생산 단계에서 유전자 발현을 유도하기 위해 어둠 대신 빛을 사용하는 것은 광유전학적 조절의 능력을 크게 확장시킬 것이지만, 또한 발효의 이 단계에서 전형적으로 직면하는 높은 세포 밀도의 광 침투 한계를 극복할 필요가 있을 것이다. 이를 위해 우리는 청색광 자극에 대한 전사 반응을 증폭시키는 OptoAMP 및 OptoQ-AMP로 알려진 회로를 개발했습니다. 이들 회로는 VP16-EL222의 야생형 또는 과민성 돌연변이체를 사용하여 각각 GAL 또는 Q 레귤론의 전사 활성화제 Gal4p 또는 QF2의 생산을 조절하여, light12,13으로 향상된 감도와 더 강한 유전자 발현을 달성한다(도 1C). 광유전학 회로는 또한 대장균에서 화학 및 단백질 생산의 동적 제어를 위해 개발되었다. OptoLAC 회로는 YF1/FixJ 두 구성 요소 시스템6을 기반으로 하는 광 반응 pDawn 회로를 사용하여 박테리아 LacI 리프레저를 제어합니다(그림 3). OptoINVRT5와 유사하게, OptoLAC 회로는 빛에서 유전자 발현을 억제하고 어둠 속에서 유도하도록 설계되었습니다.
