzellen ahmt dieses Modell die klinischen Entwicklungsmerkmale des Tumorosteosarkoms am besten nach. Eine genaue Anzahl von Osteosarkomzellen wird mit einer Mikrovolumenspritze direkt in die Tibia injiziert, was identische Tumorbildungsraten und Tumorvolumina ermöglicht.Am Tag der Osteosarkom-Zellinjektion waschen Sie die Zellen zweimal mit PBS. Trypsinisieren Sie die Zellen mit 1,5 Millilitern 0,25% Trypsin für drei Minuten. Dann fügen Sie sechs Milliliter 10% serumhaltige MEM-Medien hinzu.Sammeln Sie die Zellen in einem 15-Milliliter-Zentrifugenröhrchen. Saugen Sie 20 Mikroliter Zellsuspension in die Kammer der Zellzählplatte und berechnen Sie die Zellkonzentration mit einem automatischen Zellzähler. Zentrifen Sie die Zellen bei 800 mal g für fünf Minuten bei Raumtemperatur.Saugen Sie den Überstand mit einer Pipette ab, suspendieren Sie das Zellpellet in einer 8,5-Milligramm-pro-Milliliter-Basalmembranmatrix und legen Sie sie auf Eis. Stellen Sie die Maus in Rückenlage. Halten Sie den Knöchel der Maus mit Daumen und Zeigefinger fest und desinfizieren Sie die Injektionsstelle der Tibia mit einem 70% igen Ethanolabstrich.Drehen Sie das Sprunggelenk jeder Maus nach außen, um die Tibia und das Wadenbein zu bewegen, und beugen Sie das Kniegelenk in eine geeignete Position, bis das proximale Tibiaplateau durch die Haut sichtbar ist. Befestigen Sie die Nadel an einer Ein-Milliliter-Spritze und richten Sie die Nadelspitze parallel zur Tibiaachse auf die Injektionsstelle. Führen Sie die Nadel perkutan ein, um ein Loch durch die Tibia-Plattform in Richtung des distalen Endes der Tibia zu bohren.Laden Sie die zuvor auf Eis gelegte Osteosarkom-Zellsuspension in eine Mikrovolumenspritze und ersetzen Sie die Ein-Milliliter-Spritze in der Tibia durch die mit Osteosarkomzellen beladene Mikrovolumenspritze. Injizieren Sie langsam etwa 10 Mikroliter Osteosarkom-Zellsuspension in die Tibia jeder athymischen Maus, ohne viel Druck auszuüben. Entfernen Sie dann die Mikrovolumenspritze von der Injektionsstelle und drücken Sie die Stelle mit dem Wattestäbchen etwa 30 Sekunden lang an.Setzen Sie die Maus zurück in einen sauberen Käfig und beobachten Sie ihre Erholung für 10 Minuten. Diese Abbildung zeigt das Mausmodell mit orthotopem Osteosarkom, das sich aus der intratibialen Osteosarkom-Zellinjektion entwickelt hat, und seiner isolierten Form. Jede Woche wird eine allmähliche Zunahme des Tumorvolumens beobachtet.Die Röntgenbilder, die von der ersten bis zur sechsten Woche nach der Injektion von Luciferase-markierten Osteosarkomzellen aufgenommen wurden, zeigten eine allmähliche Zunahme des Volumens. Darüber hinaus zeigte das nach der Injektion in die Maustibia erhaltene Bild ein orthotopes Osteosarkomwachstum in vivo. Die durch die intratibielle Injektion der Osteosarkomzellen verursachte pulmonale Metastasierung wurde in vivo mit einem Biolumineszenz-Live-Imaging-System erfolgreich verfolgt.Die Beobachtung unter einem Stereomikroskop ergab metastasierende Kolonien in den isolierten Lungengeweben. Die HE-Färbung auf paraffineingebettetem Lungengewebe zeigte metastasierende Läsionen. Die Nadel sollte perkutan durch das Patellaband in die Gelenkkapsel eingeführt werden, um ein Loch durch die Tibia-Plattform in Richtung der digitalen Tibia zu bohren, indem die Spritze gedreht wird.
