皮膚は、環境への重要なインターフェースとして機能する体内最大の器官であり、多くの神経線維1,2,3によって密に神経支配されている。皮膚神経支配は、全マウント皮膚および組織切片4,5,6に対する染色など、様々な組織学的方法を用いて以前に広く研究されてきたが、皮膚神経線維の詳細な効果的な実証は依然として課題である7,8。これを踏まえ、本プロトコールは、厚い組織切片において皮膚神経線維をより明確に示すための独自の技術を開発した。
切片の太さによる限界のため、神経支配皮膚神経線維の観察は、取得した画像情報からカルシトニン遺伝子関連ペプチド(CGRP)神経線維と局所組織・臓器との関係を正確に描くほど正確ではない。3D組織クリアリング技術の出現は、この問題を解決するための実行可能な方法を提供する9,10。組織クリアリングアプローチの急速な発展は、近年、組織構造、器官全体、ニューロン突起、および動物全体を研究するための多くのツールを提供してきました11。透明な皮膚組織は、共焦点顕微鏡によってはるかに厚い部分で画像化され、皮膚神経線維を視覚化するためのデータを得ることができる。
現在の研究では、ラット後足の足底および背部皮膚を、毛状およびグラブラス性皮膚の2つの標的部位として選択した3、4、7。より長い距離で皮膚神経線維をトレースするために、皮膚組織を免疫組織化学的および組織化学的染色のために300μmの厚さでスライスし、続いて組織透明化処置を行った。CGRPは、感覚神経線維12、13を標識するために使用した。さらに、組織背景上の皮膚神経線維を強調するために、ファロイジンおよびリンパ管内皮ヒアルロン酸受容体1(LYVE1)をさらに用いて、それぞれ血管およびリンパ管を標識した14、15。

我々のプロトコルは、共焦点顕微鏡を用いて同期部を観察する実行可能な方法を提供し、神経線維の観察における精度を保証することができた。シンプルで簡単な組織洗浄プロトコルは、この技術の主な利点です。組織洗浄プロセスは効率的であり、約30分〜1時間かかる。テキスト原稿に記載されているように、安楽死したラットに灌流を行う。メスを使用して、背骨とソールの皮膚を後足から取り除きます。凍結切片を必要とするサンプルの場合、組織を0.1モルPB中のアルデヒドから4%で2時間固定する。洗浄後、皮膚組織を摂氏37度で2%のアガロースに埋め込み、冷却のためにアイスボックスの中に置きます。取り付けた組織を氷水で振動マイクロトーンに固定し、横方向に500マイクロメートルの厚さでスライスします。スライス後、切片からアガロースを取り出し、1x PBSを含む6ウェルプレートに保存します。組織切片を1x PBS中の2%トリトンX-100の溶液中で摂氏4度で一晩インキュベートする。組織切片をブロッキングバッファーに入れ、シェーカー上で毎分72回転で摂氏4度で一晩回転させます。マイクロ遠沈管内の希釈緩衝液中のマウスモノクローナル抗CGRPおよびヒツジポリクローナル抗LYVE-1抗体の一次抗体を含む溶液に組織切片を移す。その後、シェーカーを摂氏4度で2日間回転させます。組織切片を室温の洗浄バッファーで2回洗浄し、次いで洗浄バッファー中の摂氏4度で一晩シェーカーに保管する。翌日、マイクロ遠沈管内の二次抗体を含む混合溶液に組織切片を移し、シェーカー上で毎分72回転で摂氏4度で5時間回転させる。6ウェルプレート中の組織切片を洗浄バッファーで室温で1時間2回洗浄する。組織切片をシェーカー上の洗浄バッファーに4°Cで一晩保管してください。組織切片を組織透明化試薬に移し、シェーカー上で毎分60回転で室温で1時間穏やかに回転させる。透明な組織をスライドに取り付け、スペーサーで組織を囲み、新鮮な組織透明化試薬とカバースリップで隙間を貼り付けます。3Dパターンは、CGRP陽性神経線維が皮下組織および真皮を通って表皮に通過することを実証した。これらの神経線維は、真皮内で分岐し、表皮で終端した皮下組織において束状に走った。これに対し、陽性血管およびLYVE-1陽性リンパ管におけるファロイジンは、皮下組織および真皮に分布している。一般に、CGRP陽性神経線維は、毛むくじゃらでグラブラスな皮膚において3Dネットワークを形成する血管およびリンパ管に平行に、または周囲を走っていた。従来の手法では、神経線維血管やリンパ管は、薄切片にCGRPファロイジンやLYVE-1で明確に標識されていましたが、これらの構造の観察はスライスの厚さによって制限され、完全に観察することはできませんでした。厚さ30マイクロメートルの区間では、CGRP陽性神経線維の表面積において、毛むくじゃらの皮膚とグラブラスな皮膚との間に差はなかった。厚さ300マイクロメートルの組織切片では、毛むくじゃらの皮膚と比較して、グラブラス皮膚のCGRP陽性神経線維の表面積が有意に増加した。抗体インキュベーション後には、浸透圧の高い洗浄バッファーを使用することが重要です。数回、免疫蛍光染色に要する洗浄時間が必要である。この手順に従えば、6つの切片に他の組織や臓器の染色や観察も存在する可能性があると考えています。この技術は、神経繊維の表面積と状態に遭遇する神経科学研究者を助けることができる。また、組織や臓器の革新を観察するのにも役立ちます。
