異常なタンパク質恒常性は、タンパク質の合成と分解の不均衡によって引き起こされます。いくつかの疾患は、タンパク質恒常性の変化に関連しています。いくつかの疾患の特徴は、異なる細胞内位置および脳領域に凝集体が存在することです。タンパク質の恒常性は、疾患において重要であるだけでなく、正常な臓器および細胞機能においても重要な役割を果たします1。例えば、タンパク質合成は、タンパク質合成を阻害する化学阻害剤の使用によって決定されるように、多くの形態のニューロン可塑性2,3に必要である4。しかし、学習と記憶をサポートするためにプロテオームがどの細胞型で変化しているかは明らかではなく、各細胞型のどの特定のタンパク質がそれらの合成または分解において増加または減少するかも理解されていません。したがって、タンパク質の恒常性を包括的に研究するには、特定の細胞型に由来するプロテオームを区別する能力が必要です。実際、多細胞環境で起こる細胞プロセスを研究するための細胞種特異的プロテオームの同定は、プロテオミクスにおける重要なハードルでした。このため、MetRS*発現と生体直交法を組み合わせた技術を開発し、細胞型特異的プロテオームを同定および精製する効果的な方法であることが証明され、このギャップを埋めました5,6,7。
