Het neurale netvlies is een zeer georganiseerd weefsel dat goed gedefinieerde neuronale lagen presenteert. Hierin zijn neuronen (ganglion,amacrine, bipolaire, horizontale en fotoreceptorcellen) met elkaar verbonden en ook met Müller-gliacellen (MGC's) en astrocyten, wat leidt tot adequate fototransductie en verwerking van visuele informatie 1,2. Van MGC's is bekend dat ze een belangrijke rol spelen bij het onderhoud van retinale homeostase omdat ze het hele retinale gedeelte doorkruisen en dus kunnen interageren met alle celtypen die meerdere beschermende processen moduleren. Er is gemeld dat MGC's verschillende belangrijke functies hebben voor het onderhoud en de overleving van retinale neuronen, waaronder glycolyse om energie te leveren aan neuronen, de verwijdering van neuronaal afval, de recycling van neurotransmitters en de afgifte van neurotrofe factoren, onder andere 3,4,5.
Aan de andere kant zijn ontsteking, oxidatieve en nitrosatieve stress betrokken bij de pathogenese en progressie van veel menselijke ziekten, waaronder retinopathieën 6,7,8,9,10,11. De redoxbalans in cellen is afhankelijk van een strakke regulering van ROS-niveaus. ROS worden constant gegenereerd onder fysiologische omstandigheden als gevolg van aërobe ademhaling voornamelijk. De belangrijkste leden van de ROS-familie zijn reactieve vrije radicalen zoals het superoxide-anion (O2͘͘͘͘•−), hydroxylradicalen (•OH), verschillende peroxiden (ROOR′), hydroperoxiden (ROOH) en het geen radicale waterstofperoxide (H2O2)12,13. In de afgelopen jaren is gebleken dat ROS een belangrijke signaalrol speelt in de cellen door essentiële processen te beheersen. MGC's hebben een sterke antioxidantafweer door de activering van de transcriptionele nucleaire factor erytroïde-2-gerelateerde factor 2 (Nrf2) en de daaropvolgende expressie van antioxidanteiwitten om de overmatige productie van ROS onder pathologische omstandigheden te elimineren 14,15,16. Wanneer de cellen hun redoxbalans verliezen als gevolg van een overdreven productie van ROS of een defect vermogen om ROS te verwijderen, bevordert de accumulatie van oxidatieve stress schadelijke modificaties in eiwitten, lipiden en DNA, wat leidt tot cellulaire stress of de dood. De toename van het retinale antioxidantafweersysteem verbetert de resolutie en preventie van retinopathieën, zoals ROP en RD 17,18,19,20,21,22,23,24. Daarom is het meten van ROS-productie in real-time een krachtig en nuttig hulpmiddel.
Er zijn verschillende methoden voor het meten van ROS-productie of oxidatieve stress in cellen. Onder deze, 2′,7′-dichlorofluorescein diacetaat (DCFH-DA) sonde is een van de meest gebruikte technieken voor het direct kwantificeren van de redoxtoestand van een cel 25,26,27,28. Deze sonde is lipofiel en niet-fluorescerend. Diffusie van deze sonde over het celmembraan maakt de splitsing mogelijk door intracellulaire esterasen bij de twee esterbindingen, waardoor een relatief polair en celmembraan-ondoordringbaar product ontstaat, 2′,7′-dichlorofluorescein (H2DCF). Dit niet-fluorescerende molecuul accumuleert intracellulair en daaropvolgende oxidatie door ROS levert het zeer fluorescerende product DCF op. De oxidatie van de sonde is het product van de werking van meerdere soorten ROS (peroxynitriet, hydroxylradicalen, stikstofmonoxide of peroxiden), die kunnen worden gedetecteerd door flowcytometrie of confocale microscopie (emissie bij 530 nm en excitatie bij 485 nm). De beperking van deze techniek is dat superoxide en waterstofperoxide niet sterk reageren met H2DCF25,29. In dit artikel gebruiken we DCFH-DA probe om ROS te meten en te kwantificeren door middel van flowcytometrie. Om die reden induceren we ROS-productie door MGC's te stimuleren met ROS-inductor, A of B, voorafgaand aan het laden van de cellen met de fluorescerende sonde. Daarnaast gebruiken we een antioxidantverbinding. Ten slotte tonen we representatieve en betrouwbare gegevens die met behulp van dit protocol zijn verkregen.

De redoxbalans speelt een belangrijke rol bij het handhaven van cellulaire homeostase. De verhoogde generatie van reactieve zuurstofsoorten (ROS) bevordert de modificatie van eiwitten, lipiden en DNA, wat uiteindelijk kan leiden tot verandering in cellulaire functie en celdood. Daarom is het gunstig voor cellen om hun antioxidantafweer te verhogen als reactie op schadelijke beledigingen, hetzij door een antioxidantroute zoals Keap1 / Nrf2 te activeren of door redox-aaseters te verbeteren (vitamine A, C en E, β-caroteen en polyfenolen, onder anderen). Ontsteking en oxidatieve stress zijn betrokken bij de pathogenese en progressie van retinopathieën, zoals diabetische retinopathie (DR) en retinopathie van prematuriteit (ROP). Aangezien Müller-gliacellen (MGC's) een sleutelrol spelen in de homeostase van neuraal netvliesweefsel, worden ze beschouwd als een ideaal model om deze cellulaire beschermende mechanismen te bestuderen. In die zin is het kwantificeren van ROS-niveaus met een reproduceerbare en eenvoudige methode essentieel om de bijdrage van pathways of moleculen die deelnemen aan het antioxidant celafweermechanisme te beoordelen. In dit artikel geven we een volledige beschrijving van de procedures die nodig zijn voor het meten van ROS met DCFH-DA-sonde en flowcytometrie in MGC's. Belangrijke stappen voor flowcytometriegegevensverwerking met de software worden hier gegeven, zodat de lezers ROS-niveaus (geometrische middelen van FITC) kunnen meten en fluorescentiehistogrammen kunnen analyseren. Deze hulpmiddelen zijn zeer nuttig om niet alleen de toename van ROS na een cellulaire belediging te evalueren, maar ook om het antioxiderende effect van bepaalde moleculen te bestuderen die een beschermend effect op de cellen kunnen hebben.

Quantification of Reactive Oxygen Species Using 2ÃƒÂ¢Ã¢â€šÂ¬Ã‚Â²,7ÃƒÂ¢Ã¢â€šÂ¬Ã‚Â²-Dichlorofluorescein Diacetate Probe and Flow-Cytometry in MÃƒÆ’Ã‚Â¼ller Glial Cells

Hier stellen we een gesystematiseerd, toegankelijk en reproduceerbaar protocol voor om cellulaire reactieve zuurstofsoorten (ROS) te detecteren met behulp van 2′,7′-dichlorofluorescein diacetate probe (DCFH-DA) in Müller gliacellen (MGC's). Deze methode kwantificeert de totale cellulaire ROS-niveaus met een flowcytometer. Dit protocol is zeer eenvoudig te gebruiken, geschikt en reproduceerbaar.

Er zijn verschillende methoden voor het meten van ROS-productie of oxidatieve stress in cellen. Onder deze, 2'7'dichlorofluoresceïne diacetaat sonde is een van de meest gebruikte techniek voor directe meting van redox toestand. Deze sonde is lipofiel en niet-fluorescerend. Diffusie van deze sonde over het celmembraan zorgt ervoor dat de intracellulaire esterasen zich splitsen bij de twee esterbindingen, waardoor een relatief polair en celmembraan-ondoordringbaar product ontstaat. Deze niet-fluorescerende moleculen accumuleren intracellulair en daaropvolgende oxidatie door ROS levert het zeer fluorescerende product dichloorfluoresceïne op. De oxidatie van de sonde is het product van de werking van meerdere soorten ROS. Die kan worden gedetecteerd door flowcytometrie of confocale microscopie. In dit artikel hebben we een dichloormelincetaatsonde gebruikt om ROS te meten en te kwantificeren door middel van flowcytometrie. Breng 6-well platen over in een laminaire stroomkap van de incubator. Aspirateer de supernatant. En was de cellen een keer met PBS. Voeg 2 ml laag serum DMEM per putje toe. En incubeer de 6-well plaat gedurende twee uur in de incubator. Behandel de cellen met de antioxidant gedurende zes uur. Behandel de cellen met de ROS-inductor, A of B, gedurende 30 minuten. Aspirateer de supernatant. En was de cel drie keer met PBS om alle prikkels te verwijderen. Doe het licht van de laminaire flowkap uit en werk in het donker. Voeg 1 ml dichloorbelincetaatprode toe in DMEM zonder fenolrood. Voeg 1 ml DMEM zonder fenolrood toe aan de controleautofluorescentiecontrolecellen. Incubeer de 6-well plaat gedurende 30 minuten in de incubator. Breng de platen op ijs over naar het laboratorium. Was de cellen drie keer met 2 ml koude PBS per put. Voeg 0,5 ml koude losmaakbuffer toe aan elke put. Oogst de cellen door voorzichtig op en neer te pipetteren met een P1000-pipet. Verzamel ze in een gelabelde buis van 1,5 ml. En centrifugeer ze bij 600 x g gedurende vijf minuten bij 4 C.Gooi het supernatant voorzichtig weg en dissociëer de pellet voorzichtig met tikken. Voeg 1 ml FACS-buffer toe om de cellen aan elke gelabelde buis te wassen. Gebruik indien nodig vortex in de laagste intensiteit voor de volledige dissociatie van de pellet. Centrifugeer ze bij 600 x g gedurende vijf minuten bij 4 C.Herhaal deze stap twee keer meer. Drie wasbeurten in totaal. Resuspend celkorrels in 200 L FACS-buffer. Dissociëer voorzichtig de pellet in elke buis met behulp van vortex. En overbrengen naar gelabelde 5 ml voor flowcytometerbuizen. Houd de buisjes op ijs totdat ze op de flowcytometer worden geanalyseerd. Maak met behulp van de flowcytometriesoftware een nieuw experiment. Klik op het exemplaar en open een lijst met buizen. Dubbelklik en wijzig de naam. Plaats de autofluorescentiecontrolebuis op de aspiratorarm en beweeg de basis voorzichtig alleen naar links. Klik op Gegevens ophalen en vervolgens op Gegevens opnemen en selecteer de gewenste stopvoorwaarde. Teken een poort rond de cellen van belang, met uitzondering van dode cellen en puin. Verwijder de tube. Klik op Volgende buis en voer het basale controlemonster uit. Klik op Gegevens ophalen en vervolgens op Gegevens vastleggen.Open de software. Voeg de voorbeelden toe met de actieknop Voorbeelden toevoegen. Klik op Voorbeeld toevoegen.Selecteer de map Experimentele datum en klik op Kiezen.Dubbelklik op het eerste voorbeeld in uw werkruimte, in dit geval autofluorescentiebesturingselement.
