マイクロ流体プラットフォームは、誘電体移動、磁気泳動、ビーズ抽出、フィルタリング、遠心マイクロ流体学および慣性効果、および弾性表面波1,2に基づく方法に加えて、医療および環境サンプルから細菌を単離するために開発されている。病原菌の検出はポリメラーゼ連鎖反応(PCR)を用いて継続されますが、通常は手間がかかり、複雑で、時間がかかります3,4。マイクロ流体音響泳動システムは、合理的なスループットと非接触細胞分離5、6、7を通じてこれに対処する代替手段です。アコーストフォレーシスは、音波を介した物質の動きの現象を利用してビーズを分離または濃縮する技術です。マイクロ流路に音波が入ると、ビーズの大きさ、密度等に応じて選別され、浮遊培地7,8の生化学的・電気的性質に応じて細胞を分離することができる。そこで、多くの音響フォレーシス研究が盛んに進められ9、10、11、最近では、立地弾性表面波マイクロ流体における境界駆動音響ストリーミングによって誘導される音響ホア運動の3D数値シミュレーションが導入されている12。

このプロトコルでは、アプタマー修飾マイクロビーズを使用して培地からグラム陰性菌を迅速かつ効率的に分離するために使用できる当社のマイクロ流体音響泳動技術について説明します。マイクロ流体音響泳動システムは、合理的なスループットと非接触細胞分離でグラム陰性菌分離を可能にします。マイクロ流体音響泳動は、医療サンプルや環境サンプルから細菌を分離するために最適化することができます。まず、ターゲット出口でPSビーズを収集し、PSサンプル混合物をサンプル入口に注入した後に残りを廃棄するマイクロ流体音響泳動チップを設計します。シリコン層の上下の層をホウケイ酸ガラスに接着したチップを準備します。そして、陽極酸化を使用して、1000ボルトと摂氏400度を追加する3番目のホウケイ酸ガラス層。10マイクロリットル未満のシアノアクリレート接着剤を使用して、マイクロ流体チャネルに沿ってホウケイ酸ガラス層にPZTを取り付けます。GNおよびGP細菌をルリア・ベルターニ培地に接種し、摂氏37度、毎分220回転で16時間インキュベートします。培養細菌を9, 056 RCFで室温で1分間遠心分離します。その後、1X PBSバッファーで2回洗浄します。選択したGNおよびGP細菌を結合バッファーに再懸濁して分析用に準備します。使用前に、10マイクロメートルのストレプトアビジンコーティングされたマイクロビーズ混合物を再懸濁してください。混合物を20秒間ボルテックスします。アプタマーを摂氏95度で3分間変性させ、次に摂氏0度で2分間折り直して準備します。再懸濁したストレプトアビジンコーティングされたマイクロビーズ混合物250マイクロリットルを1.5ミリリットルのチューブに移し、100マイクロリットルのビオチン化DNAアプタマーをチューブに加えます。混合物を室温で30分間インキュベートし、毎分25回転で回転させます。反応混合物を室温で40秒間9, 056 RCFで遠心分離する。次に、200マイクロリットルのTris-HClバッファーでチューブを2回洗浄します。洗浄したサンプルチューブに10マイクロリットルの100ミリグラム/ミリリットルBSAを加え、毎分25回転で回転させながら室温で30分間インキュベートします。最後に、アプタマー修飾マイクロビーズをTris-HClバッファー中で2回洗浄し、室温で40秒間9, 056RCFで遠心分離する。PEEKチューブを2つのサンプルとバッファーを注入するための2つの入口と、廃棄物を収集および排出するための2つの出口に接続します。10ミリリットルのシリンジを使用して、マイクロ流体音響泳動チャネルに気泡のない脱塩水を満たします。流体の流れを制御するために、2つ以上の出力チャネルを備えた精密圧力コントローラーを準備します。装置を準備した後、精密圧力制御装置を使用して、サンプル入口に2キロパスカル、バッファー入口に4キロパスカルの圧力を加えて、サンプルとバッファーを注入します。顕微鏡を使用してビーズに焦点を合わせ、PZTを使用してマイクロ流体チャネルの中心に移動します。シングルチャンネルのファンクション・ジェネレータを使用して3.66メガヘルツの共振周波数を生成し、パワーアンプを使用して標準信号を16デシベル増幅します。音響流体チップ上の分離および濃縮プロセスを、蛍光顕微鏡と毎秒1, 200フレームで動作する高速カメラで観察します。本研究で紹介した音響膜チップは、PZTの電圧が上昇すると、10マイクロメートルサイズのビーズの中心濃度が増加することを確認した。PZT電圧の5ボルトでは、10マイクロメートルサイズのビーズのほとんどが中心に集中していました。注入された10マイクロメートルサイズのビーズの総数に対する出口で採取したビーズの数の割合を回収率とした。採取したビーズの総数に対する10マイクロメートルサイズのビーズの数の割合を純度と称した。その結果、本装置は10マイクロメートルサイズのビーズに対して高い分離効率を有することが示された。各アプタマー修飾マイクロビーズに結合したGNおよびGP細菌の数を、高速度カメラを備えた顕微鏡を用いて測定した。5つのGN細菌すべてがビーズに結合しましたが、テストされた物質に結合したGP細菌は大幅に少なかった。シグナル強度はGN菌とGP菌で有意に異なっていた。PZTの取り付けに使用する接着剤は、波形の精度のためにできるだけ使用せず、PZTとマイクロチャネルを平行にする必要があります。この装置は、細菌感染症の早期診断の期間に生きた細菌または細菌由来のバイオマーカーを検出するために使用することができる。
