이러한 패턴의 관련성은 불분명하지만, 이러한 패턴은 환경 및 화학적 단서에 의존하는 것으로 보입니다12. 환경 단서는 온도, 염분, 빛 및 pH를 포함한 다양한 측면을 포괄하는 반면, 화학적 단서에는 미생물 쿼럼 감지 분자, 생화학 부산물 및 영양소의 존재가 포함됩니다. AHL(N-헥사노일-L 호모세린 락톤)과 같은 자동 유도기 정족수 감지 신호전달 분자는 계면활성제13,14의 생산을 조절함으로써 표면 떼에 영향을 줄 수 있다. 피토알렉신 화합물인 레스베라트롤은 박테리아의 떼 지어 모이는 운동성을 제한할 수 있다15.
본 연구에서는 야생형 에스케리치아 콜라이 K12 균주에 대한 레스베라트롤의 구배 농도의 영향을 조사하고 조작된 대장균 K12-YdeH 및 슈도모나스 aeruginosa PAO1-YdeH 종의 아라비노스 유도성 떼 운동성을 조사하였다. YdeH 효소의 생산은 araBAD 프로모터를 통해 아라비노스에 의해 유도되며, 이로 인해 세포 c-di-GMP 교란이 발생하고 박테리아 떼 운동성에 영향을 미친다16,17. 이 유도성 떼 거동은 대장균 K12-YdeH 및 P. aeruginosa PAO1-YdeH 균주를 갖는 아라비노스 구배 떼 플레이트를 사용하여 연구된다.
구배 떼 플레이트는 이중층 매질을 연속적으로 고형화하여 제조된다(그림 1B). 바닥층은 유도제와 함께 첨가된 배지를 포함하며, 소품이 올라간 페트리 접시의 한쪽에 부어진다. 바닥층이 응고되면 페트리 접시는 평평한 표면으로 되돌아 가며, 여기서 유도제가없는 배지를 포함하는 상층이 플레이트의 다른면에서 추가됩니다. 떼 플레이트가 완전히 응고된 후, 등각 방향으로 배열된 홀딩 웰은 고정된 레이아웃(그림 1C)에 따라 떼 플레이트에 구멍을 뚫거나 중간 경화 공정 중에 못을 포함하는 플레이트 커버의 3D 프린팅된 모델을 사용하여 웰을 각인함으로써 생성됩니다(보충 그림 S1). 겔 이미징 시스템은 서로 다른 시점에서 떼를 지어 모은 형태를 포착하는 데 사용됩니다(그림 2).
