神経血管疾患は、死亡率と罹患率の主な原因である虚血性脳卒中、および視力喪失につながる網膜血管疾患の原因となる主要な医療問題です1,2。神経血管疾患をモデル化するために、網膜静脈閉塞(RVO)のマウスモデルを採用しています。このモデルは非侵襲的であり、臨床現場で網膜血管疾患のある人を検査するために使用されるものと同様のin vivoイメージング技術を利用しています。したがって、このモデルを使用すると、このモデルを利用した研究の翻訳の可能性が高まります。すべてのマウスモデルと同様に、モデルの再現性を最大化することが重要です。
網膜血管疾患は、70歳未満の人々の視力喪失の主な原因です。RVOは、糖尿病性網膜症に次いで2番目に多い網膜血管疾患です3。RVOに特徴的な臨床的特徴には、虚血性損傷、網膜浮腫、およびニューロン喪失の結果としての視力喪失が含まれる3,4。主要血管のレーザー光凝固を用いたRVOのマウスモデルが開発され、ヒトRVO 5,6,7で観察される主要な臨床病理を再現するために改良されています。眼科イメージングの進歩により、ヒトで使用される非侵襲的診断ツール、すなわちフルオレセイン血管造影(FA)および光干渉断層撮影(OCT)の複製も可能になりました6。フルオレセイン血管造影は、小さな蛍光色素であるフルオレセインの注射を使用して、血液網膜関門(BRB)の破壊による漏出、および閉塞部位を含む網膜の血流動態の観察を可能にします8,9。OCTイメージングは、網膜の高解像度断面画像の取得および網膜層の厚さおよび組織の研究を可能にする10。FA画像の分析は歴史的に主に定性的であり、研究間の直接的で再現性のある比較の可能性を制限しています。最近、OCTイメージングにおける層の厚さを定量化するための多くの方法が開発されましたが、現在標準化された分析プロトコルはなく、OCT画像取得のサイトはさまざまです11。これらのツールを適切に活用するためには、標準化された定量的で複製可能なデータ分析方法論が必要です。本論文では、RVO-フルオレセイン漏出、OCT層の厚さ、および網膜層の混乱のマウスモデルにおける病理学的損傷を評価するために使用される3つの血管読み出しを紹介します。
