光干渉断層撮影は、さまざまな網膜疾患の臨床診断に広く使用されていますが、マウスモデルに適用するのはまだ困難です。このプロトコルは、動物におけるOCT技術を管理する方法を説明しています。この技術は、眼球のサイズが異なる動物に適用できます。非侵襲的、高解像度、リアルタイムの網膜断面イメージングと、網膜の厚さの定量的測定を可能にします。OCTは、病変の位置、範囲、形状、サイズなどの病変の特徴、および網膜新生血管などの疑わしいすべての網膜異常を特定することにより、網膜症の診断を支援します。このプロトコルは、網膜疾患を含む動物実験または眼底疾患に適用されます。手順を開始する前に、準備をしてプロトコルを段階的に実行してください。動物が麻酔をかけられたときに実験を行います。ある程度の練習をすれば、高品質のOCT画像を取得できるはずです。まず、コンピューターの電源を入れてソフトウェアを起動します。テストプログラムボタンをクリックして、テストプログラムを完了します。次に、サーモスタットをオンにして、摂氏37度に予熱します。プログラムのテストが完了したら、OCT モジュール プロシージャを開始します。新しい件名を作成し、マウス情報を入力します。次に、電気毛布を予熱し、サージカルタオルで覆います。麻酔をかけたマウスを電気毛布のプラットフォームに置き、すぐに両目を医療用ヒアルロン酸ナトリウムゲルでコーティングします。次に、マウス角膜に100Dコンタクトレンズを置き、凹面側が角膜表面のヒアルロン酸ナトリウムゲルに触れます。次に、cSLOデバイスの小型で恒温の動物プラットフォームにマウスを置きます。瞳孔をレンズの中央に保つために、鉗子でコンタクトレンズの角度を調整します。頭の調整を微調整して、目の向きをまっすぐにします。OCT ボタンをクリックし、マウス モジュールを選択して、cSLO プログラムを起動します。次に、操作レバーを使用して、プリセットレンズをコンタクトレンズに向かってゆっくりと動かします。視神経乳頭を中心に、前後極の画像を揃えるためにさらに調整します。次に、OCT プログラムを起動し、OCT イメージが表示されるまで進行状況バーを上下にクリックします。最小範囲を0〜20、最大範囲を40〜60に調整します。次に、理想的なOCT画像が得られるまで、プリセットレンズ距離と位置方向を調整します。cSLOの標準ラインを移動して、スキャン位置を選択します。次に、平均をクリックしてcSLOとOCTの画像信号をオーバーレイし、ショットボタンをクリックしてSLO OCT画像を取得します。実験後、100Dコンタクトレンズを取り外し、レボフロキサシンアイジェルを適用して角膜を保護します。網膜層別化補正を行うには、OCTインターフェイスの負荷検査をクリックし、ポップアップウィンドウからOCT画像を呼び出します。次に、メディアコンテナ内の画像をダブルクリックして画面に表示します。次に、レイヤー検出をクリックして、網膜の自動レイヤー化を完了します。解析用に準備したレイヤーの両側の分割線を選択したら、[レイヤーの編集] をクリックして間隔と制限範囲を調整します。次に、赤い円を移動して線を変更します。網膜積層の厚さを測定するには、マーカーの測定ボタンをクリックし、分析する層の分割線を選択して、OCT画像に層の境界を表示します。次に、[レイヤーに接続]を選択し、移動中に接続したままにします。次に、結果を表示する領域を選択し、OCT画像上で解析する位置をクリックして測定線を表示します。次の列をクリックすると、後続の測定と以前のデータが表示されます。網膜の厚さ全体を測定するには、右上隅のリストで1行目と7行目を選択します。次に、視乳頭の端にある網膜構造の出現から、水平定規上で200マイクロメートル間隔で4つの値を測定します。C57BL/6マウスのOCT画像は、明確な境界を有する様々な網膜層を示した。対照的に、Vldlrノックアウトマウスは異常な過反射病変を示した。OCT画像は、Vldlrノックアウトマウスの網膜表面にいくつかの中央反射バンドを示しました。これらのバンドは網膜血管に接着し、不完全な硝子体剥離のOCT特性と一致した。1つの過反射病変が網膜下腔に現れ、外顆粒層に拡がったが、外叢状層を突破しなかった。この病変のOCT外観は病理学的所見と一致した。病理部では,新生血管が網膜色素上皮層,感光体内外層,外限界膜を突破し,外顆粒層には侵入したが,外叢状層には侵入しなかった。網膜下腔に位置する過反射病変の別のバンドは、外顆粒層を伴わなかった。病理学的所見と一致して、この網膜下血管新生は外限界膜を突破しなかった。両群の右眼の網膜厚を後極の側頭、鼻、上、下方向で比較したところ、Vldlrノックアウトマウスの網膜厚はC57BL/6Jマウスよりも有意に低いことが明らかになりました。プロトコルを実行するときは、角膜に100Dコンタクトレンズを適切に配置してから、画像の位置を調整します。スキャン場所を選択し、撮影前に画像をオーバーレイします。この手順に続いて、前眼部OCTが実現可能であり、プリセットレンズと角膜の間の距離を変更する必要があります。角膜、前房、虹彩、毛様体、水晶体を示すことができます。この技術は、小動物モデルにおける網膜症研究のための重要なツールを提供し、基礎研究における網膜症の非侵襲的な検出と測定を促進します。
