このプロトコルは、生検由来の腸オルガノイドと臓器チップ技術を使用して生理学的に関連する腸チップを生成し、薬物動態と薬物間相互作用を予測し、腸上皮バリア機能障害を研究する方法を示しています。オルガノイドを臓器チップ技術に統合することで、腸上皮の複雑さを再現することができます。さらに、機械的手がかり、流れ分布、生化学的勾配の実験的制御も可能になります。この方法は、人間の腸の複雑な生理機能をエミュレートし、正常および病理学的腸機能の細胞および分子基盤をより深く理解します。これは、薬物動態と薬力学、および炎症性損傷を防ぐための潜在的な薬剤候補をより適切に予測するのに役立ちます。シードはプロトコルの重要なステップです。腸管オルガノイド断片の過度の断片化または不正確な播種密度は、上皮バリアの発達および分化を損なう可能性がある。まず、静的オルガノイド培養から培地を吸引します。ミリリットルあたり800万細胞の最終播種密度を達成するのに十分なウェルから培地を回収します。500マイクロリットルの氷冷BMM解離溶液を各ウェルに加え、可溶化BMMをウェル表面から付着させます。懸濁液を15ミリリットルの低タンパク質結合チューブに集め、氷の上に置きます。チューブを15分ごとに1時間静かに振ってください。摂氏4度で5分間Gの300倍で遠心分離し、オルガノイドペレットを得た。ペレットの上に透明なゲル層が観察される場合は、チューブから溶液を吸引する。ペレットを再懸濁し、等量のBMM解離溶液を加える。氷上で10分間インキュベートし、遠心分離します。可溶化されたBMM残基が存在しなくなるまで繰り返します。.その後、上清を捨て、ペレットを2ミリリットルのオルガノイド消化溶液に再懸濁します。チューブを摂氏37度で1〜3分間インキュベートし、8ミリリットルの高度なDMEM F12を加えて酵素反応を停止します。ペレットを遠心分離し、完全なオルガノイド増殖培地に再懸濁して、ミリリットルあたり800万細胞を達成します。滅菌1.5ミリリットルの低タンパク質結合チューブに360マイクロリットルを分注します。次に、コーティングされたチップの上部チャネルから培地を取り除き、30マイクロリットルの細胞懸濁液を追加します。チップを摂氏37度で一晩インキュベートし、オルガノイドフラグメントをメンブレンに接着させます。3ミリリットルの事前平衡化完全オルガノイド増殖培地を上部の入口リザーバーに追加し、3ミリリットルの事前平衡化された内皮細胞増殖培地を下部の入口リザーバーに追加します。同じ培地をそれぞれ上部と下部の出口リザーバーに300マイクロリットル追加します。ポータブルモジュールを運ぶトレイを培養モジュールに移します。培養モジュールで、チップを正常に接続するために十分な液滴が形成されるまで、プライムサイクルを繰り返し実行します。次に、チップキャリアをポータブルモジュールにスライドさせます。次に、2時間のレギュレーションサイクルを開始して、ポータブルモジュールとチップ内の培地を加圧した後、プログラムされた状態を再開します。次に、ストレッチ設定を変更し、培養モジュールを起動します。24時間後、10%ストレッチと同じ頻度でサイクルを繰り返します。投与培地またはビヒクルコントロールを調製するには、CYPインデューサーストック溶液またはDMSOを完全オルガノイド増殖培地および内皮細胞増殖培地で希釈します。培養モジュールを一時停止し、トレイをバイオセーフティキャビネットに持ち込みます。すべての入口および出口リザーバーの培地を、インデューサーまたは車両制御を備えた2ミリリットルの投与培地と交換します。ポータブルモジュールを培養モジュールに戻し、毎時30マイクロリットルでフローを再開します。24時間ごとに培地を新たに調製した誘導溶液と交換し、48〜72時間培養を継続します。次に、トレイをバイオセーフティキャビネットに持ち込み、すべてのリザーバーから投与培地を吸引します。上部のインレットリザーバーを洗浄し、温かい高度なDMEM F12培地で交換し、下部のリザーバーを内皮細胞増殖培地と交換します。洗浄媒体を1ミリリットルのプローブ基板溶液と交換します。チップを毎時1, 000マイクロリットルの高流量で5分間灌流し、上部と下部の両方の出口リザーバーを吸引します。チップを培養モジュールに戻し、毎時300マイクロリットルの一定流量下で1時間インキュベートします。1時間の治療後、流れを止めてトレイをバイオセーフティキャビネットに持ってきます。上部の出口リザーバーから100マイクロリットルの排水を収集し、200マイクロリットルの停止液を含むチューブに追加します。チューブをすぐにドライアイスの上に置き、サンプルを摂氏マイナス80度で保管します。両方のチャネルを200マイクロリットルの滅菌DPBSで洗浄します。次に、200マイクロリットルのフィルターピペットチップで下部チャネル出口をブロックします。50マイクロリットルの解離溶液をボトムチャネルに通し、室温で2分間インキュベートします。内皮細胞の完全な剥離を確実にし、ピペッティングによってチャネルから解離溶液を除去する。洗浄を繰り返します。次に、トップチャンネル出口をブロックし、75マイクロリットルのタンパク質溶解バッファーを灌流します。ピペットチップを挿入したまま、室温で5分間インキュベートします。細胞ライセートを5〜10回ピペッティングして1.5ミリリットルのチューブに集めます。灌流と採取を繰り返して細胞を完全に剥離させ、分析するまでライセートを摂氏マイナス80度で保存します。RNA溶解の場合は、150マイクロリットルのRNA溶解バッファーを使用しながら同じ手順に従います。まず、ストック溶液を脱気した内皮細胞増殖培地で希釈してインターフェロンガンマ投与溶液を調製する。バイオセーフティキャビネットで、下部チャネル入口リザーバーから培地を取り出し、毎日3ミリリットルの投与液と交換します。次にトレイを培養モジュールに入れ、チップを毎時1, 000マイクロリットルの高流量で5分間灌流します。流量を毎時60マイクロリットルに切り替えて、流体培養を続けます。ウェルあたり100マイクロリットルのDPBSを96ウェルの黒壁プレートに追加します。マルチチャンネルピペットを使用して、すべての貯水池からそれぞれの井戸に50マイクロリットルの排水を追加します。検量線を作製するには、DPBSで3キロダルトンデキストランカスケードブルーを1ミリリットルあたり100マイクログラム含む培地の3倍希釈を行う。次に、DPBSで内皮細胞増殖培地の3倍希釈を用いて連続妄想を行う。十二指腸チップと結腸チップの明確な形態学的特徴は、小腸の構造を表す十二指腸チップの絨毛様の形成の存在によって強調されました。シトクロムP450誘導物質リファンピシンとビタミンD3に曝露された十二指腸チップでは、薬物代謝酵素シトクロムP450 3A4のmRNA遺伝子発現の上昇とシトクロムP450酵素の触媒活性の増強があり、3つのオルガノイドドナーすべてで適切な誘導応答を示しました。結腸チップの上皮単層をインターフェロンガンマで処理すると、細胞形態の損なわれおよび柱状上皮の喪失が観察された。また、インターフェロンガンマによる処理は、ビヒクル対照と比較してタイトジャンクションタンパク質と接着ジャンクションタンパク質の細胞質シグナルの増加をもたらし、タイトジャンクションマーカーZO-1とクローディン4の変位、およびオクルジンとE-カドヘリンのインターナリゼーションを示しました。上皮傍細胞透過性の有意な増加は、結腸チップ上でのインターフェロンガンマ刺激の48時間後に観察された。同様に、インターフェロンガンマは、切断されたカスパーゼ3の細胞内含量の増加によって示されるアポトーシスの活性化を誘導する。インターフェロンガンマは、VCAM-1およびインターロイキン-6の基底外側分泌ならびにICAM-1および血清アミロイドタンパク質Aの頂端分泌によって示されるように、結腸チップにおける炎症誘発性分子の分極分泌を誘導した。腸管チップの確立に成功すると、腸管吸収、輸送、代謝、栄養素レベル、腸内ホルモン分泌、宿主微生物病原体相互作用、薬物の安全性と有効性、患者固有の疾患メカニズム、および治療への応答を研究することができます。
