De grootschalige productie van oplosbare, menselijke, recombinante MMP's blijft een uitdagende taak. Zoogdiercellen kunnen functionele MMP's tot expressie brengen tegen hoge kosten en lange wachttijden, terwijl E. coli snel grote hoeveelheden MMP-inclusielichamen produceert die moeten worden gezuiverd en opnieuw moeten worden gevouwen11,16. R2DP-cellen verhogen de opbrengst van MMP-inclusielichamen aanzienlijk, waardoor een kosteneffectiever en productie...

De meeste complexe eukaryote eiwitten ondergaan uitgebreide posttranslationele modificaties na expressie, waarbij zeer geassisteerde eiwitvouwing en co-factoren functioneel moeten zijn1. Het produceren van grote hoeveelheden oplosbaar menselijk eiwit in een bacteriële gastheer blijft een aanzienlijke uitdaging vanwege de hoge kosten en het gebrek aan robuuste expressie- en zuiveringsmethoden, zelfs voor kleinschaligere laboratoriumexperimenten2,3. MMP's, menselijke endopeptidasen met een groot molecuulgewicht, worden gewoonlijk uitgedrukt als onoplosbare inclusielichamen wanneer ze worden uitgedrukt in E. coli. Extractie van oplosbare menselijke MMP's leidt vaak tot een moeizaam, tijdrovend oplos- en hervouwproces4.
MMP's spelen een cruciale rol in zowel fysiologische als pathogene processen. Menselijke MMP's zijn een familie van 23 zink endopeptidasen, gecategoriseerd naar structuur en substraatspecificiteit, en differentieel uitgedrukt ondanks een sterk geconserveerd katalytisch domein5,6. MMP's worden uitgescheiden als inactieve zymogenen, gereguleerd via posttranslationele activering en hun endogene remmers, weefselremmers van metalloproteinasen (TIMPs)7,8,9,10. Hoewel aanvankelijk erkend voor hun rol in ecm-omzet, zijn MMP's ook betrokken geweest bij ontwikkeling, morfogenese, weefselherstel en remodellering8. Ontregeling van MMP's is met name in verband gebracht met kanker, samen met neurodegeneratieve, cardiovasculaire en fibrotische ziekten, naast andere ziekten5,7.
De ontwikkeling van robuuste grootschalige MMP-productiemethoden is van cruciaal belang om het succes van toekomstige studies van MMP-mechanismen door middel van biochemische en celgebaseerde assays te garanderen. Verschillende MMP's zijn eerder tot expressie gebracht in bacteriën11, waaronder Hisx6-gelabelde MMP's, zonder de MMP-activiteit12,13,14,15 te veranderen. Deze methoden omvatten echter vervelende, lange stappen die moeilijk te repliceren kunnen zijn.
Zoogdiercellen kunnen ook worden gebruikt om veel verschillende menselijke eiwitten tot expressie te brengen en tegelijkertijd de juiste posttranslationele modificaties te garanderen16. Hoewel het expressiesysteem van zoogdieren een ideale keuze is om recombinante menselijke eiwitten te produceren met de juiste posttranslationele modificaties, zijn de belangrijkste nadelen van deze methode aanvankelijk lage opbrengsten, dure groeimedia en reagentia, lange tijdlijnen om stabiele expressielijnen te bereiken en het risico op besmetting met andere soorten zoals schimmels of bacteriën2,11 . Bovendien levert MMP-productie in cellijnen van zoogdieren onzuiverheden op van geassocieerde cellulaire eiwitten zoals TIMPs of fibronectines11. In tegenstelling tot de langzame celgroei die wordt waargenomen in zoogdiercellen, biedt het bacteriële expressiesysteem grootschalige eiwitproductie in een korte periode, samen met eenvoudigere media en groeivereisten. Vanwege het gebrek aan andere geassocieerde cellulaire eiwitten (d.w.z. TIMPs) in bacteriële expressiesystemen, zijn actieve MMP's in hogere concentraties echter onderhevig aan afbraak door autoproteolyse, wat resulteert in een slechte MMP-opbrengst17.
Dit artikel beschrijft een gedetailleerde methode voor bacteriële expressie, zuivering en activering van recombinant Hisx6-pro-MMP-3cd met behulp van E. coli als expressiegastheer vanwege de betaalbaarheid, eenvoud en het succes bij het produceren van hogere opbrengsten van MMP's2,3,18. Aangezien E. coli de eiwitvouwingsmachines en posttranslationele verwerking mist die nodig zijn voor recombinante MMP's en andere complexe eiwitten, zijn veel E. coli-stammen ontworpen om deze beperkingen te overwinnen, waardoor E. coli een geschiktere gastheer is voor expressie van recombinant humaan MMP-3cd,19,20 . De R2DP-stam die in deze studie wordt gebruikt, verbetert bijvoorbeeld de eukaryote expressie door een chlooramfenicol-resistent plasmide te leveren dat codons bevat die zelden worden gebruikt in E. coli.
Zoals beschreven in dit protocol, worden hisx6-pro-MMP-3 katalytische domein (MMP-3cd) eiwitten geëxtraheerd en gedenatureerd4 na overexpressie van relatief zuivere inclusielichamen van de pET-3a-vector (figuur 1) in R2DP-cellen. Hisx6-pro-MMP-3cd3,19 werd gezuiverd met behulp van affiniteitstagchromatografie. Bij hervouwen en dialyse werd de pro-MMP-3cd (zymogeen) geactiveerd door 4-aminofenylkwikacetaat (APMA) en SDS-PAGE-analyse wordt gebruikt om de opbrengsten en de noodzaak van verdere zuivering te evalueren5,21. Dit protocol beschrijft expressie, zuivering en activering van oplosbare MMP-3cd als voorbeeld. Het kan echter ook worden gebruikt als leidraad voor de expressie van andere MMP's en menselijke proteasen met vergelijkbare expressie en activeringsmechanismen (figuur 2). Voor andere eiwitten dan MMP-3cd wordt de lezer geadviseerd om optimale buffersamenstellingen en methoden voor hun doeleiwit te bepalen voordat dit protocol wordt geprobeerd.

bacterial expression and purification of human matrix metalloproteinase-3 using affinity chromatography

Matrix metalloproteinases (MMP's) behoren tot de familie van metzincineproteasen met centrale rollen in extracellulaire matrix (ECM) afbraak en remodellering, evenals interacties met verschillende groeifactoren en cytokines. Overexpressie van specifieke MMP's is verantwoordelijk voor verschillende ziekten zoals kanker, neurodegeneratieve ziekten en hart- en vaatziekten. MMP's zijn onlangs het middelpunt van de aandacht geweest als doelwitten om therapeutica te ontwikkelen die ziekten kunnen behandelen die gecorreleerd zijn aan MMP-overexpressie.
Om het MMP-mechanisme in oplossing te bestuderen, zijn meer gemakkelijke en robuuste recombinante eiwitexpressie- en zuiveringsmethoden nodig voor de productie van actieve, oplosbare MMP's. Het katalytische domein van de meeste MMP's kan echter niet worden uitgedrukt in Escherichia coli (E. coli) in oplosbare vorm vanwege een gebrek aan posttranslationele machines, terwijl zoogdierexpressiesystemen meestal duur zijn en lagere opbrengsten hebben. MMP-inclusielichamen moeten het vervelende en moeizame proces van uitgebreide zuivering en hervouwen ondergaan, waardoor de opbrengst van MMP's in inheemse conformatie aanzienlijk wordt verminderd. Dit artikel presenteert een protocol met behulp van Rosetta2(DE3)pLysS (hierna R2DP) cellen om matrix metalloproteinase-3 katalytisch domein (MMP-3cd) te produceren, dat een N-terminale His-tag bevat gevolgd door pro-domein (Hisx6-pro-MMP-3cd) voor gebruik in affiniteitszuivering. R2DP-cellen verbeteren de expressie van eukaryote eiwitten door middel van een chlooramfenicol-resistent plasmide met codons die normaal zeldzaam zijn in bacteriële expressiesystemen. Vergeleken met de traditionele cellijn bij uitstek voor recombinante eiwitexpressie, BL21 (DE3), verbeterde zuivering met behulp van deze nieuwe stam de opbrengst van gezuiverde Hisx6-pro-MMP-3cd. Na activering en ontzouting wordt het pro-domein samen met de N-terminal His-tag gesplitst, waardoor actieve MMP-3cd wordt geleverd voor onmiddellijk gebruik in talloze in vitro toepassingen. Deze methode vereist geen dure apparatuur of complexe fusie-eiwitten en beschrijft de snelle productie van recombinante menselijke MMP's in bacteriën.

Bij het uitvoeren van monsters op SDS-PAGE, omdat het eiwit tot expressie wordt gebracht in de vorm van onoplosbare inclusielichamen, moeten de gelyseerde en gesonificeerde fracties weinig tot geen Hisx6-pro-MMP-3cd-extract bevatten, omdat het eiwit nog niet is opgelost in ureum. Figuur 3 vergelijkt de His-tag zuiveringsemissiefracties van Hisx6-pro-MMP-3cd van BL21(DE3)-cellen en R2DP-cellen. Dit gedetailleerde protocol beschrijft een zeer efficiënte en kosteneffectieve methode van bacteriële expressie voor recombinant MMP-3 katalytisch domein in E.Coli, die kan worden toegepast op andere MMP therapeutische doelen. E.coli mist een complex eiwitvouwings- en posttranslationele verwerkingssysteem. Deze methode maakt gebruik van de R2DP-celstam om de expressie van eukaryote eiwitten in E.coli te verbeteren.Overproductie van specifieke MMP's leidt tot verschillende ziekten zoals kanker, cardiovasculaire en neurodegeneratieve ziekten. Actieve oplosbare MMP's zijn nodig voor de ontwikkeling van MMP-remmende therapieën. Ent een enkele geïsoleerde kolonie van pET His-tag pro MMP-3 katalytische domeintransformatiemiddel uit een LB-ampicilline chlooramfenicolresistente plaat in vijf milliliter LB-ampicilline chlooramfenicolresistente media bij 37 graden Celsius.Bewaar aliquots van elke cultuur en bereid 40% glycerol aandelen. Ent een kolf van één liter met 500 milliliter LB ampicilline chlooramfenicolbestendig medium tot een optische dichtheid van 0,05 tot 0,1 bij 600 nanometer met de nachtcultuur. Meet de optische dichtheid op 600 nanometer op verschillende tijdstippen, meestal gedurende drie tot vier uur totdat deze tussen 0,4 en 0,6 valt.Induceer de culturen met één molaire IPTG-stam tot een eindconcentratie van één millimolar. Incubeer nog eens drie tot vier uur in de shaker van 37 graden. Centrifugeer de cellen in conische flessen van 250 milliliter bij 13.000 G en vier graden Celsius gedurende 10 minuten en herhaal totdat de culturen volledig zijn gepelletiseerd.Voeg drie milliliter lysisbuffer per gram pellet toe en reuspend de pellet door vortexing of pipetteren. Voeg 1,25 milliliter 10%volumegewicht natriumdeoxycholaat per liter cultuur toe. Voeg 10 microliter DNase I toe per liter cultuur.Schud bij kamertemperatuur gedurende 30 minuten bij 150 RPM. Centrifugeer gedurende 10 minuten bij 13.000 G en vier graden Celsius. Gooi het supernatant weg.Resuspend de pellet in 100 milliliter per liter cultuur van inclusielichaambuffer door op en neer te pipetteren. Houd de monsters op ijs tijdens ultrasoonapparaat om oververhitting te voorkomen. Soniceer elk monster gedurende zes cycli van 15 seconden, een output van vijf en een 50% puls.Centrifugeer de monsters gedurende 10 minuten bij 13.000 G en vier graden Celsius. Controleer de pellet. Als de pellet compact is, gooit u het supernatant weg.Resuspend elke pellet uit een cultuur van één liter en vijf milliliter oplosbuffer. Incubeer gedurende ten minste 30 minuten op ijs om de eiwitten te laten oplossen. Centrifugeer de cellen gedurende 10 minuten bij 13.000 keer G in vier graden Celsius.Als de pellet zich na centrifugeren vormt, giet het supernatant dan in een aparte conische buis van 50 milliliter. Resuspend de pellet in nog eens vijf milliliter oplosbuffer per liter cultuur door op en neer te pipetteren. Centrifugeer gedurende 10 minuten bij 13.000 G en vier graden Celsius.Trek vervolgens de supernatanten. Gooi de pellet weg of bewaar deze bij min 80 graden Celsius voor extra ultrasoonapparaat. Maak een oogje in het zeil tegen de HT-wasbuffer en noteer de A280 van de eerste wasfractie.Herhaal dit met andere breuken totdat A280 de basislijn nadert. Eluteer onmiddellijk His-tagged eiwitten door vijf milliliter HT-elutiebuffer toe te voegen. Verzamel de doorstroom bij 0,5 tot één milliliterfractie in microfugebuizen met het label HT-elutie.Meet de A280 van de elutiedractie. Als de A280 groter is dan 0,3 milligram per milliliter, verdunt u de fractie met HT-equilibratiebuffer. Dompel de geëlueerde MMP-fracties onder in dialysebuizen in een liter dialysebuffer één en roer de slang en de inhoud ervan gedurende ten minste acht uur op een magnetische roer bij vier graden Celsius.Breng over op één liter dialysebuffer twee en roer de slang en de inhoud ervan op een magnetische roer gedurende ten minste acht uur bij vier graden Celsius. Breng vervolgens over naar één liter dialysebuffer drie en roer de slang en de inhoud ervan op een magnetische roer gedurende maar liefst acht uur bij vier graden Celsius. Breng het monster over in een geschikte container en label ze als gebeld MMP.Onderzoek de buis op neerslag. Voeg per één milliliter aliquot van één milligram per milliliter MMP 50 microliter van 20 millimolaire APMA toe om een uiteindelijke APMA-concentratie van één millimolar te bereiken. Als zich een neerslag vormt, centrifugeer het dan op maximale snelheid gedurende 10 minuten bij vier graden Celsius.Bewaar het supernatant in een microfugebuis van 1,5 milliliter met het label geactiveerde MMP. Gooi het neerslag weg in een container die is gemarkeerd voor APMA-afval. SDS-PAGE werd uitgevoerd om de His-tag zuiveringsemissiefracties van His-tag pro MMP-3 katalytisch domein van BL21 DE3-cellen in R2DP-cellen te vergelijken.De eerste acht elutiefracties van het His-tagged MMP katalytische domein en BL21 DE3 cellen vertoonden minder eiwit dan die in het His-tagged MMP katalytische domein in R2DP cellen. De geïnduceerde, opnieuw gevouwen, geconcentreerde, geactiveerde en ontzoute fracties van MMP3 CD- en R2DP-cellen worden getoond. Bij activering is het molecuulgewicht van de geactiveerde MMP-3 CD-band ongeveer 20 kilodalton, in tegenstelling tot het His-tagged zymogeen dat op ongeveer 30 kilodalton blijft.Wanneer het centrifugeren van cellen lyseert, zijn pellets mogelijk niet compact. Wanneer dat gebeurt, soniceer dan meer en centrifugeer langer. Bij het reconcentreren van het eiwit kan neerslag optreden.Los het op in oplosbuffer en herhaal het proces. De hier beschreven procedure biedt een gedetailleerde methode voor oplosbare expressie van actieve menselijke MMP's die uiteindelijk kan worden gebruikt om het moleculaire mechanisme van MMP-enzymatische activiteit te begrijpen. Alternatieve eiwitzuiveringsmethoden zoals FPLC kunnen worden uitgevoerd op gezuiverde MMP-eiwitten.
