1. 准备清算解决方案
<ol><li>要制备CS溶液，将10%（w / v）木糖醇，15%（w / v）脱氧胆酸钠和25%（w / v）尿素溶解在磁力搅拌器上的蒸馏水中。 注意：脱氧胆酸钠粉末应在通风室中称量，因为它很容易漂浮在空气中。CS可以在室温下在黑暗中储存超过1年。</li>
	<li>为了制备CSA溶液，将亚硫酸钠（50mM终浓度）加入到上述获得的CS溶液中。 注意：使用前将亚硫酸钠加入CS溶液中，因为还原剂很容易失活。</li>
</ol>2. 固定液的制备
<ol><li>将40毫升无菌水转移到锥形管中，加入2克多聚甲醛。加入200μL的2 N NaOH以增加溶液的pH值。关闭并用parafilm密封管后，在60°C下孵育，偶尔倒置，直到所有东西都溶解。</li>
	<li>将溶液冷却至室温后，加入5mL10x磷酸盐缓冲盐水（PBS）以将pH值调节至7.4。加入灭菌水以补充体积至50 mL。 注意：最好使用新鲜制备的固定溶液。该溶液也可以在-30°C下储存数月。</li>
</ol>3. 样品固定
<ol><li>将植物样品浸入微管的固定溶液中（图1A），确保固定溶液的体积是样品体积的五倍以上。</li>
	<li>用石膏膜密封微管，并用针头打孔。不要将试管打开，因为在真空减压过程中存在样品溢出的风险。</li>
	<li>将微管置于干燥器中，并缓慢调节真空度（〜690mmHg），以便从样品中逐渐出现气泡（图1B-C）。抽真空干燥器后关闭真空泵。在室温下保持微管不受干扰30分钟。</li>
	<li>小心地通风干燥器，以防止干扰样品。再次打开真空泵，并在抽空干燥器后将其关闭。在室温下保持微管不受干扰30分钟。 注意：在通风干燥器时应注意防止损坏样品。通过两次真空处理，固定溶液对样品的渗透得到增强。进一步的真空处理有助于将固定溶液渗透到较厚的样品中。</li>
	<li>小心打开干燥器，不要将固定溶液撞到微管中。使用微量移液管，取出固定溶液并加入1x PBS。储存1分钟后，将旧的PBS更换为新的1x PBS（图1D）。</li>
</ol>4. 清算
<ol><li>除去PBS后，加入五倍于澄清溶液的样品体积。</li>
	<li>用石膏膜密封微管，并用针头打孔。将样品放入干燥器中，如步骤3.3中抽真空，然后关闭真空泵。在室温下保持微管不受干扰60分钟。</li>
	<li>轻轻打开干燥器。用parafilm关闭微管，并将其储存在室温下的黑暗中，以避免荧光蛋白的光漂白。每1-2天倒置微管以加速清除过程。</li>
	<li>当清除溶液变为绿色时，用新的清除溶液替换它，直到溶液保持无色（图1E-H）。</li>

5. 化学染料染色
<ol><li>加入微管中，加入Hoechst 33342（终浓度为10μg/ mL）进行核染色或Calcofluor White（终浓度为1mg / mL）用于细胞壁染色并等待1小时。除去染料溶液后，用新鲜的清除溶液洗涤样品1小时。 注意：过夜染色和洗涤可以改善荧光染料对组织的渗透并减少背景荧光。各种荧光染料与CS溶液相容，例如碱性紫红素9（木质素），Auramine O9（木质素，苏伯林和角质素），Nile Red9（苏贝林），Direct Yellow 969，Direct Red 239和SR220010，11（细胞壁）。</li>
</ol>6. 观察
<ol><li>用剃须刀片切割硅橡胶板，为垫片准备框架（图2A）。 注：根据样品的厚度调整硅橡胶片的厚度。由于用清除溶液处理的样品是软的，如果它们直接用盖玻片覆盖，它们将被损坏。</li>
	<li>将硅胶框架放在盖玻片上（例如，25 x 60 mm）（图2B）。将处理后的样品置于框架内，并加入约100μL清除溶液以除去框架中的任何气泡。用另一个盖板玻璃（18 x 18 mm或24 x 24 mm）盖住，以防止清除溶液蒸发（图2C）。</li>
	<li>在荧光显微镜下观察样品。观察后，将样品返回微管中取出的澄清溶液，并在室温下在黑暗中储存。</li>

这种方法包括固定，洗涤和清洁。固定是该协议中的关键步骤。如果在PFA固定后未观察到荧光蛋白，则在用清除溶液处理后不会观察到荧光蛋白。PFA溶液渗透到组织中至关重要，但不建议进行高真空处理，因为它会破坏细胞结构。应针对每种类型的组织和物种优化真空条件和固定期。建议即使在固定后也要检查荧光蛋白。虽然样品通常在室温下固定30-60分钟，但它们可以在4°C下固定更长的时间（过夜或更长时间）。
