인간 각막은 5개의 주요 층으로 구성되어 있으며 안구 굴절에 중추적인 역할을 하여 안구 조직을 보호하기 위해 시력과 구조적 무결성을 유지합니다1. 각막의 가장 바깥쪽 부분은 각막 상피로, 기저 세포에서 순차적으로 분화하여 안구 표면에서 위쪽으로 이동하여 흘리는 5-6개의 세포층으로 구성되어 있다1. 인간 및 뉴질랜드 토끼의 각막과 비교하여, 마우스 각막은 유사한 각막 구조를 갖지만, 상피 및 간질의 감소된 두께로 인해 중심 부분보다 주변부가 더 얇다2. 안구 광학 시스템에서의 독특한 위치 때문에 기계적 손상, 박테리아 접종 및 화학 작용제와 같은 많은 외부 손상은 쉽게 상피 무결성을 위험에 빠뜨릴 수 있으며 더 나아가 시력을 위협하는 상피 결함, 감염성 각막염, 각막 용해 및 각막 천공을 유발할 수 있습니다.

기계적 및 화학적 각막 손상을 연구하기 위해 생체 외 및 생체 내 동물 모델을 설정했습니다. 이 기술은 연구자들이 각막의 기계적 및 화학적 손상을 연구할 수 있는 플랫폼을 쉽게 구축할 수 있는 플랫폼을 제공합니다. 쥐 각막 상피 상처를 만들려면 피부 생검 펀치를 사용하여 마취 된 마우스의 중앙 각막을 표시하여 상처의 경계가 잘 정해져 있고 측정 가능한 부위를 확인합니다.펀치를 중앙 각막 위로 부드럽게 움푹 들어가 원형 표시를 남기고 Bowman's layer가 손상되지 않도록 0.5mm 버가 있는 휴대용 각막 녹 링 제거제를 사용하여 각막 상피를 Bowman's 층까지 괴사시킵니다. 각막 집게로 상처 가장자리에 남아 있는 느슨한 조직을 제거합니다. 플루오레세인 종이에 생리 식염수 한 방울을 떨어뜨려 플루오레세인을 용해시킨 다음 플루오레세인 함유 방울을 쥐 상피 결손에 올려 코발트 블루 빛 아래에서 시각화하여 형광 염색으로 괴사조직 제거 부위를 확인합니다.쥐 각막 찰과상 모델의 생체외 배양을 진행하여 안락사된 마우스의 상부 및 하부 안와 테두리를 부드럽게 눌러 안구를 밀어냅니다. 닫힌 각막 가위의 끝을 안구 벽을 따라 구후 공간에 삽입하여 안구를 관통하지 않도록합니다. 0.3mm 각막 겸자로 안구를 안정적으로 잡은 다음 각막 가위로 시신경과 안와 주위 연조직을 절단하여 안구를 분리합니다.쥐 안구의 생체 외 배양을 위해 우물 내부에 녹은 왁스가 있는 48웰 플레이트를 준비하고 응고를 기다린 다음 결막 겸자 끝으로 안구를 수용할 수 있도록 응고된 왁스 표면에 둥근 구멍을 만듭니다. 채취한 안구를 왁스로 덮인 바닥과 측벽이 있는 48웰 플레이트에 직접 놓아 안정화합니다. 연구 목적에 따라 항생제 유무에 관계없이 1% 소 태아 혈청을 함유한 DMEM으로 안구를 배양합니다.안구가 뜨지 않고 안구 표면을 배양액으로 담그고 플루오레세인 염색을 통해 상처 치유 과정을 기록하고 코발트 블루 라이트 아래에서 디지털 카메라로 사진을 수집합니다. 알칼리성 화상을 유발하려면 페트리 접시에 직경 8mm의 원형 여과지를 놓습니다. 점 적기를 사용하여 페트리 접시에 0.5 일반 수산화 나트륨을 첨가하여 여과지를 담그고 여과지에서 초과 용액을 배출 한 후 마취 된 토끼 각막에 놓습니다.검경 눈꺼풀로 눈꺼풀을 연 후 토끼 니티팅 멤브레인이 여과지 삽입을 방해하지 않는지 확인하고 알칼리에 적신 여과지를 중앙 각막에 30초 동안 놓습니다. 여과지를 제거하고 안구 표면을 10ml의 생리 식염수로 헹구어 알칼리 물질을 씻어냅니다. 각막 결손을 완성하려면 각막 녹 고리 제거제를 사용하여 불투명 부위 내의 각막 상피를 Bowman의 막까지 괴사제거합니다.코발트 블루광 아래에서 플루오레세인 염색으로 괴사조직 제거 부위를 확인하고 각막 겸자를 사용하여 잔류 각막 상피를 제거합니다. tarsorrhaphy로 상처 상태를 확보하기 위해 니티팅 멤브레인이 안구 표면과 코 쪽의 각막 상피 결손을 매끄럽게 덮고 있는지 확인합니다. 안구 표면을 보호하고 토끼가 긁는 것을 방지하기 위해 6-0 봉합사를 사용하여 국소 제제의 유무에 관계없이 일시적인 족근을 수행하고, tarsorrhaphy에 대한 봉합사가 토끼가 봉합사를 부러 뜨리는 것을 방지하기 위해 4-5 개의 넥타이와 더 긴 매듭으로 위아래 눈꺼풀 가장자리에서 3-4 밀리미터에 있는지 확인하십시오.마우스 각막 상피의 생체외 상처 치유 모델에서, 마우스 각막 상피의 생체 내 괴사조직 제거 후 중앙 2 밀리미터에서 양성 플루오레세인 염색을 갖는 약간 함몰된 중앙 각막 영역이 관찰되었다. 왁스로 코팅된 48웰 배양 플레이트에 고정된 쥐 안구의 생체 외 배양을 검사하고 실체 현미경으로 48웰 배양 플레이트 내에서 매일 기록했습니다. 쥐 각막 상피를 괴사제거한 지 하루 후, 직경 2mm의 원형 플루오레세인으로 염색된 상피 결손 하나가 코발트 블루광 하에서 얻은 디지털 사진에서 밝혀졌습니다.토끼 각막 상피에 알칼리 손상을 일으킨 후, 선명하고 완전한 원형 마진을 갖는 중앙 각막에 코발트 블루 광을 포함하거나 포함하지 않고 양성 플루오레세인 염색이 관찰되었습니다. 윤부에서 pannus ingrowth를 동반한 각막 상피 상처 재상피화가 관찰되었습니다. 가장 중요한 단계는 마우스와 토끼 각막 표면에 매끄럽고 균일 한 상피 결함을 만드는 절차입니다.이러한 플랫폼에서 새로운 의료 및 외과 치료제를 테스트할 수 있습니다. 재 상피화의 효과는 절차 후에 모니터링 할 수 있습니다. 이 프로토콜에서 손상 후 각막 신생혈관 형성 및 혼탁은 추가 연구에 대한 충족되지 않은 요구가 될 것입니다.
