这项技术使我们能够研究模拟微重力对免疫细胞的影响，以帮助我们更好地了解空间环境的各个方面如何在分子水平上影响人体生理。与将细胞培养物发送到太空或其他在地球上模拟微重力下进行细胞培养的方法相比，该技术在经济上是可行的，并且相对容易设置和维护。建议在填充容器时缓慢进行，以避免溢出和溢出。初始气泡去除后，使用注射器确保血管完全充满。这将最大限度地减少在整个治疗过程中形成大气泡的机会。首先将培养容器从塑料包装中取出。根据所使用的细胞类型或细胞系标记边缘上的每个容器，无论是在对照还是处理中以及任何其他相关信息。稳定容器并小心打开加注口，不要接触加注口盖钉或 O 形圈。将盖子放在乙醇垫上，将钉子或 O 形圈朝上，同时填充容器。不要从注射器端口上取下盖子。使用无菌血清移液管在容器中装入 10 mL 用于细胞培养的适当完整培养基，并小心地将盖子放在进样口上。确保注射器端口盖和填充端口已牢固关闭。然后，将填充的容器放入培养箱中，同时执行后续步骤以启动用于细胞培养的容器。从培养箱中取出培养基引水容器。对于烧瓶对照，从上述制备的原液中装入装有10 mL细胞培养物的T25悬浮培养瓶。将 10 mL 原液细胞培养物添加到单独的 15 mL 锥形管中，该锥形管将用于加载注射器。拧下注射器端口盖以将其从容器中取出，并确保库存旋塞阀处于打开位置。稳定容器并小心打开加注口，不要接触加注口盖钉或 O 形圈。将盖子放在乙醇垫上，钉子或 O 形圈朝上，同时填充容器。确保旋塞阀处于打开位置。通过使用血清移液管将容器中的培养基倒入废物容器中，可以清空容器。或者，使用无菌玻璃将培养基吸过连接到真空系统的移液器。使用移液器或真空系统时，请确保不要触摸氧合膜，因为这可能会损坏它，并使容器无法使用。紧紧关闭含有制备好的原液培养物的 50 mL 锥形管，轻轻倒置几次以彻底混合内容物。用新鲜的无菌血清移液管从 50 mL 管中吸取 10 mL 细胞培养原液。拿起容器并倾斜它，使填充口朝顶部。然后小心地通过进样口将细胞培养液分配到容器中。将容器填充到填充口的尖端，同时将容器向下倾斜以避免溢出。小心不要用移液器接触氧合膜，因为它非常脆弱。装载容器后，小心地将加注口盖放回原处，不要接触 O 形圈。从包装中取出前 3 mL 注射器，然后泵送几次以使其松开。然后将注射器连接到其中一个注射器端口，确保注射器完全压下。紧紧关闭含有 10 mL 等分试样细胞培养物的 15 mL 锥形管，轻轻倒置几次以彻底混合内容物。同样，从包装中取出第二个 3 mL 注射器并泵送几次。然后小心地将该注射器半浸没在15mL锥形管的细胞培养物中，并抽出一些培养物。通过将注射器完全分配回管中并吸取 3 mL 培养物来最大限度地减少气泡。将填充的注射器连接到剩余的注射器端口，并用乙醇垫仔细消毒注射器和填充端口周围。不要在氧化膜上沾上乙醇。对其余船只重复该过程。如果最后一个容器的 50 mL 锥形管中剩余的细胞培养物略微不足，请从用于装载注射器的 15 mL 锥形管中回收剩余量。确保注射器端口旋塞阀关闭，以限制细胞和气泡从注射器迁移到容器中。将容器倒置并侧击几次以收集初始气泡。快速将容器翻转回朝上，然后以轻微的角度背对着，使气泡全部漂浮到容器的向上一侧。用空注射器操纵端口下方的气泡，并观察它们开始进入端口。打开两个注射器端口旋塞阀。轻轻敲击容器以鼓励气泡漂浮到空注射器中。用空注射器慢慢吸起较大的气泡，同时小心地按下完整的注射器以保持容器内的压力，使氧合膜不会破裂。重复此过程几次以确保去除所有气泡。当气泡被有效去除后，关闭注射器端口旋塞阀。在治疗期间保持注射器打开，以便随后去除气泡，确保两个注射器中的体积大致相等。用 70% 乙醇小心地擦拭旋转底座和带状电缆的表面。确保带状电缆已连接到电源。将旋转底座放入培养箱中，并将带状电缆连接到底座上。将电源放在培养箱附近，但放在培养箱外。通过将容器螺纹排列在旋转钉上并逆时针轻轻转动底座上的旋转钉来连接 SMG 处理容器。确保容器牢固连接。通过在水盘中填充高压釜RO水，将培养箱保持在大约100%的湿度。调整培养箱的旋转速度以匹配细胞的沉降速度，使细胞不会从培养基中掉落。在设置治疗的第一天，每隔几个小时检查一次气泡形成，如文本手稿中所述。每天至少去除一次气泡，直到所需治疗长度结束。一旦超过计划的治疗长度，停止旋转并拆卸设备。通过轻轻保持容器静止，同时顺时针方向转动设备的旋转钉来取出治疗容器。将对照瓶、对照和处理过的容器带入无菌生物安全柜中。取每个容器（不包括对照瓶）并翻转它，使其朝下。然后轻轻敲击它，使所有细胞悬浮。然后将容器侧转，将填充口朝向底部，并再次敲击它以引导细胞朝向填充口以进行有效抽吸。单独处理每个容器。从两个端口拧下注射器，然后将它们分配到生物危害废物中。打开注射器端口上的旋塞阀。小心地取下进样口盖，并使用无菌 10 mL 血清移液管抽取内容物，在清空容器时倾斜容器。将所有对照组和治疗组的内容物分配到单独标记的 15 mL 锥形管中。关闭每个管子并轻轻倒置几次，以确保它们正确混合。使用首选方法确定所得细胞培养物的浓度和活力，为随后的实验测定做准备。将NK-92细胞系的起始活力和座密度与所得的终生存力和终浓度进行比较。经过72小时的SMG治疗后，比较了两个阴性和阳性结果。相比之下，所用NK-92细胞系的最佳浓度范围为每毫升0.3至120万个细胞，倍增时间约为两到三天。SMG治疗组的增殖在对照组和治疗组之间大致相等。这些参数对于下游实验的成功至关重要，来自两个对照组的细胞在下游实验测定中表现相似。在整个处理过程中监测和说明气泡形成至关重要，尤其是在承受模拟微重力的细胞培养物中。大气泡的形成会导致容器内的流体动力学中断，这将基本上否定模拟的微重力条件。
