유전체 전달, 자기 영동, 비드 추출, 필터링, 원심 미세 유체 공학 및 관성 효과, 표면 음향파 1,2에 기반한 방법 외에도 의료 및 환경 샘플에서 박테리아를 분리하기 위해 미세 유체 플랫폼이 개발되고 있습니다. 병원성 박테리아의 검출은 중합효소 연쇄 반응 (PCR)을 사용하여 계속되지만, 일반적으로 힘들고 복잡하며 시간이 많이 걸립니다 3,4. 미세유체 acoustophoresis 시스템은 합리적인 처리량 및 비접촉 세포 분리 5,6,7을 통해 이를 해결하기 위한 대안이다. 여기서, 본 글에서는 앱타머 변형된 마이크로비드를 사용하는 배지로부터 그람 음성(GN) 박테리아를 신속하고 효율적으로 분리하는데 사용될 수 있는 미세유체 acoustophoresis 기술 프로토콜을 설명한다. 이 시스템은 긴 직사각형 마이크로채널 내에서 두 개의 직교 공진을 동시에 자극하여 단일 압전 작동을 통해 2차원(2D) 음향 스탠딩파를 생성하여 노드와 안티노드 포인트에서 압타머 부착 마이크로비드를 정렬하고 집중시켜 분리 효율 2,11,15,16 . 입구와 출구 모두에 분기 채널이 있어 분리, 정화 및 농축을 동시에 수행할 수 있습니다.
이 프로토콜은 박테리아 전염병의 조기 진단 분야뿐만 아니라 실시간 물 모니터링을 통해 병원성 박테리아 감염에 대한 신속하고 선택적이며 민감한 반응에 도움이 될 수 있습니다.
