このプロトコルは、細胞を選別することなく、複雑な組織内の少数の細胞から高品質の細胞種特異的RNAを取得することを目的としています。最近入手可能なマウスモデルを採用しており、研究者はGLPプルダウンを使用してリボソーム結合RNAを単離することができます。この方法は、任意のEGFP発現細胞からリボソーム結合RNAを単離するのにも適している。まず、2ミリリットルの氷冷均質化作業バッファーをガラスティッシュグラインダーセットに追加します。凍結したサンプルをグラインダーにすばやく入れ、緩い乳棒を使用して氷上で30ストロークで組織を均質化します。ホモジネートを2ミリリットルの丸底チューブに移し、摂氏4度で10分間12, 000gで遠心分離します。次に、ペレットを乱すことなく上清を新しい2ミリリットルのチューブに移し、入力として100マイクロリットルを予約します。上清を抗GFP抗体とともに摂氏4度でエンドオーバーエンドローテーターで一晩インキュベートします。ホモジネートと抗体の混合物を、洗浄したプロテインgビーズを含む新しい2ミリリットルの丸底チューブに移し、ローテーター上で摂氏4度、24 RPMで2時間インキュベートしました。磁気ラックを使用して磁気ビーズを上澄みから分離します。上清を、EGFP陰性細胞のRNAおよびリボソームに結合していないEGFP陽性細胞のRNAを含む陰性画分として保存する。1ミリリットルの高塩分洗浄バッファーをビーズに加え、チューブを短時間ボルテックスしてビーズを洗浄します。次に、チューブを磁気ラックに入れます。洗浄バッファーを除去し、洗浄ステップをさらに2回繰り返して、EGFP陽性細胞からビーズリボソームRNA複合体を得た。RNA分離キットからの50マイクロリットルの抽出バッファーでビーズをサーモミキサーで30分間インキュベートし、ビーズからRNAを放出します。磁気ラックでビーズを分離し、上清を1.5ミリリットルのチューブに移します。チューブを3000gで2分間遠心分離します。次に、上清を新しい1.5ミリリットルのチューブにピペットで入れます。RNA精製カラムをプレコンディショニングするには、250マイクロリットルのコンディショニングバッファーを精製カラムにピペットで移し、室温で5分間インキュベートします。次にカラムを16, 000 gで1分間遠心分離します。等量の70%エタノールを細胞抽出液にピペットで入れ、ピペッティングでよく混ぜます。最大200マイクロリットルの混合物を、予め調整されたRNA精製カラムにピペットで入れます。サンプルの損失を減らすために、カラムをその容量の80%を超えて満たさないでください。すべてのRNAが各フラクションの同じカラムに収集されるまで、この手順を数回繰り返します。カラムを2分間遠心分離して、RNAをカラム内のメンブレンに結合させます。その後、遠心分離を30秒間続けます。フロースルーを廃棄し、100マイクロリットルの洗浄バッファー1をカラムにピペットで入れます。1分間遠心分離してから、フロースルーを廃棄します。ピペット75マイクロリットルのDNA溶液を精製カラム膜に直接混合します。室温で15分間インキュベートします。次いで、40マイクロリットルの洗浄バッファー1をカラムにピペットで入れ、さらに30秒間遠心分離する。フロースルーを破棄します。100マイクロリットルの洗浄バッファー2液をカラムにピペットし、8, 000 gで1分間遠心分離します。フロースルーを破棄します。PET100マイクロリットルの洗浄バッファー2をカラムに入れ、16, 000 gで2分間遠心分離します。フロースルーを廃棄し、同じカラムを16, 000 gで1分間再遠心分離して、洗浄バッファーの痕跡をすべて除去します。カラムを新しい1.5ミリリットルのマイクロ遠心チューブに移します。12マイクロリットルのRNase遊離水を精製カラムのメンブレンに直接ピペットで送り、ピペットチップがメンブレンに触れないようにします。室温で1分間インキュベートし、1000 gで1分間遠心分離します。その後、16, 000 Gで1分間RNAを溶出します。バイオアナライザーを使用して、抽出されたRNAの質と量を評価します。RNAを80度の冷凍庫に保存するか、さらなる分析のために送り出します。遺伝子操作された遺伝子対立遺伝子であるGli1-CreERT2とNuTRAPの両方を持つマウスに、タモキシフェンを1日1回、1日おきに3回注射します。採取した組織の免疫蛍光分析は、EGFPが精巣の間質細胞で発現したことを示しています。EGFPは、NuTRAPマウスであるGli1-CreERT2の副腎莢膜細胞にも見られました。ソニックヘッジホッグ-CreERT2では、NuTRAPマウス EGFP陽性細胞集団は、カプセルの下の副腎の外皮質に存在し、EGFP事前発現細胞の発現を確認した。細胞種特異的RNAの抽出後、抽出したRNAをマイクロアレイ解析のために送った。結果は、陰性画分の遺伝子と比較して、約3000個の遺伝子が陽性画分で濃縮されたのに対し、陰性画分では約4, 000個の遺伝子が富化されていることを示した。差次発現遺伝子のうち、ライディッヒ細胞関連遺伝子Hsd11b1およびHsd3b6は陽性画分に富んでいた。陰性画分では、セルトリ細胞関連遺伝子デザートヘッジホッグおよびGstm6が富化されていた。陰性画分と入力を比較することで、発現差のある遺伝子はごくわずかしか同定されなかった。リアルタイム定量RT-PCRを使用して、陽性および陰性画分における主要遺伝子の発現を確認しました。マイクロアレイアッセイで見出されたものと同様に、ステロイド生成酵素3−βヒドロキシステロイドデヒドロゲナーゼおよびコレステロール側鎖切断酵素が陽性画分において富化されていた。一方、セルトリ細胞マーカーSRYボックス転写因子9種および生殖細胞マーカーシナプトン複合体タンパク質3種は陰性画分に富化していた。このプロトコルを試みる場合、使用前にのみDTT、シクロヘキシミド組換えリボヌクレアーゼ、およびプロテイナーゼ阻害剤カクテルをホモジナイズストック溶液に加える新鮮なホモジナイズ作業バッファーを調製することが重要です。使用前に新鮮な低塩および高塩洗浄バッファーを準備することも重要です。
