1. 청산 용액의 준비

</ol>2. 고정제액의 제조
<ol><li>멸균수 40mL를 원뿔형 튜브에 넣고 파라포름알데히드 2g을 첨가한다. 200 μL의 2 N NaOH를 첨가하여 용액의 pH를 증가시킨다. 튜브를 파라필름으로 닫고 밀봉한 후, 모든 것이 용해될 때까지 가끔 역전하여 60°C에서 배양한다.</li>
	<li>용액을 실온으로 냉각시킨 후, 5 mL의 10x 인산완충식염수(PBS)를 첨가하여 pH를 7.4로 조정하였다. 멸균수를 첨가하여 부피를 50mL까지 구성하십시오. 참고 : 갓 준비된 고정 용액이 선호됩니다. 용액은 또한 수개월 동안 -30°C에서 저장될 수 있다.</li>
</ol>3. 샘플의 고정

5. 화학 염료 염색
<ol><li>마이크로튜브에 핵 염색을 위한 Hoechst 33342(최종 농도 10 μg/mL) 또는 세포벽 염색을 위한 Calcofluor White(최종 농도 1mg/mL)를 추가하고 1시간 동안 기다립니다. 염료 용액을 제거한 후, 샘플을 1 h 동안 신선한 클리어링 용액으로 세척한다. 참고: 하룻밤 동안 염색하고 세척하면 조직으로의 형광 염료 침투를 개선하고 배경 형광을 줄일 수 있습니다. 다양한 형광 염료는 베이직 Fuchsin9 (리그닌), 오라민 O9 (리그닌, 수베린 및 커틴), 나일 레드9 (수베린), 다이렉트 옐로우 969, 다이렉트 레드 239, 및 SR220010,11 (세포벽)과 같은 CS 용액과 상용성이다.</li>
</ol>6. 관찰
<ol><li>실리콘 고무 시트를 면도날로 절단하여 스페이서용 프레임을 제조하였다(그림 2A). 참고 : 샘플의 두께에 따라 실리콘 고무 시트의 두께를 조정하십시오. 클리어링 용액으로 처리 된 샘플은 부드럽기 때문에 커버 글라스로 직접 덮으면 손상됩니다.</li>
	<li>실리콘 프레임을 커버 글래스(예: 25 x 60mm) 위에 놓습니다(그림 2B). 처리 된 샘플을 프레임 내에 놓고 ~ 100 μL의 클리어링 용액을 첨가하여 프레임 내의 기포를 제거하십시오. 클리어링 용액의 증발을 방지하기 위해 다른 커버 글래스(18 x 18mm 또는 24 x 24 mm)로 덮으십시오(그림 2C).</li>
