Toenemend onderzoek wijst uit dat interacties tussen plant en bacteriën voornamelijk in de rhizosfeer voorkomen en worden beïnvloed door wortelexsudaten20,21,22,23,24. Plantenwortelexsudaten omvatten een breed scala aan primaire metabolieten, waaronder fenolzuren, organische zuren en aminozuren, evenals complexere secundaire verbindingen25,26,27.<sup class="...

Chemotaxis is belangrijk voor de kolonisatie van plantengroeibevorderende rhizobacteriële (PGPR) op wortels en voor het begrijpen van plant-microbe interacties1. Een klasse van verbindingen met een laag molecuulgewicht (chemoattractanten) in plantenwortelexsudaten induceert de chemotactische beweging van PGPR naar de rhizosfeer2. Appelzuur, citroenzuur en andere componenten in de wortelexsudaten stimuleren chemotaxis van Bacillus-stammen3. Glucose, citroenzuur en fumaarzuur in maïswortelexsudaten rekruteren bijvoorbeeld bacteriën naar het worteloppervlak4. D-galactose, dat is afgeleid van wortelexsudaten, induceert de chemotaxis van Bacillus velezensis SQR95. Organische zuren, waaronder fumaraat, appelzuur en succinaat, beïnvloeden chemotaxis en kolonisatie van verschillende PGPR in het Cajanus cajan - Zea mays intercropping-systeem6. Oleanolic acid in rijstwortel exsudaten, werkt als een chemoattractant voor de stam FP357. Andere plantenexsudaten (waaronder histidine, arginine en aspartaat) kunnen een cruciale rol spelen in de chemotactische respons van bacteriën8. Plantenexsudaten functioneren als een signaal om de beweging van bacteriën te sturen, wat de eerste stap is tijdens de kolonisatie van de rhizosfeer. Plantenkolonisatie door PGPR is een proces van enorme relevantie, omdat PGPR gunstig is voor de plantgastheer.
Veel methoden zijn gebruikt voor het analyseren van bacteriële chemotaxis. De zwemplaatmethode is een van de eerder beschreven methoden9. Bij deze methode werden platen gemaakt met een halfvast medium. Een chemotactische buffer met agar (1,0%, w/v) werd aan de plaat toegevoegd. De buffer wordt verwarmd en vervolgens gemengd met het chemoattractant. Vervolgens werd 8 μL bacteriesuspensie druppelsgewijs aan het midden van de plaat toegevoegd en werd de plaat bij 28 °C in een incubator geplaatst. De plaat werd regelmatig geobserveerd en gefotografeerd. De experimentele cyclus van de zwemplaatmethode was echter erg lang. Bij de capillairachtige methode10 dient een pipetpunt als kamer voor het vasthouden van 100 μL bacteriële suspensie. 1 ml spuitnaald werd gebruikt als capillair. Een spuitnaald met chemoattractanten met verschillende concentratiegradiënten werd in de pipetpunt van 100 μL ingebracht. Na incubatie bij kamertemperatuur gedurende 3 uur werd de naald van de spuit verwijderd, de inhoud werd verdund en op het medium verguld. De bacteriële accumulatie in de spuit werd weergegeven door kolonievormende eenheden (CFU's) in de platen. De experimentele fout binnen replicaties voor de capillair-achtige methode was echter groot. Een andere methode gebruikte een microfluïdisch SlipChip-apparaat11. Kortom, runderserumalbumine (BSA) oplossing werd in alle kanalen geïnjecteerd en verwijderd met behulp van een vacuüm. De oplossingen die verschillende chemoattractanten bevatten (1 mM concentratie alleen voor kwalitatieve detectie), bacteriële cellen gesuspendeerd in fosfaat-gebufferde zoutoplossing en fosfaat gebufferde zoutoplossingbuffer (negatieve controle) werden toegevoegd aan respectievelijk de bovenste, middelste en onderste microwells. Incubatie werd vervolgens uitgevoerd in een donkere omgeving bij kamertemperatuur gedurende 30 minuten. De bacteriële cellen werden vervolgens gedetecteerd in de microwells. Het microfluïdische SlipChip-apparaat was echter duur. Daarom had elk van de hierboven beschreven methoden voor- en nadelen.
We hebben een verbeterde chemotaxis-test vastgesteld voor de snelle identificatie van rhizobacteriële chemoattractanten in wortelexsudaten met behulp van steriele glasglaasjes zonder ingewikkelde stappen. In deze studie, door middel van meerdere vergelijkingen van experimentele resultaten, overwon de methode meerdere tekortkomingen van traditionele bacteriële chemotaxismethoden. De methode verminderde de fout van het tellen van platen en verkortte de experimentele cyclus. Daarom kan deze nieuwe methode, indien gebruikt om een chemoattractante stof te identificeren, 2-3 dagen besparen en de kosten van experimentele materialen verlagen.

an improved chemotaxis assay for the rapid identification of rhizobacterial chemoattractants in root exudates

Chemotaxis identificatie is erg belangrijk voor het onderzoek en de toepassing van rhizosfeer groeibevorderende bacteriën. We hebben een eenvoudige methode ontwikkeld om snel de chemoattractanten te identificeren die de chemotactische beweging van rhizosfeergroeibevorderende bacteriën op steriele glazen dia's via eenvoudige stappen kunnen induceren. Bacterieoplossing (OD600 = 0,5) en steriele chemoattractante waterige oplossing werden druppelsgewijs op de glasplaat toegevoegd met een interval van 1 cm. Een inentingslus werd gebruikt om de chemoattractante waterige oplossing met de bacteriële oplossing te verbinden. De glijbaan werd 20 minuten op kamertemperatuur gehouden op de schone bank. Ten slotte werd de chemoattractante waterige oplossing verzameld voor bacterieel tellen en microscopische observatie. In deze studie, door middel van meerdere vergelijkingen van experimentele resultaten, overwon de methode meerdere tekortkomingen van traditionele bacteriële chemotaxismethoden. De methode verminderde de fout van het tellen van platen en verkortte de experimentele cyclus. Voor de identificatie van chemoattractante stoffen kan deze nieuwe methode 2-3 dagen besparen in vergelijking met de traditionele methode. Bovendien stelt deze methode elke onderzoeker in staat om systematisch een bacterieel chemotaxis-experiment binnen 1-2 dagen te voltooien. Het protocol kan worden beschouwd als een waardevolle bron voor het begrijpen van plant-microbe interacties.

In totaal werden 584 en 937 bekende metabolieten gedetecteerd in respectievelijk de positieve en negatieve ionenindices. Eerdere studies hebben aangetoond dat chemoattractanten meestal organische zuren, aminozuren en koolhydraten zijn17,18.

Het artikel rapporteerde identificatie van chemoattractant van rhizobacteriën in wortelexsudaten met behulp van een verbeterde chemotaxis-test. Chemotaxis implantaat wortel actuaten is belangrijk voor wortelkolonisatie voor plantengroei, het bevorderen van rhizobia bacteriën en voor het verstrekken van de nuttige facties van plant-microbe interacties. Veel methoden werden gebruikt voor het analyseren van bacteriële chemotaxis.Bijvoorbeeld zwemplaatmethode, Cappilary-achtige methode, microfluïdisch slipchipapparaat. De experimentele cyclus van de zwemplaatmethode was lang. De experimentele fout van Cappilary-achtige methode werd gelanceerd, microfluïdisch slipchipapparaat is duur om te experimenteren.We hebben een eenvoudige methode ontwikkeld om snel het chemoattractant te identificeren dat de chemotactische beweging van rhizosfeergroeibevorderende bacteriën zou kunnen induceren met behulp van steriele glasglaasjes zonder ingewikkelde stappen. De methode verminderde de fout van het tellen van de platen en verkortte de experimentcyclus. Met betrekking tot het identificeren van een chemoattractante stof, kan de nieuwe methode 2-3 dagen besparen en ook de kosten van experimentele materialen verlagen.Willekeurig verdelen rijstzaden in de kweekkamer ontstaat gedurende een week en voeg twee keer steriel water toe. Selecteer de rijstzaailingen van vergelijkbare grootte en plant in een 50 milliliter Ms vloeibaar medium. Incubeer gedurende 48 uur bij 22 graden Celsius en een septische conditie.Rijstwortelexsudaten worden vrijgegeven in het Ms-medium. Vloeistofchromatografie massaspectrometrie analyse werd uitgevoerd met behulp van UHPLC-systeem. Bereid chemoattractant waterige oplossing zorg ervoor dat deze steriel is.Filter de chemoattractieve oplossing met een bacteriefilter van 0,22 micrometer. De chemoattractante waterige oplossing was de enige stof die uit de LCMS-onderzoeken werd verkregen, opgelost in water. Citrix acid solution werd als voorbeeld gebruikt.Alle acties moeten aan de zijkant van de lamp worden uitgevoerd. Markeer de middelste positie van de glasplaat met een interval van 1 centimeter. Zorg ervoor dat de glasplaat meerdere keren op de vlam wordt gesteriliseerd.Bij de 30 microliter chemoattractante oplossing links van de glasplaat zorgen ervoor dat bacteriën tot in het logaritmische stadium worden gekweekt. Voeg de 30 microliter bacterieoplossing rechts van de glasplaat toe. Steriliseer de entingslus meerdere keren op de vlam met behulp van de entlus om de chemoattractante waterige oplossing met de bacterieoplossing te verbinden en houd deze gedurende 20 minuten op kamertemperatuur op een schone bank.Koppel na 20 minuten de verbindingsleiding los met filterpapier. Zorg ervoor dat de 1,5 milliliter gecentrifugeerde buis steriel is. Verzamel de chemoattractante waterige oplossing aan de linkerkant van de glasplaat.Breng de oplossing over in de steriele gecentrifugeerde buis van 1,5 milliliter. Voeg het juiste volume van de voorkleurstof van de cel toe aan de gecentrifugeerde buis. Verzamel na 2 minuten de toegestane vloeistoffen voor het tellen van bacteriën en microscopische observatie met een bloedtelkamer.Bepaal het aantal van de aangetrokken levensvatbare micro-organismen met behulp van een volgende vergelijking. Een hogere RCI duidt op een sterke reactie op het chemoattractant. De resultaten gaven aan dat citroenzuur en cafeïnezuur de hoogste chemotaxis-indices hebben.De resultaten van de nieuwe chemotaxis-testmethode schreven dat bacteriële canconcentratie van citroenzuur, cafeïnezuur en galactose-experimenten significant hoger was dan die van de controlegroep en positieve controle. Citroenzuur schreef de sterkste chemotaxis. De experimentele resultaten waren in overeenstemming met de resultaten van traditionele methoden.Dit videoprotocol kan worden gepresenteerd als een waardevolle bron voor verduidelijking van plant-microbe interacties.
