뼈는 뼈 형성 세포, 조골 세포 및 뼈 흡수 세포인 파골 세포로 구성된 조밀한 결합 조직입니다. 골절 치유는 조골 세포와 파골 세포의 조정 된 작용에 의해 뼈 결함을 재생시키는 생리적 과정입니다1. 골절이있을 때, 골절 부위의 조골 세포 및 파골 세포 활성은 뼈 치유를 결정하는 중요한 요소 중 일부입니다2. 골절 치유가 정상적인 과정에서 벗어나면 지연된 결합, 부결합 또는 비유합이 발생합니다. 골절은 9개월 동안 골절의 유합이 실패하고 최근 3개월 동안 수리가 진행되지 않은 경우 비유합 상태라고 합니다3. 모든 골절의 약 10%-15%는 비유합으로 진행될 수 있는 수리 지연을 경험합니다4. 모든 골절에 대한 비유합률은 5%-10%이며 관련된 뼈와 골절 부위5에 따라 다릅니다.
골절 비유합 치료를 위한 현재 요법은 수술 및/또는 의료 양식을 포함합니다. 현재 골절의 지연 또는 비결합은 뼈 이식과 같은 수술 전략으로 극복할 수 있습니다. 그러나 뼈 이식에는 이식 조직의 가용성, 기증자 부위 통증, 이환율 및 감염과 같은 한계와 합병증이 있습니다6. 의학적 치료는 뼈 형태 형성 단백질(BMP) 및 테리파라타이드(파라트호르몬 유사체)와 같은 골동화 약물로 구성됩니다. 현재 사용되는 골동화 작용제는 골절 복구를 증가시킬 가능성이 있지만 과도한 비용이나 바람직하지않은 부작용과 같은 제약이 있습니다7. 따라서 뼈 치유를 위한 비용 효율적인 비수술적 대안을 식별할 수 있는 여지가 있습니다. 약물의 뼈 치유 잠재력은 처음에는 시험관 내 연구에 의해 결정될 수 있지만 최종 개념 증명을 위해서는 생체 내 연구가 필요합니다. 뼈 치유를 향상시키는 것으로 알려진 약물은 시험관 내에서 평가되어야하며, 유망한 것으로 밝혀지면 생체 내 동물 모델 연구에 사용할 수 있습니다. 약물이 생체 내 모델에서 골 형성 및 리모델링을 촉진하는 것으로 입증되면 다음 단계 (즉, 임상 시험)로 진행할 수 있습니다.
동물의 골절 치유를 평가하는 것은 인간 실험을 거치기 전에 뼈 치유를 위해 도입 된 새로운 약제를 평가하기위한 논리적 단계입니다. 골절 치유의 생체내 동물 모델 연구를 위해, 설치류는 점점 더 인기 있는 모델8이 되었다. 설치류 모델은 낮은 운영 비용, 제한된 공간 필요성 및 뼈 치유에 필요한 시간 감소로 인해 관심이 증가하고 있습니다9. 또한 설치류는 광범위한 항체와 유전자 표적을 가지고있어 뼈 치유 및 재생의 분자 메커니즘에 대한 연구를 가능하게합니다10. 합의 회의는 다양한 작은 동물 뼈 치유 모델을 종합적으로 강조하고 뼈 치유에 영향을 미치는 다양한 매개 변수에 초점을 맞추고 몇 가지 작은 동물 골절 모델 및 임플란트를 강조했습니다11.

protocol for developing a femur osteotomy model in wistar albino rats
