この方法は、基礎となるグラフェン格子を維持しながらPMMA残基を効果的に除去する。この機能デバイスは、血清中のIgG抗体を検出する際に一貫した結果を示す。また、このプロトコルは、CVDグラフェンのリアルタイムのラベルフリーバイオセンシングデバイスへの実装を保証します。手順は非常に簡単で、最小限のトレーニングで実行できます。このデバイスは、他のバイオセンシングデバイスよりも高い選択性、高感度、およびリアルタイム検出を提供します メスを使用して銅基板上のグラフェンシートを半分に切断することから始めます。耐熱テープを貼り、グラフェン四角の四隅をスピナーガスケットに固定します。グラフェンの正方形のシートをPMMA 495K A4の100〜200ナノメートルの薄層でスピンコートし、毎分500回転で10秒間回転させ、次いで毎分2,000回転で50秒間回転させる。その後、サンプルを摂氏150度で5分間焼きます。毎分15標準立方センチメートルの速度流を用いて30ワットの酸素プラズマでグラフェンの裏面を5分間除去する。プラズマ処理したグラフェンを正方形に幅1センチ、高さ2センチメートルの寸法にカットし、デバイス作製に役立てます。あらかじめ洗浄したシリカ基板を、幅約4センチ、高さ2センチの小片に切断します。グラフェンエッチング液を希釈せずに塩化第二鉄を用いて銅をエッチングオフする。銅側を下にしてPMMA側を上にしてサンプルを液体エッチング液に浮かべます。銅エッチング後、プラズマ処理基板を用いてグラフェン膜をゆっくりと持ち上げる。転写したグラフェン膜を2時間風乾し、その後ホットプレート上で焼成する。PMMAを除去するには、PMMA側を下に向けて4分間アセトン蒸気より約2センチメートル上に保ち、アセトン蒸気でサンプルを70°Cで温めることから始めます。次いで、試料をアセトンに5分間浸漬する。サンプルをイオン交換水で慎重に洗ってください。最後に、試料を窒素で穏やかに吹き付け乾燥させる。転写したグラフェンで基板をアセトン、イソプロピルアルコール、およびイオン交換水を用いて洗浄する。その後、基板を75°Cのホットプレート上で30分間焼成する。電子ビーム蒸発器を用いて、グラフェン試料上にそれぞれ5ナノメートルおよび45ナノメートルの厚さのニッケルおよび金を堆積させる。電極のパターニングのためにマスクAを用いた第1のフォトリソグラフィープロセスを適用する。サンプル上にAZ 5214Eポジ型フォトレジストを毎分2,000回転で45秒間スピンし、120°Cで1分間サンプルを硬化させます。サンプルを紫外線洪水暴露システムに入れ、1平方センチメートルあたり200ミリジュール以下で約10秒間露光する。フォトレジスト現像剤AZ 300MIFで約2分間サンプルを現像し、イオン交換水ですすいでください。試料を金エッチング液に浸漬して金層を10秒間エッチングし、イオン交換水ですすぎ、アセトンに10分間浸漬して残りのフォトレジスト層を除去した。アセトン、イソプロピルアルコール、およびイオン交換水を用いて、サンプルを洗浄した後、75°Cのホットプレート上で30分間焼成した。次に、マスクBを用いた第2のフォトリソグラフィー工程を適用して、グラフェンチャネルをパターニングする。60°Cのニッケルエッチング液にサンプルを浸し、ニッケル層を10秒間エッチングする。脱イオン水ですすぎ、窒素で吹き飛ばして乾かします。サンプルをプラズマアッシャーに入れ、酸素プラズマを使用して露出したグラフェンを除去します。その後、アセトンに10分間浸漬してフォトレジスト層を除去する。アセトン、IPA、脱イオン水でサンプルを洗浄し、75°Cのホットプレートで30分間焼きます。マスクCを用いた第3のフォトリソグラフィープロセスを適用してパッシベーションフォトレジスト層をパターニングし、基板上の下地グラフェンを保護する。ポジ型フォトレジストによるスピン、サンプルの硬化、フォトレジスト現像液による現像など、前述ののと同じプロセスパラメータを使用します。その後、試料を60°Cのニッケルエッチング液に10秒間浸漬して残りのニッケル層を除去した後、イオン交換水ですすぎ、窒素を用いてブロードライした。最後に、サンプルを摂氏120度のホットプレートで30分間焼きます。代表的な結果は、ラマンおよび原子間力顕微鏡によって特徴付けられる転写されたCVDグラフェンを示す。ラマン画像のGピークと2次元ピークは、転写された単層グラフェンの存在と品質に関する包括的な情報を提供します。図は、試料を収容するための塩化銀参照電極およびポリジメチルシロキサンウェル中の標準銀と一体化したEEG FETバイオセンサを示す。さらに、グラフェンチャネルの拡大図は、ソース電極からグランドへの接続、ドレイン電極、およびソースへのゲート電極への接続を示しています。グラフェンの広く使用されている官能化試薬であるPBASEは、グラフェンの電気的特性を損なうことなく、π-pi相互作用を介してグラフェン表面に吸収することができる。5プライムアミノ修飾IgGアプタマーは、PBASEにおける反応性およびヒドロキシ−6シンアミドエステルとの間のアミド結合結合、およびIgGアプタマーの5プライム末端上のアミン基によってPBASEと共役する。単一強度PBSでデバイスをすすぎた後に残りの非共役部位をブロックするために使用したウシ血清アルブミンインキュベーションを図は、異なる電解質条件下でのIgG検出を示す。グラフェンの品質は、このデバイスの最高の性能の鍵です。したがって、プラズマエッチング中に、プラズマがグラフェンの有用な領域に害を及ぼさないことを確認する必要があります。また、PMMA残基は、クリーンなグラフェン表面を得るために完全に洗浄する必要があります。この機能デバイスは、ヒトIgG抗体の検出において一貫した結果を示すため、この手順は、界面相互作用およびバイオセンシングを研究するための他のナノ材料を用いてデバイスを構築するための基準として使用することができる。
