皮膚には、ケラチノサイト、線維芽細胞、および神経シュワン細胞を含む複数の細胞型が含まれる。このプロトコールは、インビトロ実験のためのこれらの細胞型の各々の単離を可能にする。単一のドナーからの個々の初代細胞培養物の単離および確立は、遺伝的変異の影響を軽減しながら、堅牢な比較および共培養実験を可能にする。このプロトコルでは、個々の細胞とその相互作用の分析により、皮膚の恒常性および病態生理学におけるシュワン細胞の役割の分析が可能になります。これらの別個の細胞集団は、シュワン細胞、線維芽細胞、またはケラチノサイトを、正常な皮膚生理学、創傷治癒または皮膚疾患状態において、個々にまたは組み合わせて研究するために使用することができる。私の研究室のポスドク研究員であるJagadeeshaprasad M Gは、手順の実演を手伝ってくれます。標準的なケア手順に従って新生児包皮を取得した後、切除した包皮を8〜10ミリリットルの氷冷DMEM基礎培地を含むマイクロ遠心チューブに入れ、包皮を細胞培養実験室に輸送して細胞単離する。抗生物質と抗真菌薬を含む20〜25ミリリットルのダルベッコリン酸緩衝生理食塩水またはDPBSで包皮を2回すすいでください。次いで、洗浄した皮膚を25〜30ミリリットルの氷冷DMEM基礎培地に10センチメートルの組織培養皿に浸す。15分後、包皮をメスを用いてスライスし開き、真皮及び皮下脂肪組織を露出させた。鉗子、メス刃、はさみを使用して皮下脂肪組織を除去し、廃棄する。次いで、きれいな包皮を3〜5個の小片に切断し、滅菌円錐管内の16〜20ミリリットルのDispace1DMEM培地中で皮膚片をインキュベートする。Dispaceとのインキュベーションの最初の30分間の間に、1つは断続的に反転によって円錐形チューブ内の皮膚片を混合する。その後、円錐形のチューブを摂氏4度に16〜18時間置きます。翌日、皮膚片を取り出し、乾燥した滅菌培養皿の上に置き、外表皮層が培養皿に触れるように各皮膚片を展開させる。組織の縁から始めて、2組の鉗子を使用して表皮を真皮から離す。分離した表皮断片を10センチメートルの培養皿中のHBSの5ミリリットルに加え、室温で10分間インキュベートする。次いで、HBS溶液を50ミリリットルの円錐管に集め、5ミリリットルのトリプシン中和緩衝液またはTおよびBを円錐管に加える。チューブを脇に置きます。培養皿中の表皮断片に3ミリリットルのトリプシン溶液を加え、摂氏37度で10分間インキュベートする。インキュベーションに続いて、トリプシンケラチノサイト溶液が濁るまで表皮断片を鉗子で穏やかに攪拌する。次にトリプシンケラチノサイト溶液をHBS TおよびBチューブに加える。未消化表皮断片をトリプシンEDTA溶液で溶解した後、トリプシンEDTA溶液を入れたHBS T及びBチューブに2ミリリットルのFBSを加え、2本を約870倍Gで摂氏4度で5分間遠心分離した。次いで、上清を吸引し、細胞ペレットを3ミリリットルのケラチノサイト完全増殖培地に再懸濁し、続いて滅菌100マイクロメートルフィルターで細胞懸濁液を滅菌50ミリリットルの円錐管に濾過した。細胞数と細胞生存率が定量化されたら、培養プレートに細胞を播種し、5%の二酸化炭素中摂氏37度のインキュベーターに入れます。2日後、非接着細胞を吸引し、継代または下流実験のためにケラチノサイトが50〜80%コンフルエントに達するまで1日おきに細胞培養培地をリフレッシュする。5ミリリットルのDPBSを用いて、T25フラスコをポリ-l-リジンまたはPLLでプレコートし、プレコートフラスコを摂氏37度で3時間置く。その後、PLLコーティングマトリックスをDPBSで2回洗浄します。真皮の単離片を5ミリリットルのDMEM基礎培地で洗浄した後、真皮をはさみで小片に刻む。ミンチした真皮を摂氏37度で5ミリリットルのコラゲナーゼで2時間半消化し、真皮が完全に解離するまで30分ごとにピペットチップを使用して穏やかな滴定を行います。消化後、細胞懸濁液を70マイクロメートルのセルストレーナーで濾過し、続いて濾過した細胞懸濁液を5ミリリットルの完全DMEM培地で希釈して、コラゲナーゼの酵素活性を停止した。次に細胞懸濁液を室温で5分間870倍Gで遠心分離し、細胞投票用紙を2ミリリットルのDMEM完全培地に再懸濁し、細胞を分裂させた。記載したように遠心分離プロセスを繰り返した後、上清を吸引し、細胞ペレットを使用してシュワン細胞または線維芽細胞を単離する。シュワン細胞を単離するには、細胞ペレットを5ミリリットルの新鮮なDMEM完全培地に再懸濁し、細胞数および生存率を定量してから、5ミリリットルの再懸濁細胞を予めコード化されたT25フラスコに4.0倍の密度で10ミリメートル当たり3つの細胞に播種する。フラスコを摂氏37度および二酸化炭素5%でインキュベートする。16時間後、倍率10倍の顕微鏡観察により細胞接着を確認した。その後、非接着細胞を除去し、フラスコを5ミリリットルのDPBSで3回洗浄する。次に、DMEM完全培地中で5ミリリットルの10マイクロモルシトシンアラビノシドと共に細胞をインキュベートする。24時間後、前に実証したようにフラスコをすすぐ前にシトシンアラビノシド含有培地を吸引する。培養フラスコに5ミリリットルのシュワン細胞を補充し、培養培地を完成させ、シュワン細胞が80%のコンフルエントに達するまで1日おきに培地をリフレッシュしながら48時間培養する。線維芽細胞を単離するために、収集したペレットを5ミリリットルの線維芽細胞完全培地に再懸濁する。細胞を非コード化T25培養フラスコ中で、シュワン細胞について実証したように24時間培養した。非接着細胞を吸引し、フラスコを洗浄した後、細胞を5ミリリットルの新鮮な線維芽細胞完全培地と共に摂氏37度および5%二酸化炭素で48時間インキュベートする。単離された初代細胞をそれぞれの細胞培養培地で培養したところ、初代ケラチノサイトは7日目までに85%コンフルエントに達し、石畳形状で特徴的な細胞形態を示した。シュワン細胞は5日目までに95%の合流点に達し、双極性または三極の形をしたように見えた。線維芽細胞培養物は4日目までに95%コンフルエントに達し、ほとんどの細胞は紡錘体形状形態を示した。免疫蛍光染色により、細胞培養物中の細胞型特異的タンパク質発現を確認した。ケラチノサイトはK10およびK14タンパク質に対して陽性であった。ケラチン免疫蛍光とDAPI核染色のマージ画像は、細胞培養物の純度97.8%を示した。同様に、Schwann細胞はS100およびP75 NTRに対して陽性であり、培養において95.2%の純度を有していた。この線維芽細胞はビメンチンを陽性に発現したが、筋線維芽細胞マーカーであるα平滑筋アクチンを発現しなかった。培養物の純度は約97.2%であった Shanseの証拠を高めるために、プレコートされたポリ-l-リジン皿を使用することができる。そしてさらに繊維汚染を緩和するために、シトシンアラビノシドおよび抗真菌剤は、細胞が短時間添加した後に添加した。これは、単一のヒトの包皮から、シュワン細胞、ケラチノサイト、線維芽細胞などの3つの初代細胞を確立するための簡単な安価な実験手順である。このプロトコルは、細胞細胞通信または細胞型間のクロストークを含むインビトロ実験に最適です。
