この制限に対処するために、我々は、チップ上の生体内微小血管ネットワークを現実的に再現する新規な3D生体模倣マイクロ流体アッセイ(bMFA)(図2)を開発し、広範囲に検証した16、17、18。このデバイスの微細加工のためのプロトコルは、以前に17で公開されており、ここでは簡単に説明されています。マウスクレマスター筋の微小血管系を、修正地理情報システム(GIS)アプローチ19を用いてデジタル化した。次いで、合成微小血管網を、デジタル化された微小血管網14、17、20、21、22に基づくソフトリソグラフィープロセスを用いて、ポリジメチルシロキサン(PDMS)上に生成した。簡単に言えば、デジタル化されたネットワーク画像をマイラーフィルムに印刷し、それをマスクとして使用して、シリコンウェーハの上にSU-8ポジ型フォトレジストをパターニングし、製造用のマスターを作成しました。微細加工されたピラー(直径10μm、高さ3μm)を用いて、高さ3μm、幅100μmの細孔を作成し、白血球遊走に最適なサイズ23、24、25、血管チャネルと組織区画を接続した。PDMSはメーカーの指示に従って調製され、開発されたマスターに注がれました。さらに、PDMSを脱気し、オーブン(65°C)中で一晩硬化させて、PDMS中に相補的なマイクロチャネルを作成した。続いて、硬化したPDMSをSU-8マスターから剥離し、続いて入口/出口用のパンチングポートを取り付けた。次いで、PMDSをスライドガラスにプラズマ接合した。マイクロ流体装置の表面は、天然ガラスおよびPDMSを含む。細胞の付着、拡散および増殖を促進するためには、細胞外マトリックス(ECM)コーティングが必要である。bMFAには、微小血管網と、高さ3μm、幅100μmの細孔を介して接続された組織区画が含まれています(図2)。このマイクロ流体システムは、循環、縁取り、転がり、接着および単一系における血管外組織区画への白血球の移動を含む、相互接続された血管および分岐を有する完全な微小血管網の生理学的に関連する3D環境における白血球接着/遊走カスケード全体を再現する14,16,17,21,26。
なお、bMFAの入口の流量が固定されていても、ネットワーク内の流動条件は場所によって異なり、単純な数式では計算できない。計算流体力学(CFD)ベースのモデルが開発され、ネットワーク内のさまざまな場所でさまざまな流れパラメータ(せん断応力、せん断速度、速度など)が計算されました。このCFDモデルは、bMFAの色素灌流パターンと流れパラメータをシミュレートするために使用されました。実験結果との交差検証により、ネットワーク全体の流れ抵抗は計算モデルによって十分に予測されることが示唆されました(図3)17。次に、このCFDモデルを使用して、bMFAのすべての容器における速度およびせん断速度プロファイルを推定し(図4)、白血球の圧延、接着および移動に対するせん断流および幾何学的形状の影響の分析を可能にした16。白血球は、分岐点の近くおよび生体内の低剪断領域において優先的に接着し、白血球接着のこれらの空間パターンは、好中球を用いたbMFAにおいて首尾よく実証された(図5)16。本稿では、ヒト肺微小血管内皮細胞(HLMVEC)およびヒト好中球を用いて炎症条件下で白血球-内皮細胞相互作用を研究するためのbMFAを調製するためのプロトコールについて記載する。bMFAなどの微生理学的システムは、好中球、単球、リンパ球および腫瘍細胞などの異なるタイプの細胞との内皮細胞相互作用を研究するために使用することができる18、27、28、29、30。bMFAを、異なる器官(例えば、肺対脳)および異なる種(例えば、ヒト対マウス内皮細胞)、ならびに内皮細胞株21、27、31、32からの初代内皮細胞で播種することができる。bMFAは、複数の細胞応答、細胞間相互作用、バリア機能、薬物送達および薬物毒性を研究するために使用することができる。
