대기 오염은 높은 모든 원인으로 인한 사망률과 관련이 있으며 수질 오염, 토양 오염 및 직업적 노출의 합보다 질병의 상당한 부담에 기여했습니다1. WHO의 보고서에 따르면 실외 대기 오염으로 인해 2016 년 전 세계 도시와 농촌 지역에서 420 만 명의 조기 사망이 발생했습니다2. 전 세계 인구의 91%가 대기 질이 WHO 지침 한도를 초과하는 장소에 살고 있습니다2. 또한, 미세 입자상 물질 (PM) (직경 ≤2.5 μm, PM2.5)은 전 세계 공중 보건3, 특히 저소득 및 중간 소득 국가의 도시에 거주하는 사람들에게 가장 심각한 대기 오염 위협으로 인식되고 있습니다.
심혈관 질환에 대한 대기 오염의 악영향은 더 많은 관심을 기울일 가치가 있습니다. 이전 연구에 따르면 PM은 심혈관 질환(CVD)의 위험을 증가시킵니다4. 고농도의 초미세 입자에 몇 시간 동안 노출되면 심근 경색 사망률이 증가 할 수 있습니다. 심근경색 병력이 있는 사람의 경우 초미세 입자에 노출되면 재발 위험이 크게 증가할 수있습니다5. 더욱이, PM 노출이 죽상 동맥 경화증6의 진행을 가속화한다는 것이 일반적으로 인정된다.
의학 연구를 위해서는 적합한 동물 모델을 선택하는 것이 중요합니다. 단순 죽상동맥경화증 동물 모델7, 심근 허혈 동물 모델8, 및 PM 노출 동물 모델9가 이미 존재한다. ApoE-/- (아포지단백질 E 녹아웃) 마우스는 죽상동맥경화증 연구에 사용되는 전통적인 마우스 모델이다. 이 프로토콜은 죽상동맥경화증(AS)과 PM 급성 노출을 조합한 심근 허혈(MI)의 동물 모델을 개발하는 것을 설명합니다. ApoE−/− 마우스는 죽상동맥경화증을 유발하기 위해 고지방 식단을 공급받았다.
