このプロトコルは、腸と甲状腺がウイルスの拡散を妨げるかどうかを判断し、ネッタイシマカによって伝染する蚊媒介ウイルスのリスク評価に役立ちます。私たちのプロトコルには、経口栄養と胸腔内注射の2つの感染方法が含まれており、アルボウイルスのベクター能力を効果的に評価することができました。このプロトコルは、ネッタイシマカのデング熱ウイルスやジカウイルス感染など、さまざまな蚊媒介動物における追加のアルボウイルス感染に適用される可能性があり、実行可能な手順であることが判明する可能性があります。手順を実演するのは、私の研究室の研究助手であるFei Wangです。人工蚊の餌付けシステムを使用して人工給餌を行うには、コラーゲン膜を適切なサイズに切断し、Oリングでリザーバーに固定します。3ミリリットルのウイルスと血液の混合物をリザーバーに加え、漏れを防ぐためにプラスチックプラグを使用してリザーバーを密閉します。蚊が入ったプラスチック製のコップをグローブボックスに入れます。密閉されたリザーバーをFU1フィーダーにねじ込み、1つのリザーバーを1つのカップに置きます。電源ユニットの電源を入れ、蚊に1時間餌をやる。給餌後、蚊が気を失うまで数秒間氷で麻酔をかけます。麻酔をかけた蚊を氷の上に置いたペトリ皿に注ぎ、蓋をすばやく覆います。鉗子で充血した雌の蚊を選び、カップあたり50匹の雌の蚊で新しいプラスチックカップに移します。カットした蚊帳メッシュでカップを包み、中央に穴の開いた蓋で覆います。スポンジを細かく切り、カップの上に置きます。プラスチック製の使い捨てスポイトを使用して、スポンジに8%グルコース溶液を追加します。雌の蚊が入っているカップを摂氏27度、湿度80%のインキュベーターに10日間入れます。72時間ごとに新しいグルコース飽和スポンジを交換してください。胸腔内接種のために、テキスト原稿に記載されているように雌の蚊を準備します。感染性ウイルス希釈液を準備するには、摂氏80度の冷凍庫からウイルスストックを取り出し、氷上で解凍します。10%FBSおよび1%ペニシリン-ストレプトマイシンを含むRPMI 1640培地で、100〜500プラーク形成単位ウイルスを含む100ナノリットルのウイルス希釈液でウイルスストックを希釈します。ウイルス希釈液を氷の上に置きます。次に、ニードルプーラープログラムのパラメータを熱指数450、力(g)を110、距離(ミリメートル)を1、秒単位の遅延をゼロに設定して、プーラー1000を使用してマイクロ注入針を準備します。引っ張った針の先端をピンセットで16倍の倍率で解剖顕微鏡で切ります。使い捨ての滅菌注射器を通して鉱油で針を埋め戻します。機械の指示に従って針をインジェクターに取り付けます。感染性ウイルス希釈液の入ったチューブに針を注意深く挿入します。次に、塗りつぶしボタンを押して、約4マイクロリットルのウイルス希釈液を針に充填します。針がいっぱいになったら、針に触れたり損傷したりしないでください。ピンセットで約50匹のメスの麻酔をかけた蚊を選び、氷板の上に置きます。プレートをインジェクターの下に置き、解剖顕微鏡下で蚊の胸腔に針を挿入します。注入量を100ナノリットルに設定し、速度を毎秒50ナノリットルに設定します。注入ボタンを押して、ウイルス溶液を蚊に流します。注射が成功すると、腹部はわずかに膨らみます。感染した蚊を氷の上に置いた新しいプラスチックカップに入れます。感染した蚊の数が十分になったら、カットした蚊帳メッシュでカップを包み、蓋で覆います。感染した雌の蚊から唾液を集めるには、麻酔をかけた蚊を氷の上に置いたペトリ皿に注ぎ、蓋をすばやく閉めます。液浸油で満たされた10マイクロリットルのピペットチップをゴム泥の上に並べて置きます。ピンセットで雌の蚊を選び、氷板の上に置きます。ピンセットで足と翼を取り外します。各ピペットチップに1匹の蚊の口の部分を置き、室温で蚊によって油に分泌される唾液を収集します。45〜60分後、200マイクロリットルのウイルス希釈培地を含む1.5ミリリットルのチューブにピペットチップを入れます。チューブを5, 000 gで摂氏4度で5分間遠心分離し、唾液をチューブに排出します。唾液含有チューブを摂氏80度で保管し、その後の透過率の決定を行います。唾液採取後、感染した雌の蚊を解剖します。ピンセットを使用して、蚊の頭を切り取り、各頭を200マイクロリットルのRPMI 1640培地を含む個々のチューブに入れます。ピンセットで胸郭をつかみます。次に、腹部の最後から2番目のセグメントを別のピンセットでつかみ、腸を引き抜きます。浮遊組織や臓器がないか腸をきれいにします。腸をPBSで洗浄し、各腸を200マイクロリットルのRPMI 1640培地を含む個々のチューブに入れます。これらのチューブを摂氏80度で保管して、ウイルスの拡散と感染率を後で決定します。この代表的な分析では、感染後10日目の雌蚊の腸、頭部、唾液中のウイルスRNAが決定されました。胸腔内接種蚊の腸内、頭部、唾液中のエビヌール湖ウイルスのウイルス力価は、口腔感染蚊のそれよりも高かった。胸腔内接種蚊の感染率と播種率は100%、口腔内感染蚊の感染率は70%と38.1%でした。胸腔内接種された蚊の感染率は最大90%に達したが、口腔に感染した蚊の感染率はわずか4.8%であり、摂食量がウイルス含有量に影響を与えるため、充血した雌蚊は一貫性を維持するために摘み取られた。
