基質粘度は細菌の運動性に影響を及ぼす。 ヘリコバクター・ピロリなどの病原性微生物の研究では、病原体の運動性はムチン層の粘度によって変化し、ヒト宿主の環境酸性化の影響を受けることが示されています6。これらの環境を再現するために、0.3%(w / v)を超える寒天濃度を使用した以前の研究では、細菌の遊泳運動性が制限され、表面群動への漸進的なシフトがもたらされた。1%(w / v)を超える寒天濃度の使用は、多くの種の群れ運動性を防ぎます7。表面に形成されるコロニーパターンは、特徴のないmat8、雄牛の目9、樹状突起10、渦11など、多様です。
このようなパターンの関連性は不明のままですが、これらのパターンは環境的および化学的手がかりに依存しているようです12。環境の手がかりは、温度、塩分濃度、光、pHなどのさまざまな側面をカバーしていますが、化学的手がかりには、微生物クォーラムセンシング分子、生化学的副産物、栄養素の存在が含まれます。AHL(N-ヘキサノイル-Lホモセリンラクトン)などの自己誘導物質クォーラムセンシングシグナル伝達分子は、界面活性剤の産生を調節することによって表面群動に影響を与えることができる13,14。フィトアレキシン化合物であるレスベラトロールは、細菌の群れ運動性を制限する可能性があります15。
本研究では、野生型大腸菌K12株に対するレスベラトロールの勾配濃度の影響を調査し、操作された大腸菌K12-YdeHおよび緑膿菌PAO1-YdeH種のアラビノース誘導性群動性を調べる。YdeH酵素の産生はaraBADプロモーターを介してアラビノースによって誘導され、細胞のc-di-GMP摂動をもたらし、細菌群動性に影響を与える16,17。この誘導性群動は、大腸菌K12-YdeHおよび緑膿菌PAO1-YdeH株を用いたアラビノース勾配群プレートを用いて研究される。
この勾配温板は、二層媒体を順次固化させることにより調製される(図1B)。最下層は、インデューサーと共に添加された培地を含み、支柱状のペトリ皿の片面に注がれる。最下層の固化に際し、シャーレは平坦な表面に戻り、そこでインデューサを含まない媒体を含む上層がプレートの反対側から添加される。スウォームプレートが完全に固化した後、固定レイアウトに従ってウォームプレートに穴を開けることによって(図1C)、または媒体硬化プロセス中にペグを含むプレートカバーの3Dプリントモデルを使用してウェルをインプリントすることによって(補足図S1)、等尺的に配列された保持ウェルが生成される。ゲルイメージングシステムは、異なる時点での群れ形態をキャプチャするために使用されます(図2)。ImageJソフトウェアを使用した表面スウォーミングの解析(補足図S2)は、表面スウォーミングプロセスの定量的結果を提供します(図3)。

このプロトコルは、半固体寒天培地上の異なるインデューサー濃度に対する細菌群集応答を同時に追跡する。これは費用対効果の高い方法で達成されます。このハイスループットスクリーニング技術は、微小走化性応答を観察するための様々なインデューサー濃度を有する寒天の必要性を否定する。この技術は、薬物スクリーニングのために設計されており、また、微小病原体SD応答を観察するためにも設計されている。また、ヒト宿主における様々な生化学的手がかりに対する微小応答を研究するために使用することができる。この方法は、細菌走化性についての洞察を提供することができる。細菌は、正の走化性または負の走化性として知られている化学的忌避剤として知られる様々な濃度の化学誘引剤に応答する。慎重に準備されたセットアップは、再現性の問題を防ぐために、下層のゲル角度間のばらつきを避けるために不可欠です。まず、13 x 13 cm 四方のペトリ皿に汚れと誘導剤の名前のラベルを付けます。それからふたの端の上に皿を支えます。アラビノース溶液を温める最下層培地に加える。40ミリリットルの暖かい最下層培地を加える。最下層の媒体を層流フード内で1時間覆い隠さずに硬化させる。最下層が完全に固化したら、蓋を外し、ペトリ皿を層流フードの中に置きます。電動ピペットフィルターを使用して40ミリリットルの暖かい上層培地を加える。覆って邪魔されない二重層プレートを1時間硬化させ、摂氏4度で24時間保管します。マークされたA4用紙を、よく固化したグラデーションプレートの下に置きます。100マイクロリットルのピペットチップの広い側をマークされた位置の半固体培地表面に押し込み、上層培地の底部に達するまでピペットチップを押します。先端が底部に触れたら、先端に垂直な力を加えるのをやめ、先端を静かに回転させて円筒形の井戸の内容物を分離します。ピペットチップを少し離れたところに水平に動かして、脇に置かれた狭い場所に空気が流れるようにします。先端を人差し指で押して、先端内部のガスの流れを遮断します。先端を垂直に引き出し、井戸の内容物を先端に保持しながら引き出します。一晩の成長培養物の80マイクロリットルをすべての井戸に加える。12時間ごとに1枚ずつインキュベーターからウォームプレートを取り出し、ゲルイメージングシステムに入れます。ゲルイメージングモードを選択し、群れプレートを白色光にさらし、焦点距離を調整して群れの鮮明な視界を得る。露出時間を300ミリ秒に調整して正確な観察のために群れの明るさを高めます。しきい値を調整して、背景ライトからの干渉を最小限に抑えます。さらに分析するために画像ファイルを保存します。イメージング時間、インデューサの種類、勾配の向き、ひずみをtxtファイルに記録します。「プロセス」をクリックし、「シャドウ」をクリックして、画像のシャープネスを高めます。[処理]をクリックしてから[バッチ]をクリックして、画像を処理します。次に、[プロセス]、[シャドウ]、[南]をクリックして、画像を参照として処理します。「プロセス」、「バッチ」、「マクロ」をクリックして、「バッチ・プロセス」ウィンドウを開きます。ウィンドウに表示されるコマンドを探します。「バッチ処理」ウィンドウで「処理」をクリックして、出力ファイルアドレスに元のイメージのフォルダアドレスを入力します。杖トレースツールを使用して群れを個別に選択し、杖トレースツールをダブルクリックして、生成されたラインが群れの境界に正しく収まるまで公差を調整します。[分析]をクリックし、[測定]をクリックしてエリア値をエクスポートします。大腸菌K12 YdeHを、隣接するウェル間で生じるオーバーラップを伴う表面群動プロセスを実証するために使用したアラビノース勾配プレート上で試験した。3つの試験ウェルを備えたプレートは、重なり合わない境界の形成を可能にした。緑膿菌PAO1 YdeH群動運動性は、最低濃度からアラビノース濃度の増加に伴って促進されたが、アラビノース濃度が高い濃度では徐々に制限された。ゼロ〜400マイクロリットル濃度範囲内のレスベラトロール勾配プレート上で試験した大腸菌K12野生型株は、レスベラトロール濃度の増加に伴う群群運動性の適度な制限を示した。群れ領域は、ImageJソフトウェアによって定量化した。群れ曲線には、複数の濃度応答が表示されました。この手順の最も重要な部分は、特定の濃度範囲のインデューサを備えた二重層ウォームプレートを製造することです。この方法により、研究者は特定の濃度範囲内で誘発された表面群動を調査し、群発に対する他の誘導物質および成分の影響を定量化することができる。
