Dit protocol beschrijft een eenvoudige en standaardmethode voor het isoleren van intacte PBS in twee soorten cyanobacteriën, eencellig model Syn6803 en filamenteuze niet-model JSC-1. De kritische stappen van het protocol zijn celhomogenisatie en ultracentrifugatie op een discontinue dichtheidsgradiënt van sucrose. Fycobilisoom (PBS) is een enorm in water oplosbaar pigmenteiwitcomplex dat zich hecht aan de cytoplasmatische kant van de fotosystemen in de thylakoïde membranen van cyanobacteriën1. PBS bestaat voornamelijk uit gekleurde fycobiliproteïnen en kleurloze linkereiwitten1,2. De fycobiliproteïnen kunnen worden onderverdeeld in vier hoofdgroepen: phycoerythrin, phycoerythrocyanine, phycocyanine en allophycocyanin3. De vier belangrijkste groepen absorberen verschillende golflengten van lichtenergie in het bereik van 490-650 nm, die chlorofylen inefficiënt absorbeerden3. De PBS kan dienen als een lichtoogstantenne voor het verzamelen van lichtenergie en deze te leveren aan Photosystem II en I4.
De structuur en samenstelling van PBS verschillen van soort tot soort. Gezamenlijk zijn drie vormen van fycobilisoom (hemidiscodiaal, bundelvormig en staafvormig) geïdentificeerd in verschillende cyanobacteriële soorten5. Zelfs bij dezelfde soort verandert de samenstelling van PBS als reactie op de omgeving, zoals lichtkwaliteit en uitputting van voedingsstoffen6,7,8,9,10,11. Daarom is de experimentele procedure om PBS te isoleren van cyanobacteriën van groot belang geweest bij het bestuderen van PBS12. Gedurende meerdere decennia hebben veel verschillende protocollen PBS geïsoleerd en de structuur, samenstelling en functie ervan geanalyseerd6,7,8,12,13,14,15,16,17. De grote verscheidenheid aan methoden voor PBS-isolatie biedt inderdaad flexibiliteit bij het isoleren van het complex in verschillende soorten met verschillende reagentia en instrumenten. Het maakt het echter ook moeilijker om een geschikt protocol te kiezen voor wetenschappers die niet bekend zijn met cyanobacteriën en PBS. Daarom wordt in dit werk een gegeneraliseerd en eenvoudig protocol ontwikkeld voor diegenen die geïnteresseerd zijn in het starten van PBS-isolatie van cyanobacteriën.

Omdat PBS een in water oplosbaar eiwitcomplex is en gemakkelijk kan worden gedissocieerd, is een fosfaatbuffer met hoge ionische sterkte nodig om PBS tijdens extractie te stabiliseren18. Verschillende onderzoeksartikelen die methoden beschrijven voor de isolatie van PBS van cyanobacterium zijn in het verleden gepubliceerd. De meeste methoden vereisen een hoge concentratie fosfaatbuffer8,14,15,18,19. De procedures voor mechanische verstoring van de cellen variëren echter, zoals glasparels-ondersteunde extractie, ultrasoonapparaat20 en Franse pers6,8,14. Verschillende fycobiliproteïnen kunnen worden verkregen door precipitatie met ammoniumsulfaat20 en gezuiverd door HPLC21 of een chromatografische kolom22. Aan de andere kant kan intact PBS gemakkelijk worden geïsoleerd door sucrosedichtheidsgradiënt ultracentrifugatie6,8,15.
In dit protocol werden één modelcyanobacterie en één niet-model cyanobacterie gebruikt als materialen voor PBS-isolatie. Het gaat om model eencellige glucosetolerante Synechocystis sp. PCC 6803 (hierna Syn6803) en niet-model filamenteuze Leptolyngbya sp. JSC-1 (hierna JSC-1), respectievelijk7,23,24. Het protocol begint met verstoring van de eencellige en filamenteuze cyanobacteriën in een fosfaatbuffer met hoge ionische sterkte. Na lysis worden de supernatanten verzameld door centrifugeren en vervolgens behandeld met een niet-ionisch reinigingsmiddel (Triton X-100) om de in water oplosbare eiwitten uit de thylakoïde membranen op te lossen. De totale in water oplosbare eiwitten worden toegepast op een discontinue sucrosedichtheidsgradiënt om de PBS te fractioneren. De discontinue sucrosegradiënt in dit protocol bestaat uit vier sucrose-oplossingen en verdeelt de intacte PBS in de laagste fracties van sucroselaag25. De integriteit van PBS kan worden geanalyseerd door SDS-PAGE, zinkkleuring en 77K fluorescentiespectroscopie6,7,8,26. Deze methode is geschikt voor wetenschappers die intact PBS willen isoleren van cyanobacteriën en de spectrale, structurele en compositorische eigenschappen ervan willen bestuderen.
Er zijn verschillende voordelen van dit protocol. (1) Deze methode is gestandaardiseerd en kan worden gebruikt voor het isoleren van intact PBS van zowel eencellige als filamenteuze cyanobacteriën. De meeste artikelen beschrijven de methode die werd toegepast in één type cyanobacteriën4,7,8,12,13,14,16,18. (2) Deze methode wordt uitgevoerd bij kamertemperatuur, aangezien PBS dissocieert bij lage temperatuur19,27. (3) Deze methode beschrijft het gebruik van een kralenklopper om de cellen te verstoren; daarom is het goedkoper en veiliger dan hogedruk Franse pers en mogelijke gehoorschade door sonicator in andere methoden8,13,14,20. (4) Deze methode isoleert intact PBS door middel van ultracentrifugatie van sucrosegradiënt. Op deze manier kan intact PBS met verschillende maten en gedeeltelijk gedissocieerde PBS worden gescheiden op basis van sucroseconcentratie.

isolation and characterization of intact phycobilisome in cyanobacteria

In cyanobacteriën is fycobilisoom een vitaal antenne-eiwitcomplex dat licht oogst en energie overbrengt naar fotosysteem I en II voor fotochemie. Het bestuderen van de structuur en samenstelling van fycobilisoom is van groot belang voor wetenschappers omdat het de evolutie en divergentie van fotosynthese in cyanobacteriën onthult. Dit protocol biedt een gedetailleerde en geoptimaliseerde methode om cyanobacteriële cellen tegen lage kosten te breken door een kralenklopper efficiënt. Het intacte fycobilisoom kan vervolgens uit het celextract worden geïsoleerd door sucrose gradiënt ultracentrifugatie. Deze methode heeft aangetoond geschikt te zijn voor zowel model- als niet-modelcyanobacteriën met verschillende celtypen. Een stapsgewijze procedure wordt ook aangeboden om de integriteit en eigenschap van fycobiliproteïnen te bevestigen door 77K fluorescentiespectroscopie en SDS-PAGE gekleurd door zinksulfaat en Coomassie Blue. Het geïsoleerde fycobilisoom kan ook worden onderworpen aan verdere structurele en compositorische analyses. Over het algemeen biedt dit protocol een nuttige startgids waarmee onderzoekers die niet bekend zijn met cyanobacteriën snel intact fycobilisoom kunnen isoleren en karakteriseren.

De Syn6803- en JSC-1-cellen werden gekweekt in kegelkolven met constant roeren in B-HEPES-medium bij 30 °C, onder een LED-wit licht (50 μmol fotonen m-2s-1) in een groeikamer gevuld met 1% (v/v) CO2. Het is een eenvoudig en snel protocol waarmee mensen die niet bekend zijn met cyanobacteriën op hun gemak fycobilisoom kunnen isoleren. Het voordeel van deze techniek is dat deze is aangepast aan een kleine schaal en het is gemakkelijk uit te voeren. Het is een eenvoudige, betrouwbare en efficiënte methode voor het isoleren van intacte fycobilisomen van model- en niet-modelcynobacteriën.Begin met het overbrengen van de celcultuur van 0,8 OD bij 750 nanometer in een kolf van één liter en verdun deze in 500 milliliter B-HEPES-medium om de OD van 0,2 bij 750 nanometer te bereiken. Kweek de cultuur met constant roeren bij 30 graden Celsius totdat de OD 0,8 tot 1 bereikt, wat wijst op een late exponentiële groeifase. Centrifugeer de cultuur met een snelheid van 10.000 maal g gedurende 20 minuten bij kamertemperatuur en gooi het supernatant weg.Suspensie van de celkorrels en tweemaal gewassen met 0,75 molaire kaliumfosfaatbuffer, pH zeven. Pellet de cellen in een centrifugebuis van 50 milliliter door centrificatie bij kamertemperatuur. Gooi het supernatant weg en resuspend de cellen met 0,75 molaire kaliumfosfaatbuffer.Meet het natte gewicht van de pellet met een elektronische balans en resuspend het natte gewicht van één gram van de pellet in vijf milliliter van de buffer. Voeg een milliliter celsuspensie en 0,4 tot 0,6 gram 0,1 millimeter glasparels toe aan een injectieflacon met schroefdop van twee milliliter. Breek de cellen gedurende 30 seconden door een kralenklopper.Laat de buizen twee minuten afkoelen in een waterbad op kamertemperatuur. Centrifugeer na lysis de injectieflacons gedurende 10 seconden bij kamertemperatuur met een snelheid van 500 maal g. Verzamel het supernatant met een pipet in een centrifugebuis van 15 milliliter.Was de kralen één keer met 0,5 milliliter van 0,75 molaire kaliumfosfaatbuffer en breng ze vervolgens over naar dezelfde centrifugebuis. Voeg Triton X-100 toe aan de gelyseerde celsuspensie en incubeer op een schommelende shaker bij kamertemperatuur en 40 RPM gedurende 20 minuten of totdat de oplossing homogeen wordt. Centrifugeer de buisjes gedurende 20 minuten bij 17, 210 maal g en bewaar het supernatant zonder de bovenste Triton-micellaag maximaal een uur bij kamertemperatuur.Maak vier concentraties sucrose in 0,75 molaire kaliumfosfaatbuffer bij pH zeven. Breng 2,8 milliliter van twee molaire sucrosebuffer aan op de bodem van de centrifugebuis van 40 milliliter en leg er drie lagen sucrose-oplossingen overheen. Voeg ten slotte drie milliliter PBS toe die de supernatantfractie bevat en centrifugeer de resulterende gradiënten.Condenseer bij ultracentrificatie de gefractioneerde PBS en fycobiliproteïnen tussen de lagen en observeer de blauwe banden in Syn6803 en interfaces. Verwijder de sucrose door de fracties herhaaldelijk te concentreren en te verdunnen met 0,75 molaire kaliumfosfaatbuffer en membraan de centrifugaalfiltereenheden. Voor het meten van PBS-fluorescentie verdunt u het geconcentreerde PBS-monster met een molaire kaliumfosfaatbuffer van 0,75 molaire kaliumfosfaat tot een volume van maximaal 500 microliter.Voeg het verdunde PBS-monster toe aan een transparante glazen buis en vries de buis in vloeibare stikstof totdat deze volledig bevroren is. Verplaats de bevroren buis naar een transparante dewar container voorgevuld met vloeibare stikstof. Complete cellysis toont de bovennatuurlijke en donkerblauwgroene kleur vóór centrificatie.De sucrosedichtheidsgradiëntscheiding van geïsoleerde PBS toont verschillende fracties van gedissocieerde fycobiliproteïnen en de laagste fractie vertegenwoordigt de intacte PBS. De absorptiespectra van de gedissocieerde en intacte PBS van JSC one en Syn6803 hebben dezelfde piek op 622 nanometer, overeenkomend met het phycocyanine. Bovendien piekt de fycoerythrinabsorptie op 571 nanometer in zowel gedissocieerde als intacte PBS van JSC-1.De emissiespectra van de gedissocieerde PBS hebben één sterke piek op ongeveer 650 nanometer in Syn6803 en JSC-1, wat de emissie van fycocyanine vertegenwoordigt. De maximale pieken van de fluorescerende emissie op 651 nanometer en 684 nanometer laten zien dat de energie die door phycocyanine wordt geoogst, efficiënt werd overgedragen aan allophycocyanine en terminale emitters, die ApcD en ApcE zijn, in de kern. De figuur toont zink gekleurd SDS-PAGE onder UV-bestraling waarbij de phycobiliproteïne-subeenheden sterk fluorescerend zijn en de ApcE zwakke fluorescentie vertoonde, aangegeven met de pijl.Kumasi blauwe kleuring laat zien dat de bovenste fracties andere niet-PBS wateroplosbare eiwitten bevatten en de intacte PBS en Syn6803 toont een prominente ApcE-band, aangegeven met een pijl, die ontbreekt in de gedissocieerde PBS-fracties. Ultracentrifugatie toont aan dat de sucrosegradiënt bereid in een verhouding van 1:3 bredere banden vertoont dan de gradiënt gemaakt in een verhouding van 1:10. Het is belangrijk om de cellen te lysisen en de supernatanten volledig te solucleren om meer fycobilisomen te verkrijgen.Andere methoden, zoals cryo-EM of gemengde spectrometrie, kunnen worden gebruikt om de eiwitstructuur of de eiwitsamenstelling van fycobilisomen te bestuderen.
