Ce protocole propose une combinaison d’hybridation in situ et d’immunohistochimie, une étape importante dans les expériences de colocalisation sur des embryons de poisson zèbre. Le poisson-zèbre est un puissant modèle vertébré pour les études de développement et de génétique 1,2. Le poisson-zèbre et l’homme partagent une homologie génétique élevée (70% des gènes sont partagés avec le génome humain), ce qui permet son utilisation comme modèle pour les maladies humaines3. Chez le poisson zèbre, il est assez courant de détecter les schémas d’expression de deux gènes et leur relation spatiale. L’immunohistochimie a été utilisée pour la première fois en 1941 pour détecter les agents pathogènes dans les tissus infectés en appliquant des anticorps marqués FITC4. La protéine cible dans la section tissulaire est d’abord marquée avec un anticorps primaire, puis la section est marquée avec un anticorps secondaire contre l’immunoglobuline de l’espèce hôte de l’anticorps primaire. La coloration par anticorps est une approche robuste pour détecter la localisation des protéines, qui offre une résolution optique élevée au niveau intracellulaire. Cependant, le nombre d’anticorps disponibles est très limité chez le poisson zèbre. Une étude récente montre qu’environ 112 000 anticorps sont disponibles dans le commerce pour les souris; Cependant, très peu d’anticorps se sont révélés fiables chez le poisson zèbre5.
Au lieu de cela, chez le poisson zèbre, l’hybridation in situ a été largement appliquée pour l’analyse du modèle d’expression génique. Cette méthode a été utilisée pour la première fois pour évaluer l’expression génique dans les embryons de drosophile dans les années 19806,7, et depuis lors, cette technologie a été continuellement développée et améliorée. Initialement, des sondes ADN radiomarquées ont été utilisées pour détecter les transcrits d’ARNm; Cependant, la résolution spatiale était relativement faible et la radioactivité présentait des risques potentiels pour la santé. Par la suite, l’hybridation in situ repose sur les sondes à ARN marquées à la digoxigénine (DIG) ou à la fluorescéine (Fluo), qui sont conjuguées à la phosphatase alcaline (AP) ou détectées par amplification fluorescente du signal tyramide (TSA)8,9. Bien que la TSA ait été utilisée pour détecter deux ou trois gènes, le marquage DIG des sondes à ARN et des anticorps conjugués antiDIG AP sont encore des approches très sensibles, stables et largement utilisées pour l’hybridation in situ. Par conséquent, les anticorps commercialisés combinés à des sondes in situ marquées par DIG sont utiles pour fournir un aperçu de la localisation et de l’expression des protéines d’un gène.
Les embryons entiers ne peuvent pas révéler la relation spatiale entre les gènes en raison de la faible résolution optique, même si les embryons de poisson zèbre sont petits et transparents10. Par conséquent, la sectionnement est nécessaire pour analyser les modèles d’expression des gènes au niveau intracellulaire. La cryosectionnement a été largement utilisée chez le poisson zèbre car elle est facile à réaliser et peut préserver efficacement l’antigène. Par conséquent, l’hybridation in situ combinée à l’immunohistochimie dans les cryosections de poisson zèbre offre un moyen puissant d’analyser les schémas d’expression de deux gènes. Une combinaison d’hybridation in situ et d’immunohistochimie a été appliquée au poisson zèbre11. Cependant, le traitement par protéinase K a été utilisé pour améliorer la pénétration de la sonde au détriment de l’intégrité de l’antigène. Pour surmonter cette limitation, ce protocole utilise le chauffage pour induire la récupération de l’antigène. Ce protocole est non seulement applicable aux embryons de différents stades et aux coupes tissulaires de différentes épaisseurs (coupes de tête de 14 μm et coupes de moelle épinière de 20 μm), mais il a également été vérifié en utilisant des gènes exprimés dans deux organes, dont la tête et la moelle épinière.
Cet article décrira comment combiner l’hybridation in situ et la coloration d’anticorps dans des embryons de poisson zèbre dans des cryosections. La polyvalence de ce protocole est démontrée par l’utilisation d’un certain nombre de combinaisons hybridation-immunohistochimie in situ, y compris des sondes d’hybridation in situ pour deux neurones différents. Ce protocole utilise une combinaison d’hybridation in situ et d’immunohistochimie, et il constitue un moyen simple et efficace d’analyser simultanément un ARNm et une protéine. Ce protocole est applicable aux embryons à différents stades, aux sections de tissus de différentes épaisseurs et aux différents organes. Pour commencer, fixez environ 10 embryons avec 0,5 à 1 millilitre de paraformaldéhyde frais à 4% dans un tube à microfuge de 1,5 millilitre à quatre degrés Celsius pendant la nuit pendant 12 à 14 heures.Déshydrater progressivement les embryons en les lavant avec du méthanol à 25, 50 et 75% dans du PBST successivement pendant cinq minutes chacun à température ambiante. Ensuite, lavez les embryons dans du méthanol à 100% pendant cinq minutes à température ambiante. Incuber les embryons dans du méthanol à 100% à moins 20 degrés Celsius pendant au moins une nuit, pendant 12 à 14 heures.Réhydrater progressivement les embryons en les lavant avec du méthanol à 75, 50 et 25% dans du PBST successivement pendant cinq minutes chacun à température ambiante. Encore une fois, lavez l’embryon trois fois avec du PBST pendant cinq minutes chacun à température ambiante. Digérer les embryons avec 10 microgrammes par millilitre de protéinase K dans PBST à température ambiante et laver les embryons trois fois avec PBST pendant cinq minutes.Ensuite, refixez les embryons lavés dans du paraformaldéhyde à 4% pendant 15 minutes à température ambiante. Encore une fois, lavez l’embryon trois fois avec du PBST pendant l’incubation pendant cinq minutes à chaque lavage. Pour pré-hybrider les embryons, les incuber dans une solution de pré-hybridation à 65 degrés Celsius pendant cinq minutes.Ensuite, remplacez la solution de pré-hybridation par la solution d’hybridation et pré-hybridez pendant au moins quatre heures. Chauffer la sonde dans la solution d’hybridation pendant cinq minutes à 95 degrés Celsius avant de l’ajouter aux embryons. Retirer autant de solution de pré-hybridation que possible sans laisser les embryons entrer en contact avec l’air.Au tube contenant les embryons, ajouter une sonde préchauffée dans la solution d’hybridation et permettre à la sonde de s’hybrider pendant la nuit pendant 12 à 14 heures à 50 à 70 degrés Celsius. Le lendemain, aspirez la solution de sonde avec une pipette et conservez-la dans un tube à moins 20 degrés Celsius afin qu’elle puisse être réutilisée plusieurs fois. Laver les embryons avec une solution d’hybridation à 100% suivie d’un lavage séquentiel avec une solution d’hybridation à 75, 50 et 25% à deux fois la concentration de SSCT pendant 15 minutes à 65 degrés Celsius.Ensuite, lavez les embryons deux fois et 0,2 fois la concentration de SSCT pendant 15 minutes à 65 degrés Celsius. Laver les embryons deux fois pendant 10 minutes dans MABT à température ambiante. Bloquer les embryons hybrides et lavés pendant au moins deux heures à température ambiante, avec la solution de blocage à 2%-1.Remplacer la solution bloquante-1 par de l’antidigoxigénine phosphatase alcaline dans une solution bloquante à 2% 1 fraîche et agiter pendant une nuit pendant 12 à 14 heures à quatre degrés Celsius. Ensuite, lavez les embryons quatre fois dans MABT pendant 30 minutes à température ambiante. Encore une fois, lavez les embryons deux fois dans NTMT et retirez autant de NTMT que possible des embryons à l’aide d’une pipette.Remplacer par le substrat de phosphatase alcaline violette BM et colorer les embryons à température ambiante dans l’obscurité. Une fois que la tache s’est développée, arrêtez la réaction en rinçant brièvement deux fois avec NTMT. Après hybridation in situ, rincer les embryons avec du PBST trois fois pendant 20 minutes.Immergez les embryons dans du saccharose à 5% dans du PBS pendant la nuit pendant 12 à 14 heures à quatre degrés Celsius. Ensuite, changez la solution recouvrant les embryons à 15% de saccharose dans PBS et incuber pendant une nuit pendant 12 à 16 heures à quatre degrés Celsius. Encore une fois, changez la solution recouvrant les embryons à 30% de saccharose dans PBS et incuber pendant un à deux jours à quatre degrés Celsius.Transférer les embryons dans un nouveau cryomoule pour tissu et le remplir doucement avec un milieu de température de coupe optimal tout en évitant la formation de bulles. Ensuite, coupez les échantillons en sections de 12 à 20 micromètres d’épaisseur à l’aide d’un cryostat et transférez rapidement les sections sur des lames de verre. Laisser les échantillons atteindre la température ambiante et entreposer les sections dans une boîte à lames scellée à moins 80 degrés Celsius pour une utilisation ultérieure.Lavez les lames contenant les sections avec du PBS pendant cinq minutes. Ensuite, placez les lames dans le tampon et chauffez la solution pendant environ 20 minutes pour la maintenir près de la température d’ébullition. Égoutter l’excès de solution et sécher soigneusement la zone autour de chaque section avec un morceau de tissu.Ensuite, tracez un cercle autour de la section avec un stylo hydrofuge pour former une barrière hydrophobe et bloquez la section pendant deux heures dans la solution de blocage 2 à température ambiante. Pipeter la solution d’anticorps primaires par lame et incuber les lames dans une boîte humide immunohistochimique à quatre degrés Celsius pendant la nuit. Lavez les lames trois fois avec du PBS pendant l’incubation pendant 10 minutes à chaque lavage et égouttez l’excès de PBS.Ensuite, incuber les lames avec l’anticorps secondaire approprié pendant une heure à température ambiante dans PBS. Encore une fois, lavez les lames trois fois avec du PBS pendant l’incubation pendant 10 minutes à chaque lavage, et égoutter l’excès de PBS. Ensuite, pipeter le support de montage sur la glissière et monter avec un couvercle de glissière.L’expression du récepteur 5-HT2C dans le système nerveux central a été détectée dans la lignée transgénique foxP2 egfp-caax. L’expression simultanée de 5-HT2C a été observée dans la lignée transgénique avec les neurones foxP2, qui ont été marqués par la protéine fluorescente verte fluorescéine. L’expression de insm1a, un facteur de transcription à doigts de zinc dans la moelle épinière et les motoneurones du poisson zèbre, a été détectée dans la lignée transgénique.L’expression simultanée d’insm1a a été observée dans la lignée transgénique avec les neurones hb9, qui ont été marqués par la protéine fluorescente verte fluorescéine. Le poisson zèbre doit être fixé avec du paraformaldéhyde fraîchement préparé.
