Gouden nanodeeltjes (AuNP's) hebben een groot potentieel getoond als biocompatibele beeldvormingsondes, bijvoorbeeld als effectieve oppervlakte-verbeterde Raman-spectroscopie (SERS) substraten in verschillende biomedische toepassingen. Belangrijke toepassingen zijn gebieden zoals biosensing, bioimaging, oppervlakte-verbeterde spectroscopieën en fotothermische therapie voor kankerbehandeling1. Bovendien is het onderzoeken van AuNP's in levende systemen cruciaal voor het beoordelen en begrijpen van de interactie tussen AuNP's en biologische systemen. Er zijn verschillende analytische technieken, waaronder Fourier transform infrared (FTIR) spectroscopie2, laserablatie inductief gekoppelde massaspectrometrie (LA-ICP-MS)3 en magnetische resonantie beeldvorming (MRI)4 die met succes zijn gebruikt om de verdeling van AuNP's in weefsels te onderzoeken. Niettemin hebben deze methoden verschillende nadelen, zoals tijdrovend zijn en complexe monstervoorbereiding3 met zich meebrengen, die lange verwervingstijden vereisen, of het ontbreken van sub-micron ruimtelijke resolutie 2,4.
In vergelijking met conventionele beeldvormingstechnieken biedt niet-lineaire optische microscopie verschillende voordelen voor het onderzoeken van levende cellen en AuNP's: de niet-lineaire optische microscopie bereikt een diepere beelddiepte en biedt intrinsieke 3D optische sectiemogelijkheden met behulp van bijna-IR ultrasnelle lasers. Met de significante verbetering van de beeldvormingssnelheid en detectiegevoeligheid is aangetoond dat twee-foton geëxciteerde fluorescentie (TPEF)5,6,7 en tweede harmonische generatie (SHG)8,9,10 microscopie niet-invasieve beeldvorming van endogene biomoleculen in levende cellen en weefsels verder verbeteren. Bovendien is het mogelijk om met behulp van nieuwe pomp-sonde niet-lineaire optische technieken zoals transiënte absorptie (TA)11,12,13,14 en gestimuleerde Raman-verstrooiing (SRS)15,16,17,18, labelvrij biochemisch contrast van cellulaire structuren en AuNP's af te leiden. Het visualiseren van AuNP's zonder het gebruik van extrinsieke labels is van groot belang, omdat chemische verstoringen van de nanodeeltjes hun fysische eigenschappen en dus hun opname in cellen zullen wijzigen.
Dit protocol presenteert de implementatie van een Spectral Focusing Timing and Recombination Unit (SF-TRU) module voor een dual-golflengte pulslaser, waardoor snelle multimodale beeldvorming van AuNP's en kankercellen mogelijk is. Het onderzoeken van gouden nanodeeltjes (AuNP's) in levende systemen is essentieel om de interactie tussen AuNP's en biologische weefsels te onthullen. Ons protocol presenteert een multimodale, niet-lineaire, optische microscopie en past deze toe om chemisch specifieke beeldvorming van gouden nanodeeltjes in kankercellen uit te voeren. Het belangrijkste voordeel van ons beeldvormingssysteem is dat het kan worden gebruikt om het biomoleculaire contrast van cellulaire structuren en gouden nanodeeltjes op een multimodale manier te beoordelen. Schakel om te beginnen de stekkers van alle apparatuur in, behalve de afstandsbedieningskast.Schakel vervolgens de achterkant van de doos in en druk op Start operation nadat het afstandsbedieningslampje blauw wordt op de bedieningskast. Schakel daarna de pc van de laserscanmicroscoop in en voer de confocale microscoopsoftware uit. Controleer vervolgens het lichtpad van de microscoop met behulp van de computersoftware en bedien de focus via het aanraakscherm of de afstandsbedieningsknoppen of de computersoftware.Selecteer de gewenste afbeeldingsinstellingen in de software, waaronder Zoom, de afbeeldingsgrootte in pixels en pixel dwell-tijd, terwijl u ervoor zorgt dat de pixel dwell time groter is dan de integratietijd. Plaats een druppel gedestilleerd water bovenop het waterdompelobject dat de laserstralen in het monster en tussen de condensor en het monster richt voor beeldvorming. Stel vervolgens de hoogte van de condensor in op ongeveer een millimeter boven het monster om de maximale hoeveelheid licht te verzamelen en verplaats de SRS-detector die op een beweegbare houder is geplaatst uit het bundelpad om zich met wit licht op het monster te concentreren.Selecteer met behulp van de software-instellingen de pompstraal en verander de lasergolflengte in 802 nanometer voor CH-trillingen en 898 nanometer voor amide I-piek, rekening houdend met grote veranderingen in de Raman-shift. Om kleine aanpassingen in de Raman-shift te bereiken, scant u de vertragingsfase in de SFTRU-eenheid zoals beschreven in het manuscript. Voor de hyperspectrale dataset selecteert u het tabblad Trigger, klikt u op Start by ttl trigger aan en selecteert u vervolgens Elk frame activeren.Stel vervolgens op het tabblad Serie de tijdreeks in op en de Z-serie uit. Stel bovendien het aantal frames in de tijdreeks in om overeen te komen met het aantal stappen in de ATM-software. Ga hierna naar het tabblad Uitvoeren in de ATM-software en stel de vertragingsfaseposities voor CH in op 90,25 en 92,25, terwijl voor het amide I-gebied op 89 en 91 in millimeters, overeenkomend met de start- en stoppositie van de hyperspectrale scan, terwijl u ervoor zorgt dat het aantal stappen overeenkomt met het aantal frames in de computersoftware.Nadat u de juiste instellingen hebt gecontroleerd, start u de hyperspectrale stack in de computersoftware door eerst naar Acquire te gaan en vervolgens op Scan starten te klikken. Klik vervolgens in de ATM-software op Start, waarmee een reeks afbeeldingen wordt verzameld met incrementele verhogingen van de vertragingsfasepositie tussen de geselecteerde start- en stoppositie. Neem vervolgens een tijdreeks afbeeldingen op verschillende posities in de vertragingsfase om een hyperspectrale scan te genereren.Met behulp van de analoge eenheid verzendt en ontvangt u ttl-signalen van en naar het SFTRU-systeem om beeldacquisitie en beweging van de vertragingsfase te synchroniseren. Voor het schakelen tussen beeldvorming in het CH-trillingsgebied en het trillingsgebied amide I, wijzigt u de pompgolflengte in de lasersoftware van 802 nanometer naar 898 nanometer. Wijzig verder de Stokes-dispersie-instelling in de ATM-software van 30 millimeter naar 5 millimeter.Pas de onderste knop op de spiegelbevestiging aan om een kleine aanpassing aan de spiegel in het verspreidingspad van de pompbundel in de SFTRU-box te maken door de spiegelhoek met een incrementele hoeveelheid te wijzigen. Om de chemische selectiviteit van de polystyreenmicrosferen te valideren, werden hyperspectrale gestimuleerde Raman-verstrooiing en spontane Raman-spectra geregistreerd. De spectra waren identiek, behalve het verschil in de relatieve intensiteit.De gestimuleerde Raman-verstrooiingsbeeldvorming van 4T1-kankercellen werd uitgevoerd op 2, 852, 2, 930 en 2, 968 golfnummers, overeenkomend met de koolstof-waterstof uitrekkende trillingsband van lipide-, eiwit- en DNA-biomoleculen. Multimodale beeldvorming van 4T1 kankercellen gedoseerd met gouden nanodeeltjes werd uitgevoerd om de nanodeeltjesverdelingen in kankercellen te onderzoeken. De verkregen gestimuleerde Raman-verstrooiingsbeelden werden verkregen voor CH2- en CH3-kanaal, LysoTracker fluorescerende sondes en off-resonantie gestimuleerd Raman-verstrooiingskanaal.Het belangrijkste om te onthouden is om het trillingsgebied, de pompreferentie, in de lasersoftware en ook de Stokes-dispersie-instelling tijdens het protocol te wijzigen. Dit systeem kan eenvoudig worden overgeschakeld van femtoseconde naar picosecondegebied zonder de optische uitlijning te beïnvloeden. Deze mogelijkheid stelt ons in staat om multifoton coherente Raman-verstrooiing en pompsondespectroscopie en hun toepassingen uit te voeren.Dit multimodale beeldvormingsplatform biedt nieuwe inzichten in nanogeneeskunde en maakt de weg vrij voor moleculaire beeldvorming van cellen, weefsels en gouden nanodeeltjes.
