Tandvorming begint met de invaginatie van het orale epitheel1. Volgens het tandontwikkelingsstadium heeft het orale epitheel verschillende namen, waaronder het binnenste en buitenste glazuurepitheel, cervicale lus en hertwig's epitheliale wortelschede (HERS). De epitheelcompartimenten communiceren met de omliggende mesenchymale cellen. Epitheliale-mesenchymale interacties reguleren tandvorming en weefselregeneratie. Het verkrijgen van tandheelkundige epitheelcellen, zoals orale keratinocyten en Hertwig's epitheliale wortelschedecellen (HERSC's), is cruciaal voor de studie van tandheelkundige epitheliale-mesenchymale interacties2.
Van knaagdieren afgeleide tandheelkundige epitheelcellen worden geïsoleerd uit de epitheelstructuur, zoals de HERS. Li en collega's isoleerden en vereeuwigden ratkiezen-afgeleide HERSC's na het oogsten van het apicale deel van de zich ontwikkelende tandkiemen van 8 dagen oude ratten3. Het HERS werd onder vergroting gescheiden van apicale weefsel. Gezien het ontwikkelingsstadium en de leeftijd van de tand, is het oogsten van de HERS van mensen bijna onmogelijk vanwege ethische kwesties: een zich ontwikkelende tandkiem moet van een jong kind worden verwijderd om de menselijke HERS te oogsten. Onrijpe tandkiemen worden zelden geëxtraheerd. Menselijke tandheelkundige epitheelcellen kunnen worden geïsoleerd uit het tandvlees en het parodontale ligament (PDL). Epitheliale structuur-afgeleide cellen nemen deel aan tandvorming samen met mesenchymale componenten en kunnen meer geschikt zijn voor de studie van tandheelkundige epitheliale-mesenchymale interacties dan orale keratinocyten. Epitheelcelresten van Malassez (ERM) zijn hers-afgeleide epitheliale restanten en verblijven in kleine aantallen in de PDL4. Studies melden de isolatie van menselijke HERSC's van PDL5. Het oogsten van menselijke HERSC's uit PDL-weefsel is echter niet altijd succesvol vanwege de schaarste van de epitheliale populatie op deze locatie5,6.
Hoewel van knaagdieren afgeleide HERSC's worden gehandhaafd in serumbevattende media3,7, worden menselijke DF-afgeleide epitheelcellen gekweekt met serumvrije media die vergelijkbaar zijn met andere menselijke epitheelcellen, zoals normale menselijke epidermale keratinocyten en normale menselijke orale keratinocyten8,9. Dit impliceert fysiologische of functionele verschillen tussen tandheelkundige epitheelcellen van knaagdieren en menselijke tandheelkundige epitheelcellen. Inzicht in het mechanisme met betrekking tot tandheelkundige epitheliaal-mesenchymale interacties kan bijdragen aan de ontwikkeling van klinische toepassingen, waaronder parodontale herbevestiging tijdens herplanting, parodontale regeneratie bij parodontitis, pulp-dentine complex regeneratie en bio-tandgeneratie. Gezien de kenmerken van translationeel onderzoek, kunnen menselijke tandheelkundige epitheelcellen geschikter zijn dan knaagdier tandheelkundige epitheelcellen voor de studie van epitheel-mesenchymale interacties.
De menselijke DF is een los bindweefsel en bevindt zich vaak in een aangetaste tand. De DF bevat mesenchymale voorlopers10. Voor zover wij weten, heeft echter geen enkele studie de isolatie van epitheelcellen uit tandzakjes vóór 2021 gemeld. Oh en Yi meldden de isolatie van epitheelcellen van menselijke DF in 20218. Het epitheliale fenotype werd bevestigd door western blotting en morfologische analyse. Analyse van de oorsprong van DF-afgeleide epitheelcellen toonde vergelijkbare resultaten met andere studies. DF-afgeleide epitheelcellen waren noch endotheel noch hematopoietisch5,11, en Oh en Yi stelden voor om deze cellen als DF-HERSC's te benoemen. De DF heeft een groter volume dan de PDL en er kunnen meer epitheelcellen uit de DF worden geïsoleerd. Dit verbetert de opkomst van epitheliale kolonies en resulteert in een hoog slagingspercentage bij het oogsten van epitheelcellen uit DF. Deze studie suggereert het gebruik van de DF als een weefselbron voor de isolatie van tandheelkundige epitheelcellen.
In de huidige studie werden afzonderlijke cellen geïsoleerd uit de DF volgens eerder beschreven procedures10,12. De DF bevat heterogene celpopulaties en verschillende celtypen kunnen in een vroeg stadium van de procedure aanwezig zijn. Morsczek en collega's isoleerden DF-afgeleide mesenchymale stamcellen10. We veronderstelden dat de DF epitheelcellen bevat en dat alleen epitheelcellen kunnen overleven onder serumvrije omstandigheden. Deze studie verschilt van die van Morsczek et al. in termen van de selectie van de epitheliale populatie en de remming van mesenchymale cellen. De selectie werd uitgevoerd met behulp van keratinocyten serumvrije media (SFM), die epitheelcelproliferatie mogelijk maken en mesenchymale celproliferatie remmen. Deze studie is voortgekomen uit een rapport van Oh en Yi8. Het doel van deze studie was om details te beschrijven van de methode die in dat rapport wordt gebruikt voor de isolatie van epitheelcellen van menselijke DF.

De tandheelkundige follikel (DF) werd geoogst tijdens het verwijderen van een aangetaste derde kies door een orale maxillofaciale chirurg. Epitheelcelisolatie werd uitgevoerd op de dag van de DF-oogst. De DF werd drie keer gewassen met DPBS en vervolgens ontleed met een weefselschaar totdat het weefsel een pulpachtige of squishy consistentie had. Eencellige populaties werden gepelletiseerd door centrifugering en gewassen met keratinocyten serumvrij medium. Heterogene celpopulaties werden verdeeld in een kweekschaal. Keratinocyten serumvrij medium werd gebruikt om de epitheelcellen te selecteren. Het kweekmedium werd dagelijks verwisseld totdat er geen drijvend puin of dode cellen werden waargenomen. Epitheelcellen verschenen binnen 7-10 dagen na de verdeling van de celpopulatie. Epitheelcellen overleefden in serumvrij medium, terwijl α-modificatie minimaal essentieel medium aangevuld met 10% foetaal runderserum de proliferatie van mesenchymale cellen mogelijk maakte. De DF is een weefselbron voor de isolatie van tandheelkundige epitheelcellen.
Het doel van deze studie was om een methode vast te stellen voor de isolatie van epitheelcellen van menselijke DF. Parodontaal ligament (PDL) werd gebruikt voor de isolatie van menselijke tandheelkundige epitheelcellen. Het verkrijgen van epitheelcellen van menselijke PDL is niet altijd succesvol vanwege het kleine weefselvolume, wat leidt tot lage aantallen epitheelcellen. DF heeft een groter volume dan PDL en bevat meer cellen. DF kan een weefselbron zijn voor de primaire kweek van menselijke tandheelkundige epitheelcellen. Dit protocol is eenvoudiger en efficiënter dan de isolatiemethode met PDL. Het verkrijgen van menselijke tandheelkundige epitheelcellen kan verdere studies van tandheelkundige epitheliale-mesenchymale interacties vergemakkelijken.

DF oogsten De operatie werd uitgevoerd door een kaakchirurg. Van de mens afgeleide materialen, waaronder het tandfragment, gingivaal weefsel en DF, werden verzameld door een chirurg (figuur 1A). De DF kan aan het tandfragment worden bevestigd. Een orale maxillofaciale chirurg zal in staat zijn om de DF te identificeren. Dit protocol demonstreert een verbeterde methode voor het verkrijgen van menselijke tandheelkundige epitheelcellen. Het is moeilijk om de geaggregeerde celkorrel te identificeren tijdens de procedures. Wees voorzichtig wanneer u supernatant hanteert om het verlies van celpopulaties te voorkomen.Om te beginnen, oogst tandheelkundige follikels tijdens de chirurgische extractie van aangetaste derde kiezen. Haal het eruit en plaats het in buis 2. Nogmaals, schud het 10 tot 15 keer.Haal het er ten slotte uit en plaats het in buis 3 en schud het vervolgens. Plaats na drie keer wassen de tandfollikel in een kweekschaal van 60 millimeter. Hak het weefsel fijn met een weefselschaar totdat de tandfollikel een pulpy of squishy uiterlijk heeft.Gebruik een klein zijgedeelte van de kweekschaal om weefselverlies te minimaliseren. Meng elk één milliliter collagenase type 1 en protease-oplossing in een conische buis van 15 milliliter. Breng het gehakte weefsel over in de conische buis van 15 milliliter, schud het voorzichtig en incubeer het gedurende een uur bij 37 graden Celsius.Voeg vijf milliliter van 0,05% trypsine-EDTA toe aan de buis. Schud het en incubeer gedurende 15 minuten bij 37 graden Celsius. Bereid keratinocytenmedium met keratinocytenserumvrij medium en 10% foetaal runderserum.Voeg drie milliliter keratinocytenmedium toe aan een nieuwe conische buis van 50 milliliter. Plaats een zeef van 40 micrometer bovenop deze buis. Zuig vervolgens het supernatant uit de buis met het weefsel met een serologische pipet van 10 milliliter en laat het door de zeef gaan.Breng de verzamelde suspensie over in een conische buis van 15 milliliter. Centrifugeer de buis en verwijder het supernatant. Voeg vervolgens drie milliliter keratinocytenserumvrij medium toe.Centrifugeer nog drie minuten en verwijder het supernatant. Plaats de eencellige populatie in een kweekschaal van 60 millimeter met behulp van het serumvrije medium voor keratinocyten. Houd de kweekschaal met een eindvolume van vijf milliliter in een bevochtigde atmosfeer van 5% koolstofdioxide bij 37 graden Celsius.Cellen met epitheliale morfologie verschenen binnen zeven tot 10 dagen na het platen van eencellige populaties. Het aantal geplaveide epitheelcellen varieerde van één tot 10 op het moment van opkomst en de cellen breidden zich in de loop van de tijd uit tot kolonies. De meest cruciale stap is het hakken van de tandfollikel in kleine deeltjes.Het verbetert de scheiding van afzonderlijke cellen van follikelweefsel. Mesenchymale cellen kunnen worden geïsoleerd uit de menselijke tandheelkundige follikel. Serumbevattende kweekomgeving selecteert mesenchymale cellen en remt epitheliale groei van eencellige populaties.Dit protocol biedt de methode voor het verkrijgen van de menselijke tandheelkundige epitheelcellen. Tandheelkundige follikel-afgeleide epitheelcellen kunnen worden gebruikt voor de studie van tandheelkundige epitheliale-mesenchymale interacties.
