quail chorioallantoic membrane - a tool for photodynamic diagnosis and therapy

日本のウズラの卵子栽培を準備する方法と、癌や微生物感染症の光線力学診断と治療のために絨毛尿膜またはCAMを使用する方法を紹介します。この方法の利点は、急速な発達成長、小さいサイズ、組織への良好なアクセス、および取り扱いの容易さです。また、動物研究を行うための3Rルールの原則を尊重しています。膀胱、肺、胎盤などの粘膜と構造的に類似しているため、潜在的な薬物のin vivo試験、毒性試験、または移植試験に適しています。手順を実演するのは、私の研究室の博士課程の学生であるバルボラ・クンデコワとマイリンダ・メタです。開始するには、孵卵開始前の最大4〜5日間、新鮮または摂氏10〜15度で保存された受精ウズラの卵を使用します。清潔で損傷のない卵のみを使用し、湿度50〜60%、摂氏37.5度の強制ドラフトインキュベーターで卵を約54時間インキュベートし、卵の回転をオフにして水平に置きます。卵を回転させずに70%エタノールで卵の表面を消毒します。次に、手袋を着用した状態で滅菌層流キャビネット内で、小型の滅菌外科用ハサミを使用して卵殻を開き、内容物を6ウェル培養プレートに移します。各卵の後、はさみを70%エタノールで消毒します。CAMの乾燥を防ぐために湿度が不可欠であるため、6ウェルプレートの隙間に約5ミリリットルの滅菌水を追加します。次に、先端が不適切な胚や未受精卵を真空吸引器で吸引し、湿度80〜90%、温度37°Cを保ちながら、さらなる実験が行われるまでインキュベーターに入れます。CAMが十分に発達したら、通常は胚の7日目から、主要な血管を避けながら、小さな毛細血管に沿ってCAM表面に滅菌されたシリコンリングを配置します。無菌条件下で、適量のヒペリシン溶液30マイクロリットルをシリコーンリングに適用し、胚を湿度80〜90%、摂氏37度のインキュベーターに保管します。光線力学的診断を行うには、紫色励起光を使用してCAMを照明し、ヒペリシン投与後に異なる時間間隔でデジタルカメラでヒペリシンおよびCAM組織および腫瘍細胞の蛍光を記録します。研究で白色光でのCAMの画像が必要な場合は、ヒペリシン投与前と組織固定の直前に、光がヒペリシンの光活性化を引き起こすため、実験終了時にCAMを記録します。ヒペリシン塗布後に光線力学療法を行うには、レーザービーム用の光ファイバーの下にCAMを配置して、シリコンリング内の領域全体をカバーします。in vivo照射を行った後、この特定のケースでは、285ミリワット/センチメートル四方のフルエンスレートで405ナノメートルのレーザー光を使用して、光線力学処理の前後に白色光および/または蛍光光を使用してCAMを記録します。CAM組織をPBS中の4%パラホルムアルデヒドを含む培養プレートに最小2時間、最大一晩固定し、パラホルムアルデヒドを除去し、CAMからシリコンリング内の組織の一部を慎重に切り取ります。これらの手順はすべて、ドラフト内で行う必要があります。CAM組織から切断した部分を水中で10分間洗浄し、続いてCAM組織を70%エタノールで3分間、エオジン溶液で2分間、スロシン96%エタノールで5分間、100%エタノールで5分間、キシレンに2回、新しいシャーレに入れて10分間、上昇アルコール系列で脱水します。続いて、へらまたは細いブラシを使用して、できるだけ早くサンプルをペトリ皿の溶解したパラフィンに移します。24時間後、組織を組織型に入れ、パラフィン包埋剤で満たして冷蔵庫で固化させ、固化したCAMを包埋媒体から切り取り、トレイ内で90度回転させ、再び包埋剤を充填して固化させます。PDT誘発損傷を決定するための組織病理学的分析のために、ミクロトーム上に5〜10マイクロメートルの切片を準備します。組織学用の凍結CAM切片を準備するには、天然または4%パラホルムアルデヒド固定CAMをスライドガラスに注意深く取り付け、埋め込み型をOCTで半分まで満たし、液体窒素またはドライアイスとエタノールの混合物で凍結します。凍結後、CAMをスライドガラスから凍結したOCT培地の上部に慎重に傾けてスライドさせます。再び型に入れ、OCT培地で覆い、前述のように凍結する。CAM血管系の分析には、CAMサンプル全体が必要です。層流キャビネットで、4%パラホルムアルデヒドとPBS中の2%グルタルアルデヒドの予熱固定溶液でCAMをオーバーフローさせ、48時間後に固定溶液を除去します。次に、マイクロハサミと細かいブラシでCAMを胚から慎重に分離し、PBSで洗浄した後、洗浄したCAMをスライドガラスにマウントし、ゆっくりと乾燥させます。その後、トランスイルミネーターのデジタルカメラを使用して、均質な白色光の光源としてスライドを撮影します。CAM表面上の腫瘍の位置は、ヒペリシンの添加時に白色光および蛍光光の下で視覚化された。組織学的解析では、浮腫と膜肥厚を伴う異常な扁平上皮細胞の同心円構造が正常組織に侵入しました。PDTによる処理後、リング内および周辺領域の血管密度に明らかな差が観察された。光増感剤の存在なしのレーザー放射は、何の治療もせずに対照に匹敵する損傷を引き起こさなかった。ヒペリシンとの3時間のインキュベーション後、レーザー照射は、血管系に広範な損傷を伴う凝集ヒペリシンのモノマー化によって引き起こされる蛍光をもたらした。組織学的解析により、未処理のコントロールCAMの厚さは比較的均一であることが明らかになりました。しかし、PDT処理により、CAMはより薄く、より壊れやすくなりました。ウズラのex ovo CAMアッセイを準備する方法とそれを使用する方法を示しました。ガイドラインを維持しながら、この方法論は習得が容易であり、迅速な結果を証明できます。
