在过去的30年里，Hyun-Joon Ha教授的团队研究了氮丙啶的化学性质，并以不对称的方式开发了含氮有价值的分子的合成。在这个视觉实验中，我们在这里展示了对映哆嗪-2羧酸酯的合成及其在双咪唑胺B，D和表皮异毒羯碱生物碱合成中的应用。该方法显示了通过选择性结晶过程大规模制备2R和对映腥醇L-甲醇酯和2S氮丙啶-2羧酸酯的方法。在这个视觉实验中，我们展示了对映卺纯氮丙啶-2羧酸酯的制备及其在双咪咪胺B，D和表皮异毒羯碱生物碱合成中的应用。本文中的方案是大规模制备非活化氮丙啶的简单方法，其中包括结晶，过滤和柱层析过程等简单步骤。首先在氮气气氛下将2， 3-二溴丙烷L-甲醇酯1A和磁力搅拌棒加入到烘干的250毫升双颈圆底烧瓶中。使用气密注射器向反应瓶中加入60毫升无水乙腈。然后，在冰浴上将反应混合物冷却至零摄氏度，并将反应混合物搅拌五分钟。在相同温度下将碳酸钾加入反应混合物中，并保持搅拌30分钟。接下来，在室温下以滴加方式加入2R-苯基乙胺，并使反应混合物搅拌12小时。然后，使用9：1体积的乙酸己烷乙酯作为淋洗液，用薄层色谱监测反应的进度。反应完成后，在滤纸上过滤混合物，并将水加入有机滤液中。使用分离漏斗用乙醚提取有机层两次。在干燥超过7.5克无水硫酸钠的组合有机提取物后，使用旋转蒸发器真空浓缩有机提取物，以获得含有两种异构体的对映异构手性aziridine的粗混合物。接下来，在70毫升甲醇中加入8.7克手性氮丙啶L-甲醇酯1衍生物的粗混合物到烘干的250毫升单颈圆底烧瓶中。然后，使用热水浴将反应混合物加热至70摄氏度，并将反应混合物冷却至零下10摄氏度，直到形成固体晶体。在滤纸上过滤固体化合物，得到2.2克R，1R，2S，5R，2-异丙基5-甲基环己基，1R1苯乙基氮丙啶-2羧酸酯，并使用旋转蒸发器再次真空浓缩滤液溶液。将剩余的反应混合物溶解在50毫米乙醇中后，在零下10摄氏度下重新结晶，得到1.2克R，1R，2S，5R 2-异丙基5-甲基环己基1R1苯乙基氮丙啶-2羧酸酯，此时使用其他醇乙醇与甲醇相同。然后在滤纸上过滤重新结晶的产品，并使用旋转蒸发器将剩余的粗5.3克滤液溶液完全真空浓缩。然后，加入50毫升戊烷烃类溶剂，同时将剩余的反应溶液保持在零下15摄氏度和近1.9克S，1R，2S，5R，2-异丙基5-甲基环己基1-苯基乙基氮丙啶-2羧酸酯的固体化合物。一旦晶体准备就绪，使用旋转蒸发器再次真空浓缩溶液，并将其溶解在30毫升戊烷烃溶剂中。在零下15摄氏度下再次重新结晶，得到8克S、1R、2S、5R 2-异丙基、5-甲基环己基1R1苯乙基氮丙啶-2羧酸酯两个素数。在氮气气氛下，将R、1R、2S、5R、2-异丙基5-甲基环己基1R1苯基乙基氮丙啶-2羧酸酯和磁力搅拌棒放入烘干的25毫升双颈圆底烧瓶中。使用密闭注射器向反应瓶中加入1.8毫升乙醇，并在室温下搅拌。然后，加入碳酸钾并使其在室温下搅拌两天，并使用薄层色谱法监测反应过程。反应完成后，在滤纸上过滤混合物，并将水加入有机滤液中。使用分离漏斗，用二氯甲烷提取有机层两次。将组合的有机提取物干燥超过三克无水硫酸钠后，使用旋转蒸发器真空浓缩。接下来，通过在硅胶上进行正相柱层析纯化粗产物，得到R-乙基-1 R1苯基乙基氮丙啶-2羧酸酯的纯产物。为了合成叠氮化物开孔化合物，将500毫克手性S-乙基1R1苯基乙基氮丙啶-2羧酸酯和磁力搅拌棒转移到开雾气氛下的50毫升双颈圆底烧瓶中。然后，向反应混合物中加入50%乙醇水溶液。将反应混合物冷却至零摄氏度后，滴加浓硫酸以保持pH 4.0并搅拌五分钟。接下来，在相同温度下加入叠氮化钠，同时搅拌反应混合物10分钟，然后在室温下加热。然后，在室温下加入氯化铝六水合物作为催化剂，并使其再搅拌三小时。使用薄层色谱法监测反应过程，并用两份20毫升饱和碳酸氢钠淬灭反应混合物。然后，用乙醇在硅藻土垫上过滤粗混合物，并使用旋转蒸发器真空浓缩反应混合物。使用分离漏斗提取有机层二氯甲烷两次。将合并后的有机层用五克无水硫酸钠干燥五分钟。使用旋转蒸发器真空浓缩粗有机层，以获得粗叠氮化物产品。用40%乙酸乙酯或己烷洗脱，在硅胶上用正相柱层析纯化粗产物，得到490毫克叠氮化物开孔化合物作为粘稠液体。2， 3-二溴丙烷L-甲酯和1R-苯乙胺导致形成R、1R、2S、5R、2-异丙基5-甲基环己基1R1苯基乙基氮丙啶-2羧酸酯和S，1R，2S，5R 2-异丙基5-甲基环己基1R1苯基乙基氮丙啶-2羧酸酯。甲醇产生对映异构体形式的R，1R，2S，5R，2-异丙基甲基环己基1R1苯基乙基氮丙啶-2羧酸酯，而N-戊烷产生S，1R，2S，5R，2-异丙基5-甲基环己基1R1苯基乙基氮丙啶-2羧酸酯。使用镁或碳酸钾与甲醇和乙醇等醇进行酯交换反应，得到手性氮丙啶-2羧酸盐的相应烷基酯，通过叠氮化物亲核试剂通过叠氮化物亲核试剂进行Regio-选择性环开口，导致双乙酰胺B和双乙酰胺D的总合成。 立体选择性变换产生减去表皮异毒羯碱。通过选择性结晶工艺，成功地制备了对映哆甲基和对映体甲基和2S氮丙啶-2羧酸酯。在制备对映草碱-2羧酸盐后，我们在这里展示了其在双咪唑胺B，D和表皮异毒蕈碱生物碱的总合成中的应用。
