Controlled Semi-Automated Laser-Induced Injuries for Studying Spinal Cord Regeneration in Zebrafish Larvae

ここで提案する技術は、経験の浅いオペレータであっても、隣接する組織への損傷を軽減し、再現性を向上させることによって、手動病変プロトコルの限界を克服することを可能にする。この技術により、回転するガラス毛細血管と標的UVレーザーを組み合わせることで、周囲の組織を損傷することなく病変を誘導する場所を正確に選択することができます。VAST機器の操作には多くのステップが関係しているので、すべてが正しく行われるように、すべてのステップをチェックすることをお勧めします。麻酔薬を含む培地を使用して幼虫を麻酔することから始めて、次いでそれらを1ウェルあたり300マイクロリットルの培地を含む96ウェルプレートに移す。アブレーション用のレーザーを含むすべてのシステムコンポーネントの電源を入れます。次に、自動ゼブラフィッシュイメージングまたはVASTソフトウェアを起動し、最初のウィンドウで[プレート]を選択し、[完了]ボタンをクリックします。毛細血管が空で清潔であるかどうかを尋ねる小さな窓がポップアップ表示されます。毛細血管の画像に内部に気泡がないか確認してください。内部に気泡がない場合は、ポップアップウィンドウの[はい]をクリックします。次に LP サンプラーウィンドウで、[ファイル] メニューに移動し、[スクリプトを開く] オプションを選択します。実行する実験に対応するスクリプトを含むファイルを選択します。次に、VAST ソフトウェアのメイン ウィンドウで、[ファイル] に移動し、[実験を開く] を選択します。計画した実験に対応する実験ファイルを選択します。ImageJ/Fijiソフトウェアを起動し、[ファイル]メニューに移動し、[新しいスクリプト]を選択してスクリプトウィンドウを開きます。次に、[ファイル]メニューに移動し、[開く]を選択してレーザー病変スクリプトをロードします。次に、Python IDE を起動するには、[ファイル] メニューに移動し、[ファイルを開く] を選択して、レーザーを管理するスクリプトを読み込みます。次に、[実行]メニューをクリックし、[デバッグなしで実行]を選択してスクリプトを実行します。レーザー・アッテネータの初期化中に、何らかのノイズとともに端末パネルに一連のメッセージが表示されることを確認します。VAST ソフトウェアのメイン ウィンドウで、矢印ボタンをクリックしてステージを移動し、顕微鏡の対物レンズに対してキャピラリーを中央に配置します。接眼レンズを覗き込み、顕微鏡の透過光を使って毛細血管の上部に焦点を合わせます。LPサンプラーの左プレートホルダー上に幼虫を含む96ウェルプレートを置く。次に、サンプラーの右側のプレートホルダーに回収用の別のプレートを置きます。プレートの A1 ウェルがホルダーの左前隅にあることを確認します。VAST ソフトウェアの LP サンプラー ウィンドウで、[プレート テンプレート] ボタンをクリックし、幼虫を含むすべてのウェルを選択します。[OK]ボタンをクリックして検証し、ウィンドウを閉じます。次に、LPサンプラーウィンドウの[プレートの実行]ボタンをクリックして、幼虫のロードを開始します。次に、顕微鏡ソフトウェアに移動し、[ライブ]ボタンをクリックして幼虫を画像化します。脊髄中央管が見えるまで顕微鏡のフォーカスノブをオンにします。蛍光のスナップショットを撮り、画像をフォルダに保存します。ImageJで画像を開き、必要に応じてコントラストを調整します。関心領域線ツールをクリックし、脊髄を中心とする短い線を描きます。顕微鏡を100%反射ミラーの位置に切り替えます。ImageJ スクリプトをロードし、繰り返しを 2、サンプルを 1、幅を 40 ミクロン、減衰を 89 に設定します。すべてのパラメータを設定したら、[OK]ボタンをクリックします。レーザーショットシーケンスが終了したら、イメージングソフトウェアで蛍光イメージングに切り替え、フォーカスを調整します。新しいスナップショットを作成して保存します。この新しい画像をImageJで開き、脊髄自体よりも大きな新しい線を描きます。顕微鏡を100%反射ミラーの位置に切り替えます。ImageJ スクリプトウィンドウに移動し、「繰り返し」を 2、サンプルを 1、幅を 40 ミクロン、減衰を 89 に設定します。すべてのパラメータを設定したら、[OK]ボタンをクリックします。レーザーショットシーケンスが終了したら、蛍光をイメージングして焦点を合わせることで、切断品質を確認します。病変部位に細胞または軸索が無傷のまま残っていないことを確認します。メインの VAST ソフトウェア ウィンドウに移動し、[収集] ボタンをクリックして、病変した幼虫を空の 96 ウェル プレートに集めます。次に、チェックボックストレイライトをクリックして、VASTシステムライトを再びオンにします。できるだけ早く96ウェルプレートから幼虫を取り出し、幼虫が病変後を回復するために新鮮な魚の水を入れたきれいなペトリ皿に移す。ペトリ皿を摂氏28度のインキュベーターに入れます。アセチル化チューブリン免疫染色およびカルシウムイメージングは、レーザー病変が脊髄組織の連続性を完全に破壊することを示している。無傷の脊髄がこの画像に示されている。病変の尾側と吻側との間の軸索の完全な破壊は、脊髄の完全な切断を確認する。不完全な断面の例を以下に示します。NBTG cAMP 6-S幼虫上のトランスセクト脊髄をこの画像に示す。長方形は、病変の吻側および尾側における蛍光強度を定量化するために使用されるROIを示す。グラフィカル画像は、吻側および尾側分析ROIにおける経時的な蛍光強度変化を表す。レーザー病変の3時間後、24時間後、および48時間後のNBT:dsRed幼虫の最大強度投影蛍光画像をここに示す。損傷後24時間後、創傷は閉じ始め、48時間後に脊髄の初期構造の部分的な回復をもたらした。部分的な機能的再接続は、カルシウムイメージングを用いて傷害後48時間後に確認された。尾部領域と吻側領域におけるスパイクの振幅の比は、損傷後3、24、および48時間の間に増加を示した。マクロファージリクルートメントは、NBT:dsRedmpeg1 GFP幼虫レーザー病変を用いてレーザー病変後に観察された。手動病変とレーザー病変の間で標識細胞数に差は認められなかった。病変していない魚は、両方の病変条件下で病変した魚よりも少ない二重標識細胞を示した。レーザー病変は、手動病変よりも少ない筋肉および皮膚損傷を誘発する。病変が完全であるかどうかを確認することが重要です。無傷の細胞または構造は病変領域に見えず、薄暗い背景のみに見えるはずである。脊髄再生を研究するために、免疫組織化学やカルシウムイメージングなど、多くの方法を使用しています。重要なことに、組織が手動で病変した動物とは対照的に、解剖に耐えることができるので、電気生理学を行うこともできます。この新しい技術により、損傷後の脊髄の構造的および機能的な組織を定量的に研究し、再現性および制御された病変を可能にすることができます。
