このプロトコルは、再現性の高い方法でさまざまな細菌から細胞外小胞を分離する方法を提供するため、重要です。この技術の素晴らしいところは、細菌培養の非常に大規模な環境からEVを分離するために使用できるため、in vivo研究が可能になることです。私たちが示すデータは前臨床アプリケーションを反映していますが、このプロトコルが治療用の細菌性EVの製造に適応できることは予測可能です。この方法は、わずかな変更でスケーラブルな細胞培養システムに適用できます。プロトコルのスケーラビリティのため、必要な開始量の細菌細胞培養を処理するために、適切なサイズのフィルターとSECカラムを用意することが重要です。手順を実演しているのは、ジャスティン・ラティア博士の研究室のアシスタントテクノロジストであるセイディー・ジョンソン、ジャスティン・ラティア博士の研究室の腫瘍学フェローであるディオニュシウス・ワトソン、そして私の研究室の研究技術者であるアキーム・サントスです。まず、滅菌ループを使用して、大腸菌の単一コロニーを250〜1000ミリリットルのルリアベルターニまたはLBブロスに接種します。次いで、培養物を振とうインキュベーター内で毎分300回転、摂氏37度で好気的にインキュベートする。48時間のインキュベーション後、細胞培養物を清浄な500ミリリットルのポリプロピレン製遠心分離機ボトルに移し、大容量の固定角度ローターで摂氏4度、5, 000倍Gで15分間遠心分離することにより、細菌培養培地を清澄化します。上清を10, 000倍Gで15分間慎重に遠心分離機に注ぎ、上清をきれいな遠心分離機ボトルに移します。次に、上澄み液を適切なサイズの0.2ミクロンのポリエーテルスルホン真空駆動フィルター装置に移し、ろ過装置を真空壁供給に接続します。ろ過速度が大幅に低下した場合は、ろ過されていない材料を新しいデバイスに移動します。ろ過された媒体は、摂氏4度で一晩保存することも、すぐにさらに処理することもできます。生細胞が完全に除去されたことを確認するには、ろ過した上清のアリコートを適切な寒天プレートに広げ、細菌株に最適な条件でインキュベートした後にコロニーがないことを確認します。100ミリリットルの容量でろ過された培地を濃縮するには、90ミリリットルのろ過された培養液を100キロダルトンの分子量カットオフを備えた遠心限外ろ過装置のリザーバーにロードし、上部のリザーバーの体積が0.5ミリリットル未満に濃縮されるまで、摂氏4度と2、000倍Gで15〜30分間隔でスイングバケットローターで培地を遠心分離します。残りのろ過された培養培地をリザーバーに補充するには、デバイスの下部にあるフロースルーを取り外してバランスを取り直します。リザーバー内の濃縮培地の粘度が目に見えて上昇した場合は、培地をPBSで希釈し、遠心分離によって再濃縮して、分子量カットオフの100キロダルトンよりも小さい非細胞外小胞またはEVタンパク質を減らします。次に、濃縮培地を低タンパク質結合チューブに移し、摂氏4度で一晩保存します 100ミリリットルを超える容量のろ過媒体を濃縮するには、分子量カットオフが100キロダルトンの適切なサイズの接線フローろ過またはTFFデバイスを選択します。原稿で説明されているようにろ過回路を組み立てた後、200ミリリットルのPBSをメス容器にポンプで送り、目的の流量に対応する適切な速度を決定します。速度が設定されたら、ろ過された馴化培地を室温で毎分約200ミリリットルで循環させ、100キロダルトン未満の分子を収集し、馴化培地の体積が100〜200ミリリットルに減少するまで、別の容器内の廃棄物として限外ろ過膜を通過します。ろ過した容量をPBSで順次2倍に希釈し、ポンプで培地を小さな容器に循環させ続け、容量を75〜100ミリリットルに濃縮し、次に25ミリリットルと10ミリリットルに濃縮します。フィードチューブをサンプルリザーバーから持ち上げ、ポンプでフィルターをパージして、最大量のサンプルを回収します。濃縮サンプルを分子量カットオフ100キロダルトンの15ミリリットルの遠心限外ろ過装置に移動し、上部リザーバー内の培地の体積が2ミリリットル未満になるまで、摂氏4度、G2, 000倍のGで15〜30分間隔でスイングバケットローターで遠心分離します。濃縮培地を低タンパク質結合チューブに移し、摂氏4度で一晩保存します。EVを単離するには、100ミリリットル未満の出発原料に対して10ミリリットルの床容量の小さなカラムを使用し、100ミリリットルを超える出発物質に対して47ミリリットルの床容量のより大きなカラムを使用して、サイズ排除クロマトグラフィー(SEC)を実行します。実験の数時間前に、SECカラムとPBSを室温に戻し、続いて標準的なラボスタンドとホルダーを使用してカラムを垂直位置に安定させます。SECカラムに接続する前に、5ミリリットルのPBSをフリットを通って廃棄物容器に流して、サンプルリザーバーをハイドレートします。次に、カラムのインレットキャップを緩めてサンプルリザーバーに2ミリリットルのPBSを追加し、PBSがフリットから滴り落ちるようにリザーバーをカラムに注意深く接続します。カラムを平衡化するために、47ミリリットルのPBSをサンプルリザーバーに加え、続いてSECカラムの底部のキャップを外します。次に、ロードされたすべてのサンプルバッファーをカラムに流し、フロースルーを破棄します。2ミリリットルのサンプルをサンプルリザーバーにロードした後、サンプルをカラムに完全に入れ、フロースルーを収集します。直ちに14.25ミリリットルの容量からサンプル容量を引いた容量でPBSをサンプルリザーバーに追加し、カラムボイドボリュームに等しい量を廃棄しながら、溶液がカラムを流れるようにします。次に、2ミリリットルの低結合マイクロチューブをSECカラムの真下に配置し、2ミリリットルのPBSをカラムに通した後、最初のフロースルーまたはフラクション1を収集します。引き続き2ミリリットルのPBSをサンプルリザーバーに追加して、後続の各フラクションを収集します。収集した画分は、短期保管の場合は摂氏4度、長期保存の場合はマイナス80°Cで保管します。EVの回収画分の無菌性試験では、アッセイに使用する画分100マイクロリットルを3ミリリットルの培養液に接種します。その後、濁度を観察しながら最適な条件下で培地を少なくとも3日間培養する。あるいは、画分サンプルを、産生細菌を増殖させるために使用される培地を含む寒天プレートに適用し、次いでコロニー形成を検査することができる。次に、凍結融解サイクルを避けるために、個々の画分またはプールされた画分の25〜50%を摂氏マイナス80度の低タンパク質結合チューブに移してEVを保管します。代表的な分析は、初期のクロマトグラフィー画分における大腸菌MP1 EVの溶出を示しています。フラクション1〜6は、平均直径が100ナノメートル未満のEVが最も多く含まれていました。同時に、その後の画分にはEVフリーのタンパク質が含まれており、EVはほとんど含まれていませんでした。EVの濃縮とサイズは、透過型電子顕微鏡によって、特にフラクション2〜6で確認されました。EV濃縮画分2〜7はナノルシフェラーゼ活性が高いが、後の画分と比較して非EV関連mCherryからの蛍光シグナルは非常に低いことが観察された。イムノゴールド標識により、画分2〜5のナノルシフェラーゼのEV会合が確認されました。他の細菌種へのプロトコルの適用性は、多様な嫌気性細菌の培養物からEVを単離することによって評価されました。初期のクロマトグラフィー画分1〜4は、直径サイズが100ナノメートル未満のEV用に濃縮されていることが観察されました。複雑な脳と心臓の注入(BHI)は、EVの総収量の25%未満でEVサイズの粒子を含んでいました。細菌細胞培養の開始量を処理できるTFFデバイスを使用することが重要です。この技術により、複雑な動物モデルにおけるEVの機能について生物学的に疑問を投げかけるのに十分なEVを生成できます。
