気道の管腔表面は、主に多繊毛、クラブ、杯、および基底幹細胞1,2からなる上皮の層によって裏打ちされている。上皮層は、肺の一次防御機構として機能し、吸入された病原体、微粒子、または化学ガスから基礎となる気道組織を保護する生物物理学的障壁として作用する。これは、細胞間タイトジャンクション形成、粘液繊毛クリアランス、抗菌および抗酸化分泌を含む複数のメカニズムを介して気道組織を保護する3,4。欠損した気道上皮は、慢性閉塞性肺疾患(COPD)5、原発性毛様体ジスキネジー(PCD)6、および嚢胞性線維症(CF)7などの壊滅的な呼吸器疾患と関連している。
肺オンチップ(LOC)技術の進歩は、ヒトの肺の発達を研究し、さまざまな肺疾患をモデル化し、厳しく規制されたin vitro環境で新しい治療材料を開発する機会を表しています。例えば、気道上皮および内皮を、薄い多孔質膜の反対側で培養して、肺組織を交換するガスを模倣することができ、忠実な疾患モデリングおよび薬物検査を可能にする8。同様に、COPD9および嚢胞性線維症10などの気道疾患をin vitroでモデル化するために、in vitro疾患モデルが作成されている。しかしながら、LOC装置の主要な課題は、肺組織の複雑な三次元(3D)アーキテクチャおよび動的細胞−組織マトリックス相互作用をインビトロ11で反復することである。
近年、エクスビボ肺組織の操作を可能にする革新的な組織工学方法論が開発されている12。これらの方法論を用いて、裸化された同種異系または異種異系組織移植片は、化学的、物理的、および機械的処置13を介して肺組織から内因性細胞を除去することによって調製され得る。さらに、脱細胞化肺足場材中の保存された天然組織細胞外マトリックス(ECM)は、移植細胞が付着、増殖、分化するための生理学的模倣的な構造的、生化学的、および生体力学的手がかりを提供する14,15。
ここでは、外植ラット気管組織の インビトロ 組織操作および培養を可能にするLOCおよび組織工学技術を組み合わせて作成されたイメージング誘導バイオリアクターシステムが報告されている。この気道組織バイオリアクターを使用して、プロトコルは、気道組織の根底にある上皮下細胞および生化学的成分を破壊することなく、内因性上皮細胞の選択的除去を実証する。次に、細胞担持コラーゲンIプレゲル溶液を点眼することにより、間葉系幹細胞(MSC)などの新たに播種した外来性細胞の、むき出し気道内腔上の均質分布および瞬間沈着を示す。また、バイオリアクターに組み込まれたマイクロ光学イメージング装置を用いて、上皮除去や内因性細胞送達時の気管内腔の可視化も行われる。また、気管および新たに移植された細胞は、4日間、顕著な細胞死および組織分解なしにバイオリアクター内で培養され得ることが示された。本研究で用いたイメージング対応バイオリアクタープラットフォーム、薄膜ベースの脱上皮化技術、細胞送達法は、 in vitro 疾患モデリングや薬物スクリーニングのための気道組織の生成に有用であると想定しています。
バイオリアクターは、プログラム可能なシリンジポンプ、灌流ポンプ、および単離ラット気管を培養するための人工呼吸器に接続された長方形のチャンバを含む。バイオリアクターは、気管または組織培養チャンバに接続された入口および出口を備え、気管の内部および外部空間に試薬(例えば、培養培地)を別々に供給する(図1)。カスタムメイドのイメージングシステムを使用して、 体外培養ラット気管の内部を細胞レベルで視覚化することができます(図2)。気管の内因性上皮は、洗剤ベースの脱細胞化溶液の点眼とそれに続く振動支援気道洗浄 によって 除去される(図3)。I型コラーゲンなどのヒドロゲル溶液は、外因性細胞を退毒した気管内腔を横切って均一かつ瞬時に播種するための送達ビヒクルとして使用される(図4)。バイオリアクターの構築と実験の実施に使用されるすべての材料は、 材料表に記載されています。
